The molecular level details of the displacement of surface adsorbed fibrinogen from silica substrates were studied by atomic force microscopy, immunochemical assays, fluorescence microscopy, and vibrational sum frequency spectroscopy. The results showed that human plasma fibrinogen (HPF) can be readily displaced from the interface by other plasma proteins near neutral pH because the positively charged alpha C domains on HPF sit between the rest of the macromolecule and the underlying surface. The alpha C domains make weak electrostatic contact with the substrate, which is manifest by a high degree of alignment of Lys and Arg residues. Upon cycling through acidic pH, however, the alpha C domains are irreversibly removed from this position and the rest of the macromolecule is free to engage in stronger hydrogen bonding, van der Waals, and hydrophobic interactions with the surface. This results in a 170-fold decrease in the rate at which HPF can be displaced from the interface by other proteins in human plasma.

The spectra revealed ion specific changes in both monolayer ordering and water structure with the former following the Hofmeister series. Separate measurements of the surface potential as a function of ion tracked closely to changes in alkyl chain structure, but less closely to changes in water structure. The disruption of the monolayer ordering could be ascribed to the relative ability of the ions to penetrate past the hydrophilic surface of the monolayer's headgroups and into the more hydrophobic portion of the thin film. The corresponding trends observed in the surface water structure showed significant deviations from the Hofmeister series, leading to the conclusion that the changes in surface water structure, often credited with being the origin of Hofmeister effects, are probably not of primary importance. On the other hand, dispersion forces almost certainly play a large role in the order of the Hofmeister series.

Vibrational sum-frequency spectroscopy (VSFS) was used to study gauche defects in octadecylamine (ODA) monolayers at the air/water interface. The VSFS spectra provide unique insights into phase transitions that occur as a result of changes in the structure of the monolayer's hydrophobic region. These changes can be attributed to the increased presence of gauche conformers in the ODA alkyl chains during the monolayer's transition from the solid to liquid phase. Temperature-dependent spectra from monolayers at several different pressures were used to assign the phase transition temperature based on the observed changes in microscopic structure. Through application of a two-dimensional form of the Clapeyron equation, the first in situ measurements of the entropy and enthalpy changes associated with gauche conformers in a monolayer were made.

Transmission of force from outside the cell through focal adhesions controls the dynamics of these adhesion sites and initiates intracellular signaling cascades that alter cellular behavior. To understand the mechanism by which living cells sense mechanical forces, and how they respond and adapt to their environment, a critical first step is to develop a new technology to investigate cellular behavior at subcellular level that integrates an atomic force microscope (AFM) with total internal reflection fluorescence (TIRF) and fast-spinning disk (FSD) confocal microscopy, providing high spatial and temporal resolution. AFM uses a nanosensor to measure the cell surface topography and can apply and measure mechanical force with high precision. TIRF microscopy is an optical imaging technique that provides high-contrast images with high z-resolution of fluorescently labeled molecules in the immediate vicinity of the cell-coverslip interface. FSD confocal microscopy allows rapid 3-D imaging throughout the cell in real time. The integrated system is broadly applicable across a wide range of molecular dynamic studies in any adherent live cells, allowing direct optical imaging of cell responses to mechanical stimulation in real time.

Vibrational sum frequency spectroscopy (VSFS) was used to explore specific ion effects on interfacial water structure adjacent to a bovine serum albumin (BSA) monolayer adsorbed at the air/water interface. The subphase conditions were varied by the use of six different sodium salts and four different pH values. At pH 2 and 3, the protein layer was positively charged and it was found that the most chaotropic anions caused the greatest attenuation of water structure. The order of the salts followed an inverse Hofmeister series. On the other hand, when the protein layer was near its isoelectric point (pH 5), the most chaotropic anions caused the greatest increase in water structure, although the effect was weak. In this case, a direct Hofmeister series was obeyed. Finally, virtually no effect was observed when the protein layer was negatively charged with a subphase pH of 9. For comparison, similar experiments were run with positively charged, negatively charged, and zwitterionic surfactant monolayers. These experiments gave rise to nearly the identical results as the protein monolayer which suggested that specific anion effects are dominated by the charge state of the interfacial layer rather than its detailed chemical structure. In a final set of experiments, salt effects were examined with a monolayer made from an elastin-like polypeptide (ELP). The peptide consisted of 120 pentameric repeats of the sequence Val-Pro-Gly-Val-Gly. Data from this net neutral biopolymer followed a very weak, but direct Hofmeister series. This suggested that direct anion binding to the amide groups in the backbone of a polypeptide is quite weak in agreement with the BSA data. The results from the variously charged protein, surfactant, and polymer monolayers were compared with a modified Gouy-Chapman-Stern model. The agreement with this simple model was quite good.

