La interacci&oacute;n entre la ATP y alta K (+)-evocaba aumento en la concentraci&oacute;n de calcio intracelular libre ([Ca2 +] i) fue investigada para conocer el mecanismo de interacci&oacute;n de ATP con los canales de calcio voltaje-sensibles. [Ca2 +] i estaba medido en el modelo neuronal, neuroblastoma x c&eacute;lulas h&iacute;bridas de glioma (NG 108-15), utilizando el indicador de fluorescencia fura-2. En presencia de 1,8 mM Ca2 + extracelular, ATP inducida por un r&aacute;pido, dependiente de la concentraci&oacute;n aumento de [Ca2 +] i. alto K + (50 mM) evoc&oacute; a [Ca2 +] me levanto de 109 +/-11 nM a 387 + /-81 nM (n = 16). La aplicaci&oacute;n de cualquiera de estos dos [Ca2 +] i-aumento provocando agentes en secuencia con el otro causado deterioro del &uacute;ltimo efecto. La desensibilizaci&oacute;n mutua de las respuestas a ATP y alto K + sugiere que ambos agentes dependen al menos en parte la misma fuente de Ca2 + para la elevaci&oacute;n de la [Ca2 +] i en NG 108-15 c&eacute;lulas.

[Ca2 +], j&#39;ai mesur&eacute; dans le mod&egrave;le neuronal, neuroblastome x cellules de gliome hybrides (NG 108-15), &agrave; l&#39;aide de l&#39;indicateur de fluorescence fura-2. En pr&eacute;sence de 1,8 mM de Ca2 + extracellulaire, ATP induite par une rapide, augmentation de la concentration-d&eacute;pendante de [Ca2 +] i. riche en K + (50 mM) a &eacute;voqu&eacute; un [Ca2 +] i rise de 109 +/-11 nM &agrave; 387 +/-81 nM (n = 16). La demande d&#39;une de ces deux [Ca2 +] i-augmentation provoquant agents en s&eacute;quence avec l&#39;autre caus&eacute; atteinte &agrave; l&#39;effet de ce dernier. La d&eacute;sensibilisation mutuelle des r&eacute;ponses &agrave; l&#39;ATP et du K + &eacute;lev&eacute; donne &agrave; penser que les deux agents d&eacute;pendent au moins en partie la m&ecirc;me source de Ca2 + pour l&#39;&eacute;l&eacute;vation de [Ca2 +] i dans NG 108-15 cellules.

The interaction between ATP- and high K(+)-evoked increase in intracellular free calcium concentration ([Ca2+]i) was investigated to gain an insight into the mechanism of interaction of ATP with voltage-sensitive calcium channels. [Ca2+]i was measured in the neuronal model, neuroblastoma x glioma hybrid cells (NG 108-15), using the fluorescence indicator fura-2. In the presence of 1.8 mM extracellular Ca2+, ATP induced a rapid, concentration-dependent increase in [Ca2+]i. High K+ (50 mM) evoked a [Ca2+]i rise from 109 +/- 11 nM to 387 +/- 81 nM (n = 16). The application of either of these two [Ca2+]i-increase provoking agents in sequence with the other caused impairment of the latter effect. The mutual desensitization of the responses to ATP and high K+ strongly suggests that both agents rely at least in part on the same source of Ca2+ for elevation of [Ca2+]i in NG 108-15 cells.

In case of surgical procedures the bleeding risk under continued antiplatelet therapy has to be balanced against the risk of ischemic complications due to withdrawal of antiplatelet therapy. Whereas surgical procedures with a low bleeding risk may be performed with continued antiplatelet therapy, there is a need for partial or total discontinuation of antiplatelet therapy in surgical procedures with higher bleeding risks.

