Circadian rhythms in rodents respond to arousing, nonphotic stimuli that contribute to daily patterns of entrainment. To examine whether the motivational significance of a stimulus is important for eliciting nonphotic circadian phase shirts in Syrian hamsters (Mesocricetus auratus), the authors compared responses to a highly rewarding stimulus (lateral hypothalamic brain stimulation reward [BSR]) and a highly aversive stimulus (footshock). Animals were housed on a 14:10-hr light-dark cycle until test day, when they were given a 1-hr BSR session (trained animals) or a 1-mA electric footshock at 1 of 8 circadian times, and were maintained in constant dark thereafter. Both BSR pulses and footshock produced nonphotic phase response curves. These results support the hypothesis that arousal resulting from the motivational significance of a stimulus is a major factor in nonphotic phase shifts.

Nonphotic phase shifts of the circadian clock in mammals are mediated by the intergeniculate leaflet (IGL) of the thalamus via a geniculohypothalamic projection to the suprachiasmatic nucleus. These shifts can be induced by arousing stimuli, such as wheel running, brain stimulation reward and foot shock. Because mesopontine cholinergic neurons are also activated by arousing stimuli, we tested the hypothesis that cholinergic input to the IGL mediates nonphotic phase shifts. Carbachol injected into the IGL of hamsters in their subjective day (CT8) induced phase advances similar to shifts that are induced by arousal at the same circadian time. Control injections of saline at CT8 did not advance phase similarly. Carbachol injections outside the IGL produced smaller shifts. Pre-injections of the muscarinic antagonist, atropine, reduced carbachol-induced phase advances relative to saline pre-injections. The results indicate that muscarinic input to the IGL can induce nonphotic phase shifts.

Feeding schedules that restrict food access to a predictable daytime meal induce in rodents food-anticipatory behaviors, changes in physiological rhythms and shifts in the rhythm of clock gene expression in the brain and periphery. However, little is known about the effects of nighttime restricted feeding. Previously, we showed that daytime restricted access to a highly palatable complete meal replacement, Ensure Plus (Ensure), shifts the rhythm of expression of the clock protein PER2 in limbic forebrain areas including the oval nucleus of the bed nucleus of the stria terminalis (BNSTov), central nucleus of the amygdala (CEA), basolateral amygdala (BLA) and dentate gyrus (DG), and induces a rhythm in the dorsomedial hypothalamic nucleus (DMH) in food deprived (restricted feeding), but not free-fed rats (restricted treat). In the present study we investigated the effects of nighttime restricted feeding (Ensure only, 2 h/night) and nighttime restricted treats (Ensure 2 h/night+free access to chow) in order to determine whether these effects were dependent on the time of day the meal was provided. We found that nighttime restricted feeding, like daytime restricted feeding, shifted the rhythm of PER2 expression in the BNSTov and CEA and peak expression was observed approximately 12 h after the mealtime. Also consistent with previous work, nighttime restricted feeding induced a rhythm of PER2 expression in the DMH and these effects occurred without affecting the rhythm in the suprachiasmatic nucleus (SCN). In contrast to previous work with daytime restricted feeding, nighttime restricted feeding had no effect on PER2 rhythms in the BLA and DG. Finally, nighttime restricted treats, as was the case for daytime restricted treats, had no effect on PER2 expression in any of the brain areas studied. The present results together with our previous findings show that the effect of restricted feeding on PER2 rhythms in the limbic forebrain and hypothalamus depend on a negative energy balance and vary as a function of time of day in a brain region-specific manner.

