In Drosophila, like in humans, Dystrophin Glycoprotein Complex (DGC) deficiencies cause a life span shortening disease, associated with muscle dysfunction. We performed the first in vivo genetic interaction screen in ageing dystrophic muscles and identified genes that have not been shown before to have a role in the development of muscular dystrophy and interact with dystrophin and/or dystroglycan. Mutations in many of the found interacting genes cause age-dependent morphological and heat-induced physiological defects in muscles, suggesting their importance in the tissue. Majority of them is phylogenetically conserved and implicated in human disorders, mainly tumors and myopathies. Functionally they can be divided into three main categories: proteins involved in communication between muscle and neuron, and interestingly, in mechanical and cellular stress response pathways. Our data show that stress induces muscle degeneration and accelerates age-dependent muscular dystrophy. Dystrophic muscles are already compromised; and as a consequence they are less adaptive and more sensitive to energetic stress and to changes in the ambient temperature. However, only dystroglycan, but not dystrophin deficiency causes extreme myodegeneration induced by energetic stress suggesting that dystroglycan might be a component of the low-energy pathway and act as a transducer of energetic stress in normal and dystrophic muscles.

Stress and muscular dystrophy: a genetic screen for dystroglycan and dystrophin interactors in Drosophila identifies cellular stress response components.

Chez la drosophile, comme chez les humains, la dystrophine glycoprot&#xE9;ine complexes (DGC) carences causer une maladie raccourcissement la dur&#xE9;e de vie, associ&#xE9;e &#xE0; une dysfonction musculaire. Nous avons effectu&#xE9; le premier dans l&#x26;#39;&#xE9;cran vivo interaction g&#xE9;n&#xE9;tique dans le vieillissement des muscles dystrophiques et des g&#xE8;nes identifi&#xE9;s qui n&#x26;#39;ont pas &#xE9;t&#xE9; indiqu&#xE9;es avant d&#x26;#39;avoir un r&#xF4;le dans le d&#xE9;veloppement de la dystrophie musculaire et d&#x26;#39;interagir avec la dystrophine et / ou dystroglycane. Des mutations dans plusieurs g&#xE8;nes qui interagissent trouv&#xE9;s provoquer fonction de l&#x26;#39;&#xE2;ge morphologiques et de la chaleur induite par les d&#xE9;fauts physiologiques dans les muscles, ce qui sugg&#xE8;re leur importance dans le tissu. La majorit&#xE9; d&#x26;#39;entre eux est phylog&#xE9;n&#xE9;tiquement conserv&#xE9;s et mis en cause dans des maladies humaines, principalement les tumeurs et les myopathies. Fonctionnellement, ils peuvent &#xEA;tre divis&#xE9;s en trois cat&#xE9;gories principales: les prot&#xE9;ines impliqu&#xE9;es dans la communication entre le muscle et le neurone, et il est int&#xE9;ressant, en m&#xE9;canique et cellulaire des voies de r&#xE9;ponse au stress. Nos donn&#xE9;es montrent que le stress induit la d&#xE9;g&#xE9;n&#xE9;rescence musculaire et acc&#xE9;l&#xE8;re d&#xE9;pendant de l&#x26;#39;&#xE2;ge la dystrophie musculaire. Muscles dystrophiques sont d&#xE9;j&#xE0; compromise, et en cons&#xE9;quence, ils sont moins adaptative et plus sensibles au stress &#xE9;nerg&#xE9;tique et aux changements dans la temp&#xE9;rature ambiante. When the main components of the DGC, Dystrophin (Dys) and Dystroglycan (Dg) are affected cognitive impairment and mental retardation in addition to muscle degeneration can occur. Previously we performed an array of genetic screens using a Drosophila model for muscular dystrophy in order to find novel DGC interactors aiming to elucidate the signaling role(s) in which the complex is involved. Since the function of the DGC in the brain and nervous system has not been fully defined, we have here continued to analyze the DGC modifiers' function in the developing Drosophila brain and eye.

Le complexe glycoprot&#xE9;ique dystrophine (DGC) est un grand complexe multi-composant qui est bien connu pour sa fonction dans le tissu musculaire. Lorsque les composants principaux de la GCR, la dystrophine (Dys) et dystroglycane (Dg) sont affect&#xE9;s d&#xE9;ficience cognitive et l&#x26;#39;arri&#xE9;ration mentale, en plus de la d&#xE9;g&#xE9;n&#xE9;rescence musculaire peut survenir. Auparavant nous avons effectu&#xE9; une s&#xE9;rie de cribles g&#xE9;n&#xE9;tiques utilisant un mod&#xE8;le de drosophile pour la dystrophie musculaire dans le but de trouver de nouveaux interacteurs GCR visant &#xE0; &#xE9;lucider le r&#xF4;le de signalisation (s) dans lequel le complexe est impliqu&#xE9;. Puisque la fonction de la GCR dans le cerveau et le syst&#xE8;me nerveux n&#x26;#39;a pas &#xE9;t&#xE9; pleinement d&#xE9;fini, nous avons continu&#xE9; ici pour analyser la fonction des modificateurs DGC &#xBB;dans le cerveau en d&#xE9;veloppement chez la drosophile et les yeux.

Das Dystrophin-Glykoprotein-Komplex (DGC) ist ein gro&szlig;er Mehrkomponenten Komplex, der bekannt f&uuml;r seine Funktion im Muskelgewebe ist. Wenn die Hauptkomponenten der DGC, Dystrophin (Dys) und Dystroglycan (Dg) sind betroffene kognitive Beeintr&auml;chtigung und Geistesverlangsamung neben Muskel-Degeneration kann auftreten. Zuvor f&uuml;hrten wir eine Reihe von genetischen Bildschirme mit einem Drosophila-Modell f&uuml;r Muskeldystrophie um neuartige DGC Inter-Akteure mit dem Ziel, die Signalisierung Rolle(n) zu erhellen, an denen der Komplex beteiligt ist, zu finden. Da die Funktion der DGC im Gehirn und Nervensystem nicht vollst&auml;ndig definiert wurde, haben wir hier weiterhin die DGC-Modifizierer-Funktion in der Taufliege sich entwickelnde Gehirn und Auge zu analysieren.

In precedenza abbiamo eseguito una serie di schermi genetici utilizzando un modello di Drosophila per distrofia muscolare al fine di trovare nuovi interattori DGC volti a delucidare i ruoli di segnalazione in cui &egrave; coinvolto il complesso. El complejo de glicoprote&#xED;na distrofina (DGC) es un gran multi-componente complejo que es bien conocido por su funci&#xF3;n en el tejido muscular. Cuando los componentes principales de la DGC, la distrofina (Dys) y distroglicano (DG) se ven afectados el deterioro cognitivo y retraso mental, adem&#xE1;s de la degeneraci&#xF3;n muscular puede ocurrir. Previamente se realiz&#xF3; una serie de pantallas gen&#xE9;ticos usando un modelo de Drosophila para la distrofia muscular con el fin de encontrar nuevos interactores DGC destinados a dilucidar el papel de se&#xF1;alizaci&#xF3;n (s) en que est&#xE1; implicado el complejo. Dado que la funci&#xF3;n de la DGC en el cerebro y el sistema nervioso no ha sido totalmente definido, hemos aqu&#xED; seguido analizando la funci&#xF3;n de los modificadores de DGC en el desarrollo de cerebro de Drosophila y los ojos.