Many nocturnal insects depend on vision for daily life and have evolved different strategies to improve their visual capabilities in dim light. Neural summation of visual signals is one strategy to improve visual performance, and this is likely to be especially important for insects with apposition compound eyes. Here we develop a model to determine the optimum spatiotemporal sampling of natural scenes at gradually decreasing light levels. Image anisotropy has a strong influence on the receptive field properties predicted to be optimal at low light intensities. Spatial summation between visual channels is predicted to extend more strongly in the direction with higher correlations between the input signals. Increased spatiotemporal summation increases signal-to-noise ratio at low frequencies but sacrifices signal-to-noise ratio at higher frequencies. These results, while obtained from a model of the insect visual system, are likely to apply to visual systems in general.

The size distribution of neuronal avalanches in cortical networks has been reported to follow a power law distribution with exponent close to -1.5, which is a reflection of long-range spatial correlations in spontaneous neuronal activity. However, identifying power law scaling in empirical data can be difficult and sometimes controversial. In the present study, we tested the power law hypothesis for neuronal avalanches by using more stringent statistical analyses. In particular, we performed the following steps: (i) analysis of finite-size scaling to identify scale-free dynamics in neuronal avalanches, (ii) model parameter estimation to determine the specific exponent of the power law, and (iii) comparison of the power law to alternative model distributions. Consistent with critical state dynamics, avalanche size distributions exhibited robust scaling behavior in which the maximum avalanche size was limited only by the spatial extent of sampling ("finite size" effect). This scale-free dynamics suggests the power law as a model for the distribution of avalanche sizes. Using both the Kolmogorov-Smirnov statistic and a maximum likelihood approach, we found the slope to be close to -1.5, which is in line with previous reports. Finally, the power law model for neuronal avalanches was compared to the exponential and to various heavy-tail distributions based on the Kolmogorov-Smirnov distance and by using a log-likelihood ratio test. Both the power law distribution without and with exponential cut-off provided significantly better fits to the cluster size distributions in neuronal avalanches than the exponential, the lognormal and the gamma distribution. In summary, our findings strongly support the power law scaling in neuronal avalanches, providing further evidence for critical state dynamics in superficial layers of cortex.

Conform&eacute;ment &agrave; la dynamique de l&#39;&eacute;tat critique, distributions granulom&eacute;triques avalanche expos&eacute; comportement de mise &agrave; l&#39;&eacute;chelle robuste dont la taille maximale d&#39;avalanche &eacute;tait limit&eacute;e que par l&#39;&eacute;tendue spatiale de l&#39;&eacute;chantillonnage (effet &laquo; taille finie &raquo;). Cette dynamique sans &eacute;chelle sugg&egrave;re la Loi de puissance comme un mod&egrave;le pour la distribution des tailles de l&#39;avalanche. Utilisant la statistique de Kolmogorov-Smirnov et une approche du maximum de vraisemblance, nous avons trouv&eacute; la pente pour &ecirc;tre proche de -1.5, qui s&#39;inscrit dans les rapports pr&eacute;c&eacute;dents. Sowohl die Gesetz Energieverteilung ohne und mit exponentiellen Cut-Off deutlich bereitgestellt passt besser zu den Cluster-Gr&ouml;&szlig;e-Distributionen in neuronalen Lawinen als die Exponentialfunktion, die lognormal und Gamma-Verteilung. Zusammenfassend l&auml;sst sich sagen st&uuml;tzen unsere Erkenntnisse stark dem Potenzgesetz Skalierung in neuronalen Lawinen, eine weitere Beweise f&uuml;r kritischen Zustand Dynamics in oberfl&auml;chlichen Schichten des Cortex.

