&#x4F7F;&#x7528; multisubtelomere &#x9C7C;&#x7684;&#x7EC8;&#x7AEF;&#x67D3;&#x8272;&#x4F53;&#x5F02;&#x5E38;&#x7684;&#x8868;&#x5F81;&#x3002;

Repetici&#xF3;n de secuencias telom&#xE9;ricas (TTAGGG) son conocidos para tapar los extremos de cada cromosoma humano. Proximal a estas secuencias repetidas son cromos&#xF3;micas espec&#xED;ficas secuencias de repetici&#xF3;n, que a su vez son distal al gen regiones ricas. Submicrosc&#xF3;pica, anormalidades sutiles, o cr&#xED;ptico en estas regiones pueden ahora ser investigado utilizando juegos comerciales de la sonda para todos los subtelomeric regiones espec&#xED;ficas del cromosoma del genoma humano. Utilizando esta tecnolog&#xED;a, el desequilibrio gen&#xF3;mico previamente no identificado se ha encontrado en una proporci&#xF3;n de pacientes con retraso del desarrollo idiop&#xE1;tico y dificultades de aprendizaje. Hemos utilizado estos conjuntos de la sonda para investigar los casos con anomal&#xED;as detectadas en la terminal al parecer, el an&#xE1;lisis cromos&#xF3;mico G-bandas. Telomeric repeat sequences (TTAGGG) are known to cap the termini of every human chromosome. Proximal to these repeat sequences are chromosome-specific repeat sequences, which in turn are distal to gene-rich regions. Submicroscopic, subtle, or cryptic abnormalities in these regions can now be investigated using commercial probe sets for all of the chromosome-specific subtelomeric regions of the human genome. Using this technology, previously unidentified genomic imbalance has been found in a proportion of patients with idiopathic developmental delay and learning difficulties. We have used these probe sets to investigate cases with apparently terminal anomalies detected on G-banded chromosome analysis. As a result of such investigations, we have found that 3 (19%) of 16 apparently terminal deletion cases were the result of more complex rearrangements involving other chromosome subtelomeres. The remaining 13 cases contained no chromosome-specific subtelomere repeats on the deleted arm, but in all 16 cases, the TTAGGG telomere repeat cap was present. A further case was investigated where extra material was found in the terminal region of the chromosome 12 short arm, found to represent a complex inversion/duplication/deletion rearrangement. Investigation of all cases with terminal anomalies, including apparently terminal deletions, is likely to uncover further cases involving complex rearrangements and should lead to a greater understanding of the mechanisms by which these abnormalities arise.

Characterization of terminal chromosome anomalies using multisubtelomere FISH.

S&#xE9;quences r&#xE9;p&#xE9;t&#xE9;es t&#xE9;lom&#xE9;riques (TTAGGG) sont connus pour plafonner les extr&#xE9;mit&#xE9;s de chaque chromosome humain. Proximale par rapport &#xE0; ces s&#xE9;quences r&#xE9;p&#xE9;t&#xE9;es sont des s&#xE9;quences chromosomiques sp&#xE9;cifiques &#xE0; r&#xE9;p&#xE9;tition, qui sont &#xE0; leur tour distale au g&#xE8;ne des r&#xE9;gions riches. Les 13 cas restants ne contenait pas de r&#xE9;p&#xE9;titions subt&#xE9;lom&#xE9;riques sp&#xE9;cifiques du chromosome sur le bras supprim&#xE9;, mais dans tous les 16 cas, le bouchon de r&#xE9;p&#xE9;tition TTAGGG des t&#xE9;lom&#xE8;res &#xE9;tait pr&#xE9;sent. Un autre cas a &#xE9;t&#xE9; &#xE9;tudi&#xE9;, o&#xF9; du mat&#xE9;riel suppl&#xE9;mentaire a &#xE9;t&#xE9; trouv&#xE9; dans la r&#xE9;gion terminale du chromosome 12 bras court, a trouv&#xE9; pour repr&#xE9;senter une inversion complexe / duplication / suppression r&#xE9;arrangement. Sequenze ripetute telomeriche (TTAGGG) sono noti per cap i termini di ogni cromosoma umano. Prossimale di queste sequenze ripetute sono ripetizione sequenze specifiche del cromosoma, che a loro volta sono distale di regioni ricche di gene. Anomalie submicroscopiche, sottile o criptici in queste regioni ora possono essere studiate mediante sonda commerciale imposta per tutte le regioni cromosomiche specifiche subtelomerica del genoma umano. Utilizzando questa tecnologia, squilibrio genomico in precedenza non identificato &egrave; stato trovato in una proporzione di pazienti con idiopatica ritardo dello sviluppo e difficolt&agrave; di apprendimento. Abbiamo usato questi set sonda per indagare sui casi con anomalie apparentemente terminale rilevati su analisi cromosomica G-fasciato. A seguito di tali indagini, abbiamo trovato che 3 (19%) di 16 casi di eliminazione terminale apparentemente erano il risultato di riarrangiamenti pi&ugrave; complessi che coinvolgono altri subtelomerici cromosoma. I restanti 13 casi non contenevano ripete subtelomeriche cromosoma-specifiche sul braccio eliminato, ma in tutti i casi di 16, il tappo di ripetizione del telomero TTAGGG era presente. Un caso ulteriore fu indagato dove &egrave; stato trovato materiale extra nella regione terminale del braccio corto del cromosoma 12, trovato per rappresentare un complesso riarrangiamento di inversione/duplicazione/cancellazione. Indagine di tutti i casi con anomalie terminale, compreso apparentemente terminale delezioni, &egrave; probabile che scoprire ulteriori casi di riarrangiamenti complessi e dovrebbe portare ad una maggiore comprensione dei meccanismi da cui derivano queste anomalie.