Horarios de alimentaci&oacute;n que restringen el acceso a los alimentos para comer durante el d&iacute;a predecible inducen conductas alimentaria-anticipaci&oacute;n de roedores, cambios en los ritmos fisiol&oacute;gicos y cambios en el ritmo de la expresi&oacute;n g&eacute;nica de reloj en el cerebro y la periferia. Sin embargo, poco se sabe sobre los efectos de la alimentaci&oacute;n restringida durante la noche. Previamente, mostramos ese acceso restringido durante el d&iacute;a a un reemplazo de comida completa altamente palatables, asegurar Plus (asegurar), cambia el ritmo de la expresi&oacute;n de la prote&iacute;na del reloj PER2 en &aacute;reas l&iacute;mbicas prosenc&eacute;falo incluyendo el n&uacute;cleo oval del n&uacute;cleo de la cama de la estr&iacute;a terminal (BNSTov), el n&uacute;cleo central de la am&iacute;gdala (CEA), la am&iacute;gdala basolateral (BLA) y el giro dentado (DG) e induce un ritmo en el n&uacute;cleo hipotal&aacute;mico dorsomedial (DMH) en alimentos privados (alimentaci&oacute;n restringida), pero no libre consumieron (convite restringida). En el presente estudio hemos investigado los efectos de noche restringido de alimentaci&oacute;n (s&oacute;lo Aseg&uacute;rese, 2 h/noche) y noche restricci&oacute;n golosinas (2 h / noche + acceso gratuito a chow asegurar) para determinar si estos efectos depend&iacute;an de la hora del d&iacute;a, la comida fue proporcionada. Encontramos que la alimentaci&oacute;n restringida durante la noche, como alimentaci&oacute;n restringida durante el d&iacute;a, cambi&oacute; el ritmo de la expresi&oacute;n de PER2 en el BNSTov y el CEA y expresi&oacute;n de pico se observ&oacute; aproximadamente 12 h despu&eacute;s de la hora de la comida. Tambi&eacute;n de acuerdo con trabajos previos, noche restringido alimentaci&oacute;n inducida por un ritmo de expresi&oacute;n PER2 en el DMH y estos efectos se produjeron sin afectar el ritmo en el n&uacute;cleo supraquiasm&aacute;tico (SCN). A diferencia de trabajos anteriores con alimentaci&oacute;n restringida durante el d&iacute;a, amamantar restringida no tuvo efecto sobre ritmos PER2 en la direcci&oacute;n general y BLA. Finalmente, trata restringida durante la noche, como fue el caso de golosinas restringidos durante el d&iacute;a, no tuvo efecto sobre PER2 expresi&oacute;n en cualquiera de las &aacute;reas del cerebro estudiadas. Estos resultados junto con los anteriores resultados muestran que el efecto de la alimentaci&oacute;n restringida en PER2 ritmos en el prosenc&eacute;falo l&iacute;mbico y el hipot&aacute;lamo dependen de una energ&iacute;a negativa del balance y var&iacute;an en funci&oacute;n de la hora del d&iacute;a en la manera de regi&oacute;n espec&iacute;fica del cerebro.

When food availability is restricted to a single time of day, circadian rhythms of behavior and physiology in rodents shift to anticipate the predictable time of food arrival. It has been hypothesized that certain food-anticipatory rhythms are linked to the induction and entrainment of rhythms in clock gene expression in the dorsomedial hypothalamic nucleus (DMH), a putative food-entrained circadian oscillator. To study this concept further, we made food availability unpredictable by presenting the meal at a random time each day (variable restricted feeding, VRF), either during the day, night or throughout the 24-h cycle. Wheel running activity and the expression of the clock protein, Period1 (PER1), in the DMH and the suprachiasmatic nucleus (SCN) were assessed. Rats exhibited increased levels of activity during the portion of the day when food was randomly presented but, as expected, failed to entrain anticipatory wheel running activity to a single time of day. PER1 expression in the SCN was unchanged by VRF schedules. In the DMH, PER1 expression became rhythmic, peaking at opposite times of day in rats fed only during the day or during the night. In rats fed randomly throughout the entire 24-h cycle, PER1 expression in the DMH remained arrhythmic, but was elevated. These results demonstrate that VRF schedules confined to the day or night can induce circadian rhythms of clock gene expression in the DMH. Such feeding schedules cannot entrain behavioral rhythms, thereby showing that food-entrainment of behavior and circadian rhythms of clock gene expression in the DMH are dissociable.