La distribuci&oacute;n de tama&ntilde;o de avalanchas neuronales en redes corticales se ha divulgado para seguir una distribuci&oacute;n de ley de potencia con exponente cerca de 1.5, que es un reflejo de las correlaciones espaciales a largo plazo en la actividad neuronal espont&aacute;nea. Sin embargo, identificar el derecho de poder escalar en datos emp&iacute;ricos puede ser dif&iacute;cil y a veces controversial. En el presente estudio, probamos la hip&oacute;tesis de ley de energ&iacute;a para avalanchas neuronales utilizando an&aacute;lisis estad&iacute;sticos m&aacute;s estrictos. En particular, realizamos los siguientes pasos: (i) el an&aacute;lisis de tama&ntilde;o finito escala para identificar la din&aacute;mica sin escala en avalanchas neuronales, estimaci&oacute;n de par&aacute;metros del modelo (ii) determinar el exponente espec&iacute;fico de la ley de energ&iacute;a y (iii) la comparaci&oacute;n de la ley de energ&iacute;a para las distribuciones de modelo alternativo. De acuerdo con la din&aacute;mica del estado cr&iacute;tico, distribuciones de tama&ntilde;o de avalancha exhibieron comportamiento escala robusto en el que el tama&ntilde;o m&aacute;ximo de la avalancha fue limitado s&oacute;lo por la extensi&oacute;n espacial de muestreo (efecto de &quot;tama&ntilde;o finito&quot;). Esta din&aacute;mica sin escala sugiere la ley de la energ&iacute;a como un modelo para la distribuci&oacute;n de tama&ntilde;os de la avalancha. In the striatal microcircuit, fast-spiking (FS) interneurons have an important role in mediating inhibition onto neighboring medium spiny (MS) projection neurons. In this study, we combined computational modeling with in vitro and in vivo electrophysiological measurements to investigate FS cells in terms of their discharge properties and their synaptic efficacies onto MS neurons. In vivo firing of striatal FS interneurons is characterized by a high firing variability. It is not known, however, if this variability results from the input that FS cells receive, or if it is promoted by the stuttering spike behavior of these neurons. Both our model and measurements in vitro show that FS neurons that exhibit random stuttering discharge in response to steady depolarization do not show the typical stuttering behavior when they receive fluctuating input. Importantly, our model predicts that electrically coupled FS cells show substantial spike synchronization only when they are in the stuttering regime. Therefore, together with the lack of synchronized firing of striatal FS interneurons that has been reported in vivo, these results suggest that neighboring FS neurons are not in the stuttering regime simultaneously and that in vivo FS firing variability is more likely determined by the input fluctuations. Furthermore, the variability in FS firing is translated to variability in the postsynaptic amplitudes in MS neurons due to the strong synaptic depression of the FS-to-MS synapse. Our results support the idea that these synapses operate over a wide range from strongly depressed to almost fully recovered. The strong inhibitory effects that FS cells can impose on their postsynaptic targets, and the fact that the FS-to-MS synapse model showed substantial depression over extended periods of time might indicate the importance of cooperative effects of multiple presynaptic FS interneurons and the precise orchestration of their activity.

Striatal fast-spiking interneurons: from firing patterns to postsynaptic impact.

En el microcircuito estriada, interneuronas (FS) de r&aacute;pido-clavar tienen un papel importante en la mediaci&oacute;n de inhibici&oacute;n en la vecina medianas neuronas espinosas de proyecci&oacute;n (MS). En este estudio, combinamos modelado computacional con medidas electrofisiol&oacute;gicas in vitro e in vivo para investigar c&eacute;lulas FS en cuanto a sus propiedades de descarga y sus eficacias sin&aacute;pticas en neuronas MS. Cocci&oacute;n en vivo de interneurons estriada FS se caracteriza por una disparo alta variabilidad. Se desconoce, sin embargo, si esta variabilidad es resultado de la entrada que reciben las c&eacute;lulas FS, o si se promueve por el comportamiento de espiga tartamudeo de estas neuronas. Tanto nuestro modelo y mediciones in vitro muestran que las neuronas de FS que exhiben al azar descarga intermitente en respuesta a la despolarizaci&oacute;n constante no muestran el comportamiento t&iacute;pico del tartamudeo cuando reciben entrada fluctuante. Lo importante, nuestro modelo predice que el&eacute;ctricamente acopladas FS c&eacute;lulas muestran sincronizaci&oacute;n punto sustancial s&oacute;lo cuando est&eacute;n en el r&eacute;gimen de tartamudeo. Por lo tanto, junto con la falta de disparo sincronizado de interneurons de FS estriado que se ha divulgado en vivo, estos resultados sugieren que las neuronas vecinas de FS no est&aacute;n en el r&eacute;gimen intermitente simult&aacute;neamente y que variabilidad de le&ntilde;a FS in vivo m&aacute;s probable es que se determina por las fluctuaciones de la entrada. Adem&aacute;s, la variabilidad en la cocci&oacute;n de FS se traduce a la variabilidad en las amplitudes postsin&aacute;pticas en neuronas de MS debido a la fuerte depresi&oacute;n sin&aacute;ptica de la sinapsis de FS-a-MS. Nuestros resultados apoyan la idea de que estas sinapsis funcionan sobre una amplia gama de fuertemente deprimido casi totalmente recuperado. Los efectos inhibitorios fuertes que c&eacute;lulas FS pueden imponer a sus objetivos postsin&aacute;pticos, y el hecho de que el FS-a-MS sinapsis modelo demostr&oacute; sustancial depresi&oacute;n durante largos per&iacute;odos de tiempo podr&iacute;an indicar la importancia de la cooperaci&oacute;n efectos de varias interneuronas presin&aacute;pticas de FS y la orquestaci&oacute;n precisa de su actividad.