A pH 2 y 3, la capa de prote&#xED;na se carg&#xF3; positivamente y se encontr&#xF3; que los aniones m&#xE1;s caotr&#xF3;picos caus&#xF3; la mayor atenuaci&#xF3;n de la estructura del agua. El orden de las sales seguido una serie de Hofmeister inversa. Por otro lado, cuando la capa de prote&#xED;na estaba cerca de su punto isoel&#xE9;ctrico (pH 5), los aniones m&#xE1;s caotr&#xF3;picos caus&#xF3; el mayor incremento en la estructura del agua, aunque el efecto era d&#xE9;bil. En este caso, una serie de Hofmeister directa fue obedecida. Por &#xFA;ltimo, pr&#xE1;cticamente ning&#xFA;n efecto se observ&#xF3; cuando la capa de prote&#xED;na fue cargado negativamente con un pH de 9 subfase. Para la comparaci&#xF3;n, experimentos similares se llevaron a cabo con monocapas tensioactivos con carga positiva, cargado negativamente, y zwitteri&#xF3;nicos. Estos experimentos dieron lugar a casi los mismos resultados como la monocapa prote&#xED;na que sugiere que los efectos espec&#xED;ficos de aniones est&#xE1;n dominadas por el estado de carga de la capa interfacial en lugar de su estructura qu&#xED;mica detallada. En una serie final de experimentos, los efectos de la sal fueron examinados con una monocapa a partir de un polip&#xE9;ptido similar a la elastina (ELP). El p&#xE9;ptido constaba de 120 repeticiones de la secuencia pentam&#xE9;ricas Val-Pro-Gly-Val-Gly. Los datos de este biopol&#xED;mero neto neutro seguido de una serie de Hofmeister muy d&#xE9;bil, pero directo. Esto sugiri&#xF3; que el ani&#xF3;n uni&#xF3;n directa a los grupos amida en la cadena principal de un polip&#xE9;ptido es muy d&#xE9;bil, de acuerdo con los datos BSA. Los resultados de la prote&#xED;na diversamente cargada, agente tensioactivo, y las monocapas de pol&#xED;mero se compararon con una modificaci&#xF3;n de Gouy-Chapman-Stern modelo. El acuerdo con este sencillo modelo era bastante bueno.

Morphological adaptations of vascular smooth muscle cells (VSMC) to the mechanically active environment in which they reside, are mediated by direct interactions with the extracellular matrix (ECM) which induces physiological changes at the intracellular level. This study aimed to analyze the effects of the ECM on RhoA-induced mechanical signaling that controls actin organization and focal adhesion formation. VSMC were transfected with RhoA constructs (wild type, dominant negative or constitutively active) and plated on different ECM proteins used as substrate (fibronectin, collagen IV, collagen I, and laminin) or poly-l-lysine as control. Morphological changes of the VSMC were detected by fluorescence confocal microscopy and total internal reflection fluorescence (TIRF) microscopy, and were independently verified using adhesion assays and Western blot analysis. Our results showed that the ECM has an important role in cell spreading, adhesion and morphology with a direct effect on modulating RhoA signaling. RhoA activity significantly affected the stress fibers and focal adhesions reorganization, but in a context imposed by the ECM. Thus, RhoA activity modulation in VSMC induced an increased activation of stress fibers and FA formation at 5h, while a significant inhibition was recorded at 24h after plating on the different ECM. Our findings provide biophysical evidence that ECM modulates VSMC response to mechanical stimuli inducing intracellular biochemical signaling involved in cellular adaptation to the local microenvironment.

Extracellular matrix effect on RhoA signaling modulation in vascular smooth muscle cells.