Es ist vermutet worden, dass bestimmte Lebensmittel &ndash; antizipativen Rhythmen der Induktion und Mitnahme von Rhythmen im Uhr-Genexpression im Dorsomedial hypothalamischen Kern (DMH), ein mutma&szlig;liches Essen mitgef&uuml;hrter circadian Oszillator verbunden sind. Um dieses Konzept zu studieren hat weitere, wir essen Verf&uuml;gbarkeit unvorhersehbar durch Vorlage der Mahlzeit zu einem beliebigen Zeitpunkt jeden Tag (Variable beschr&auml;nkt F&uuml;tterung, VRF), entweder w&auml;hrend des Tages, der Nacht oder w&auml;hrend des 24-h-Zyklus. Rad-Aktivit&auml;t und der Ausdruck des Proteins Uhr, Dauer1 ausgef&uuml;hrt (PER1), in der DMH und die gefundenes Kern (SCN) bewertet wurden. Ratten zeigte erh&ouml;hte Niveaus der Aktivit&auml;t w&auml;hrend des Teils des Tages beim Essen war zuf&auml;llig dargestellt aber, wie erwartet, Fehler beim Verladen von vorausschauenden Rad Aktivit&auml;t zu einer einzigen Tageszeit ausgef&uuml;hrt. PER1 Ausdruck in der SCN wurde durch VRF Zeitpl&auml;ne nicht ge&auml;ndert. Die DMH wurde PER1 Ausdruck rhythmische, H&ouml;chststand zu anderen Tageszeiten bei Ratten gef&uuml;ttert, nur tags&uuml;ber oder nachts. Bei Ratten Tiere zuf&auml;llig &uuml;ber den gesamten 24-h-Zyklus PER1 Ausdruck in der DMH blieb arrhythmic, aber wurde erhoben. Diese Ergebnisse zeigen, dass VRF Zeitpl&auml;ne beschr&auml;nkt sich auf Tages- und Nachtzeit Tagesrhythmus der Uhr-Genexpression in der DMH hervorrufen k&ouml;nnen. Solche F&uuml;tterung Zeitpl&auml;ne k&ouml;nnen nicht verladen Verhaltens Rhythmen, und damit zeigen, dass Lebensmittel-Mitnahme von Verhalten und Tagesrhythmus der Uhr-Genexpression in der DMH sind transponieren.

