Newborn neurons elaborate an axon that undertakes a complicated journey to find its ultimate target in the brain or periphery. Although major progress in the study of this process has been made by analysis of dissociated neurons in vitro, one would like to observe and manipulate axonal outgrowth and pathfinding as it occurs in situ, as fasciculated nerves growing within the tissue itself. Here, we present a simple technique to do this, through cultivation of embryonic mouse slices expressing enhanced green fluorescent protein (EGFP) specifically in newborn neurons. This system allows for imaging of outgrowth of peripheral nerves into structures such as the developing limb. We demonstrate a reproduction of normal innervation patterns by spinal nerves derived from spinal cord motor neurons and sensory neurons of the dorsal root ganglia. The slices can be manipulated pharmacologically as well as genetically, by crossing the EGFP-expressing line with lines containing targeted mutations in genes of interest.

Nouveaux neurones &eacute;laborer un axone qui entreprend un voyage compliqu&eacute; pour trouver sa cible ultime dans le cerveau ou de la p&eacute;riph&eacute;rie. Bien que des progr&egrave;s importants dans l&#39;&eacute;tude de ce processus a &eacute;t&eacute; d&eacute;pos&eacute;e par l&#39;analyse des neurones dissoci&eacute;s in vitro, on voudrait d&#39;observer et de manipuler la croissance axonale et pathfinding lorsqu&#39;il se produit in situ, comme les nerfs fascicul&eacute;es de plus en plus dans le tissu lui-m&ecirc;me. Nous pr&eacute;sentons ici une technique simple pour faire cela, par le biais de culture des tranches embryonnaires de souris exprimant la prot&eacute;ine fluorescente verte am&eacute;lior&eacute;e (EGFP) plus pr&eacute;cis&eacute;ment dans les nouveaux neurones. Ce syst&egrave;me permet d&#39;imagerie de l&#39;excroissance des nerfs p&eacute;riph&eacute;riques dans des structures telles que le membre en d&eacute;veloppement. Nous d&eacute;montrons une reproduction des patrons de l&#39;innervation normale par les nerfs rachidiens provenant des neurones moteurs de la moelle &eacute;pini&egrave;re et des neurones sensoriels des ganglions de la racine dorsale. Neugeborene Neuronen erarbeiten ein Axon, die eine komplizierte Reise um das ultimative Ziel zu finden, in das Gehirn oder der Peripherie vornimmt. Obwohl gro&szlig;e Fortschritte in der Studie dieses Prozesses durch Analyse der getrennten Neuronen in-vitro gemacht hat schon, m&ouml;chte man beobachten und axonale Auswuchs und Bewegungsplanungs zu manipulieren, wie es vor Ort, als fasciculated Nerven innerhalb des Gewebes selbst wachsen auftritt. Hier pr&auml;sentieren wir eine einfache Technik dazu durch Anbau von embryonalen Maus Scheiben verbessertes gr&uuml;nes fluoreszierendes Protein (EGFP) speziell in neugeborenen Neuronen zum Ausdruck zu bringen. Dieses System erm&ouml;glicht Bildgebung der Auswuchs der peripheren Nerven in Strukturen wie den Entwicklungsl&auml;ndern Gliedma&szlig;en. Wir zeigen eine Reproduktion des normalen Innervation Muster von spinale Nerven von Motoneuronen des R&uuml;ckenmarks und sensorischen Neuronen von den Spinalganglien abgeleitet. Die Scheiben k&ouml;nnen pharmakologisch als auch als genetisch manipuliert werden, durch die Trennungslinie EGFP-Ausdr&uuml;cken mit Zeilen mit gezielte Mutationen in Genen von Interesse.

Neuroni neonati elaborano un assone che intraprende un viaggio complicato per trovare la sua destinazione finale nel cervello o la periferia. Nonostante importanti progressi nello studio di questo processo compiuti dall&#39;analisi dei neuroni dissociati in vitro, uno vorrebbe osservare e manipolare escrescenza assonale e path-finding come si verifica in situ, come i nervi fasciculated crescente all&#39;interno del tessuto stesso. Qui di seguito, presentiamo una tecnica semplice per fare questo, attraverso la coltivazione di fette embrionali del mouse che esprime maggiore proteina fluorescente verde (EGFP) specificamente in neuroni neonati. Questo sistema permette per l&#39;imaging di escrescenza dei nervi periferici in strutture come l&#39;arto in via di sviluppo. Dimostriamo una riproduzione di modelli normale innervazione dai nervi spinali derivati da neuroni motori del midollo spinale e i neuroni sensitivi dei gangli spinali. Aunque importantes avances en el estudio de este proceso se han realizado mediante el an&aacute;lisis de las neuronas disociadas in vitro, uno quisiera observar y manipular consecuencia axonal y pathfinding como ocurre in situ, como nervios fasciculados creciendo dentro del propio tejido. Aqu&iacute;, presentamos una t&eacute;cnica sencilla para hacer esto, a trav&eacute;s del cultivo de l&aacute;minas embrionarias de rat&oacute;n expresan la prote&iacute;na fluorescente verde (EGFP) mejorada, espec&iacute;ficamente en las neuronas reci&eacute;n nacidas. Este sistema permite la proyecci&oacute;n de imagen del fruto de los nervios perif&eacute;ricos en estructuras como el miembro en desarrollo. Demostramos una reproducci&oacute;n de los patrones normales de la inervaci&oacute;n por los nervios raqu&iacute;deos derivados de las neuronas motoras de la m&eacute;dula espinal y las neuronas sensoriales de los ganglios de la ra&iacute;z dorsal.