Adaptations morphologiques des cellules musculaires lisses vasculaires (CMLV) &#xE0; l&#x26;#39;environnement m&#xE9;canique active dans laquelle ils r&#xE9;sident, sont m&#xE9;di&#xE9;s par des interactions directes avec la matrice extracellulaire (MEC), qui entra&#xEE;ne des changements physiologiques au niveau intracellulaire. Cette &#xE9;tude visait &#xE0; analyser les effets de l&#x26;#39;ECM RhoA induite par la signalisation m&#xE9;canique qui contr&#xF4;le l&#x26;#39;organisation d&#x26;#39;actine et la formation d&#x26;#39;adh&#xE9;sion focale. CMLV ont &#xE9;t&#xE9; transfect&#xE9;es avec des constructions de type sauvage (RhoA, dominant n&#xE9;gatif ou constitutivement active) et plaqu&#xE9; sur les prot&#xE9;ines ECM diff&#xE9;rents utilis&#xE9;s comme substrat (fibronectine, le collag&#xE8;ne IV, collag&#xE8;ne I, et de la laminine) ou poly-L-lysine comme t&#xE9;moin. Morphologische Anpassungen der vaskul&auml;ren glatten Muskelzellen (VSMC), die mechanisch aktive Umgebung, in dem sie wohnen, vermittelt durch direkte Interaktionen mit der extrazellul&auml;ren Matrix (ECM), die physiologische Ver&auml;nderungen auf der intrazellul&auml;ren Ebene hervorruft. Diese Studie soll analysieren die Auswirkungen von ECM auf RhoA-induzierten mechanischen signalisieren, die Aktin-Organisation und fokale Adh&auml;sion Bildung kontrolliert. VSMC wurden die transfected mit RhoA Konstrukte (Wildtyp, dominante negative oder konstitutiv aktiv) und plattiert auf verschiedenen ECM-Proteine, die als Substrat verwendet (Fibronektin, Kollagen IV, Kollagen I und Laminin) oder Poly-l-Lysin als Steuerelement. Adattamenti morfologici delle cellule muscolari lisce vascolari (VSMC) all&#39;ambiente meccanicamente attivo in cui risiedono, sono mediate dall&#39;interazione dirette con la matrice extracellulare (ECM) che induce cambiamenti fisiologici a livello intracellulare. Questo studio finalizzato ad analizzare gli effetti dell&#39;ECM su RhoA-indotto meccanico segnalando che controlla organizzazione actina e formazione di adesione focale. VSMC erano transfected con costrutti RhoA (wild type, dominante negativo o costitutivamente attive) e placcato su diverse proteine ECM utilizzate come substrato (fibronectina, collagene IV, collagene I e laminina) o poli-l-lisina come controllo. Cambiamenti morfologici delle VSMC sono state rilevate da microscopia confocale a fluorescenza e microscopia di fluorescenza (TIRF) riflessione interna totale e sono stati verificati in modo indipendente utilizzando dosaggi di adesione e analisi Western blot. I nostri risultati hanno mostrato che l&#39;ECM ha un ruolo importante nella diffusione delle cellule, adesione e morfologia con un effetto diretto sulla modulazione RhoA segnalazione. RhoA attivit&agrave; significativamente colpite le fibre di stress e la riorganizzazione di adesioni focali, ma in un contesto imposto dall&#39;ECM. Adaptaciones morfol&#xF3;gicas de c&#xE9;lulas musculares lisas vasculares (CMLV) en el medio ambiente mec&#xE1;nicamente activo en el que residen, son mediadas por la interacci&#xF3;n directa con la matriz extracelular (MEC), que induce cambios fisiol&#xF3;gicos a nivel intracelular. Este estudio tuvo como objetivo analizar los efectos de la ECM en RhoA inducida por la se&#xF1;alizaci&#xF3;n mec&#xE1;nica que controla la organizaci&#xF3;n de la actina y la formaci&#xF3;n de adhesi&#xF3;n focal. CMLV fueron transfectadas con construcciones RhoA (tipo salvaje, dominante negativa o activa constitutivamente) y chapada en prote&#xED;nas ECM diferentes utilizados como sustrato (fibronectina, col&#xE1;geno IV, col&#xE1;geno I, y laminina) o poli-l-lisina como control. Los cambios morfol&#xF3;gicos de la CMLV fueron detectados por microscop&#xED;a de fluorescencia confocal y microscop&#xED;a de reflexi&#xF3;n interna total de fluorescencia (TIRF), y fueron verificados de forma independiente utilizando los ensayos de adhesi&#xF3;n y el an&#xE1;lisis de Western blot. Nuestros resultados mostraron que el ECM tiene un papel importante en la propagaci&#xF3;n de c&#xE9;lulas, la adhesi&#xF3;n y la morfolog&#xED;a con un efecto directo sobre la modulaci&#xF3;n de la se&#xF1;alizaci&#xF3;n de RhoA. La actividad de RhoA afectado significativamente las fibras de estr&#xE9;s y la reorganizaci&#xF3;n adhesiones focales, pero en un contexto impuesto por el MEC. Por lo tanto, la modulaci&#xF3;n de la actividad de RhoA en las CMLV indujo un aumento de la activaci&#xF3;n de fibras de estr&#xE9;s y la formaci&#xF3;n de la FA a las 5h, mientras que una inhibici&#xF3;n significativa se registr&#xF3; a las 24 horas despu&#xE9;s de placas en el ECM diferente. Nuestros resultados proporcionan pruebas biof&#xED;sicas que ECM modula la respuesta a los est&#xED;mulos mec&#xE1;nicos CMLV induciendo intracelulares de se&#xF1;alizaci&#xF3;n bioqu&#xED;micos implicados en la adaptaci&#xF3;n celular al microambiente local.
