The photoreceptor cells of the retina are subject to a wide range of genetic diseases. This review summarizes current knowledge regarding an important group of retinal diseases caused by mutations in photoreceptor-enriched genes. In addition, progress toward treatment of a variety of these diseases in animal models via adeno-associated virus gene therapy is described. Although no human trials have yet been initiated to treat diseases caused by mutations in photoreceptor-enriched genes, there is a great deal of optimism regarding the prospects of treating these diseases using adeno-associated virus gene therapy.

The transcription factor neural retina leucine zipper (Nrl) is a critical determinant of rod photoreceptor cell fate and a key regulator of rod differentiation. Nrl(-/-) rod precursors fail to turn on rod genes and instead differentiate as cones. Furthermore, NRL mutations in humans cause retinitis pigmentosa. Despite the developmental and clinical significance of this gene, little is known about the transcriptional regulation of Nrl itself. In this study, we sought to define the cis- and trans-acting factors responsible for initiation and maintenance of Nrl transcription in the mouse retina. Utilizing a quantitative mouse retinal explant electroporation assay, we discovered a phylogenetically conserved, 30-base pair region immediately upstream of the transcription start site that is required for Nrl promoter activity. This region contains binding sites for the retinal transcription factors CRX, OTX2, and RORÎ², and point mutations in these sites completely abolish promoter activity in living retinas. Gel-shift experiments show that CRX, OTX2, and RORÎ² can bind to the critical region in vitro, whereas ChIP experiments demonstrate binding of CRX and OTX2 to the critical region in vivo. Thus, our results indicate that CRX, OTX2, and RORÎ² directly regulate Nrl transcription by binding to critical sites within the Nrl promoter. We propose a model in which Nrl expression is primarily initiated by OTX2 and RORÎ² and later maintained at high levels by CRX and RORÎ².

Le facteur de transcription neuronale r&#xE9;tine leucine zipper (LNR) est un facteur d&#xE9;terminant du destin des cellules photor&#xE9;ceptrices tige et un r&#xE9;gulateur cl&#xE9; de la diff&#xE9;renciation tige. NRL (- / -) pr&#xE9;curseurs de tige ne parviennent pas &#xE0; activer des g&#xE8;nes de tige et au lieu de diff&#xE9;rencier les c&#xF4;nes. En outre, les mutations chez l&#x26;#39;homme NRL provoquer une r&#xE9;tinite pigmentaire. Malgr&#xE9; l&#x26;#39;importance d&#xE9;veloppementale et clinique de ce g&#xE8;ne, on sait peu au sujet de la r&#xE9;gulation transcriptionnelle de Nrl lui-m&#xEA;me. Dans cette &#xE9;tude, nous avons cherch&#xE9; &#xE0; d&#xE9;finir les facteurs cis-et trans-action responsables de l&#x26;#39;initiation et le maintien de la transcription Nrl dans la r&#xE9;tine de la souris. L&#x26;#39;utilisation d&#x26;#39;une souris quantitative r&#xE9;tinienne dosage &#xE9;lectroporation explant, nous avons d&#xE9;couvert un point de vue phylog&#xE9;n&#xE9;tique conserv&#xE9;e, la r&#xE9;gion 30-paire de base imm&#xE9;diatement en amont du site d&#x26;#39;initiation de la transcription qui est n&#xE9;cessaire pour l&#x26;#39;activit&#xE9; du promoteur de la LNR. Cette r&#xE9;gion contient des sites de liaison pour les facteurs de transcription r&#xE9;tinienne CRX, OTX2, et ROR&#x3B2;, et des mutations ponctuelles dans ces sites d&#x26;#39;abolir totalement l&#x26;#39;activit&#xE9; du promoteur dans les r&#xE9;tines de vie. Gel-quarts exp&#xE9;riences montrent que CRX, OTX2, et ROR&#x3B2; peut se lier &#xE0; la r&#xE9;gion critique in vitro, alors que des exp&#xE9;riences ChIP d&#xE9;montrer la liaison de CRX et OTX2 &#xE0; la r&#xE9;gion critique in vivo. Ainsi, nos r&#xE9;sultats indiquent que CRX, OTX2, et ROR&#x3B2; directement r&#xE9;guler la transcription en se liant &#xE0; Nrl sites de critiques au sein du promoteur de la LNR. In questo studio, abbiamo cercato di definire i fattori di cis - e trans-acting responsabili dell&#39;iniziazione e la manutenzione di Nrl trascrizione nella retina del mouse.