Quando la disponibilit&agrave; di cibo &egrave; limitata a un singolo momento della giornata, i ritmi circadiani del comportamento e della fisiologia in MAIUSC roditori ad anticipare il tempo prevedibile di arrivo di cibo. &Egrave; stato ipotizzato che alcuni ritmi alimentari-preventivo sono collegati all&#39;induzione e trascinamento dei ritmi nell&#39;espressione del gene dell&#39;orologio nel nucleo dorsomedial ipotalamico (DMH), un putativo oscillatore circadiano trascinato dal cibo. Ratti esposti aumento dei livelli di attivit&agrave; durante parte della giornata quando cibo era casualmente presentato ma, come previsto, non &egrave; riuscito a salire sul treno della rotella preventiva attivit&agrave; di un singolo momento della giornata. Cuando la disponibilidad de alimento se limita a una sola hora del d&iacute;a, los ritmos circadianos del comportamiento y la fisiolog&iacute;a en cambio de roedores a anticipar el tiempo previsible de la llegada de alimentos. Se ha planteado la hip&oacute;tesis de que ciertos alimentos-anticipaci&oacute;n ritmos est&aacute;n vinculados a la inducci&oacute;n y el arrastre de ritmos en la expresi&oacute;n g&eacute;nica de reloj en el n&uacute;cleo hipotal&aacute;mico dorsomedial (DMH), un supuesta oscilador circadiano alimentos-arrastrado. Para estudiar este concepto adem&aacute;s, hicimos la disponibilidad de alimento imprevisible presentando la comida en un tiempo aleatorio cada d&iacute;a (variable restringida alimentaci&oacute;n, VRF), durante el d&iacute;a, noche o durante todo el ciclo de 24 horas. Corriente de la actividad y la expresi&oacute;n de la prote&iacute;na del reloj, Period1 de la rueda (PER1), en el DMH y el supraquiasm&aacute;tico n&uacute;cleo (SCN) se evaluaron. Ratas exhibieron mayores niveles de actividad durante parte del d&iacute;a cuando alimentos fue al azar present&oacute; pero, como era de esperar, no arrastrar la corriente de actividad a una sola hora del d&iacute;a de la rueda de anticipaci&oacute;n. PER1 expresi&oacute;n en el SCN fue sin cambios de horarios VRF. En el DMH, PER1 expresi&oacute;n lleg&oacute; a ser r&iacute;tmico, alcanzando un m&aacute;ximo de horas enfrente del d&iacute;a en las ratas alimentadas con s&oacute;lo durante el d&iacute;a o durante la noche. En las ratas alimentadas al azar durante todo el ciclo de 24 horas, PER1 expresi&oacute;n en el DMH permaneci&oacute; arr&iacute;tmico, pero fue elevado. Estos resultados demuestran que horarios VRF confinados al d&iacute;a o de noche pueden inducir ritmos circadianos de la expresi&oacute;n g&eacute;nica de reloj en el DMH. Estos horarios de alimentaci&oacute;n no pueden arrastrar ritmos de comportamiento, lo que demostrar&iacute;a eso alimento-arrastre de comportamiento y los ritmos circadianos de la expresi&oacute;n g&eacute;nica de reloj en el DMH son disociables.

A role for dopamine (DA) in the regulation of clock genes in the mammalian brain is suggested by evidence that manipulations of DA receptors can alter the expression of some clock genes outside the suprachiasmatic nucleus (SCN), the master circadian clock. The role of endogenous DA in the regulation of clock gene expression is unknown. Here, we demonstrate a direct relationship between extracellular DA levels and the rhythm of expression of the clock protein PERIOD2 (PER2) in the dorsal striatum of the male Wistar rat. Specifically, we show that the peak of the daily rhythm of extracellular DA in the dorsal striatum precedes the peak of PER2 by âˆ¼6 h and that depletion of striatal DA by 6-hydroxydopamine or Î±-methyl-para-tyrosine or blockade of D(2) DA receptors by raclopride blunts the rhythm of striatal PER2. Furthermore, timed daily activation of D(2) DA receptors, but not D(1) DA receptors, restores and entrains the PER2 rhythm in the DA-depleted striatum. None of these manipulations had any effect on the PER2 rhythm in the SCN. Our findings are consistent with the idea that the rhythm of expression of PER2 in the dorsal striatum depends on daily dopaminergic activation of D(2) DA receptors. These observations may have implications for circadian abnormalities seen in Parkinson's disease.