The proteins showing significantly decreased abundance after oral 6-gingerol stimulation were identified as glutathione S-transferase P, the heat shock protein Î²-1, the heat shock 70 kDa protein 1, annexin A1, and cytoplasmic Î²-actin, whereas prolactin inducible proteins (PIP), short palate, lung and nasal epithelium carcinoma-associated protein 2 (SPLUNC2), zinc-Î±-2-glycoproteins (Zn-Î±-GP), and carbonic anhydrase VI (CAVI) were found with increased abundance. As the effects of this study were observed instantaneously upon stimulation, any proteome modulation is very likely to result from the release of proteins from preformed vesicles and not from de novo synthesis. The elevated levels of SPLUNC2, Zn-Î±-GP, and CAVI might be interpreted to trigger innate protective mechanisms in mucosal immunity and in nonimmune mucosal defense and might play an important role during the initial stage of inflammation.

Flujo salival inducida por 6-gingerol (acre), hidroxi-&alpha;/&beta;-sanshools (hormigueo) y c&iacute;trico &aacute;cido (amargo) se midi&oacute; y se analizaron los cambios dependientes del tiempo en el proteoma de saliva todo por medio de 2D-PAGE, seguida por la digesti&oacute;n tr&iacute;ptica en gel y an&aacute;lisis de huellas dactilares masa MALDI-TOF-MS p&eacute;ptido. Las prote&iacute;nas que muestran abundancia perceptiblemente disminuida despu&eacute;s de estimulaci&oacute;n oral 6-gingerol fueron identificados como glutati&oacute;n S-transferasa P, el calor choque prote&iacute;na &beta;-1, el calor choque 70 kDa prote&iacute;na 1, anexina A1 y citopl&aacute;smica &beta;-actina, mientras que corta las prote&iacute;nas inducibles de prolactina (PIP), boca, pulm&oacute;n y prote&iacute;na asociada a carcinoma de epitelio nasal 2 (SPLUNC2), cinc-&alpha;-2-glicoprote&iacute;nas (GP-&alpha;-Zn) y anhidrasa carb&oacute;nica VI (CAVI) se encontraron con mayor abundancia. Como los efectos de este estudio se observaron instant&aacute;neamente sobre el est&iacute;mulo, cualquier modulaci&oacute;n del proteoma es muy probable que el resultado de la liberaci&oacute;n de prote&iacute;nas de ves&iacute;culas preformadas y no de s&iacute;ntesis de novo. Los niveles elevados de SPLUNC2, Zn-&alpha;-GP y CAVI podr&iacute;an interpretarse para activar mecanismos de protecci&oacute;n innatos en Inmunidad mucosal y en defensa mucosa no inmunitaria y podr&iacute;an desempe&ntilde;ar un papel importante durante la etapa inicial de la inflamaci&oacute;n.