Eine Rolle f&uuml;r Dopamin (DA) bei der Regulierung der Uhr Gene im Gehirn von S&auml;ugetieren empfiehlt sich durch Beweise, Manipulationen von DA-Rezeptoren die Expression von einigen Genen Uhr au&szlig;erhalb der gefundenes Kern (SCN), master-circadian Clock &auml;ndern k&ouml;nnen. Die Rolle des endogenen DA bei der Regulierung der Genexpression Uhr ist unbekannt. Hier zeigen wir eine direkte Beziehung zwischen extrazellul&auml;ren DA Ebenen und den Rhythmus der Ausdruck des Proteins Uhr 19992 (2Mio.) im dorsalen Striatum der m&auml;nnlichen Wistar Ratte. Insbesondere zeigen wir, dass der Gipfel der t&auml;gliche Rhythmus des extrazellul&auml;ren DA im dorsalen Striatum von &sim;6 h den Gipfel 2Mio. vorausgeht und Ersch&ouml;pfung der Striatale DA 6-Hydroxydopamine oder &alpha;-Methyl-Para-Tyrosin oder Blockade der Vorschu&szlig; DA-Rezeptoren durch Raclopride im Rhythmus der Striatale 2Mio. stumpft. Dar&uuml;ber hinaus zeitlich t&auml;gliche Aktivierung der Vorschu&szlig; DA-Rezeptoren, aber nicht D(1) DA Rezeptoren, Wiederherstellungen und bindet den 2Mio. Rhythmus im Striatum DA ersch&ouml;pft. Keiner von diesen Manipulationen hatte keine Auswirkung auf den 2Mio. Rhythmus in der SCN. Unsere Ergebnisse sind konsistent mit der Idee, dass der Rhythmus des Ausdrucks von 2Mio. im dorsalen Striatum t&auml;gliche dopaminerge Aktivierung Vorschu&szlig; DA Rezeptoren abh&auml;ngt. Predictable restricted feeding schedules limit food availability to a single meal at the same time each day, lead to the induction and entrainment of circadian rhythms in food-anticipatory activity, and shift daily rhythms of clock gene expression in areas of the brain that are important in the regulation of motivational and emotional state. In contrast, when food is delivered under a variable restricted feeding (VRF) schedule, at a new and unpredictable mealtime each day, circadian rhythms in food-anticipatory activity fail to develop. Here, we study the effects of VRF on the daily rhythm of plasma corticosterone and of clock gene expression in the limbic forebrain and dorsal striatum, of rats provided a 2-h access to a complete meal replacement (Ensure Plus) at an unpredictable time each day. VRF schedules varied the mealtimes within the 12Â h of light (daytime VRF), the 12Â h of dark (nighttime VRF), or across the 24Â h light-dark cycle (anytime VRF). Our results show that contrary to the synchronizing effects of predictable restricted feeding, VRF blunts the daily corticosterone rhythm and disrupts daily rhythms of PER2 expression in a region-specific and mealtime-dependent manner.

Alimentaci&oacute;n restringida predecible horarios l&iacute;mite la disponibilidad de alimento para una sola comida al mismo tiempo cada d&iacute;a, llevan a la inducci&oacute;n y el arrastre de los ritmos circadianos en actividad alimentaria-anticipaci&oacute;n y ritmos diarios de cambio de la expresi&oacute;n g&eacute;nica de reloj en &aacute;reas del cerebro que son importantes en la regulaci&oacute;n del estado motivacional y emocional. Por el contrario, cuando la comida se entrega bajo un programa de alimentaci&oacute;n restringida variable (VRF), en un nuevo e impredecible comida cada d&iacute;a, los ritmos circadianos en actividad alimentaria-anticipaci&oacute;n no desarrollar. Aqu&iacute;, estudiamos los efectos de VRF en el ritmo diario de corticosterona de plasma y de la expresi&oacute;n g&eacute;nica de reloj en el prosenc&eacute;falo l&iacute;mbica y estriado dorsal, de ratas proporcionada un acceso 2-h a un reemplazo de comida completa (asegurar m&aacute;s) en un momento impredecible cada d&iacute;a. VRF horarios variaron las comidas dentro de las 12 h de luz (VRF diurna), las 12 h de oscuridad (noche VRF), o a trav&eacute;s de las 24 h luz / oscuridad ciclo (en cualquier momento VRF). Nuestros resultados muestran que contrariamente a los efectos de sincronizaci&oacute;n de alimentaci&oacute;n restringida predecible, VRF embota el ritmo diario de corticosterona y perturba ritmos diarios de PER2 expresi&oacute;n de manera espec&iacute;ficas para la regi&oacute;n y dependiente de las comidas.

Alimentaci&oacute;n restringida variable altera las oscilaciones diarias de la expresi&oacute;n de Period2 en el forebrain l&iacute;mbico y estriado dorsal en ratas.
