1. Animals
<ol>
	<li>Procure C57BL/6 mice from the in-house mouse colony. 
	NOTE: Male mice 5-8 months of age at the start of the study were used. Daily running activity peaks and plateaus at around 9-10 weeks of age26. Previous studies have demonstrated that old mice (22-24 months) will also perform loaded wheel running27.</li>
	<li>House the mice individually in a standard rodent cage with a wire lid and keep the cage in a controlled environment (20-24 &#176;C with a 12:12 h light:dark cycle).</li>
	<li>Provide standard rodent chow and water ad libitum.</li>
</ol>
2. Running wheel apparatus
<ol>
	<li>Running wheel setup: 
	NOTE: Running wheels are assembled/set up similarly for all running protocols, except for adding 1 g or 2.5 g load magnets.
	<ol>
		<li>Glue a single 1 g sensor magnet to the outer middle circumference of the running wheel (Figure 1).</li>
		<li>Use this wheel with a single 1 g sensor magnet for only the first week of wheel acclimation.</li>
		<li>Loaded wheel running (LWR; identical loading protocol to PoWeR17): Follow steps 2.1.4-2.1.6.</li>
		<li>Week 2 for LWR requires 2 g of load (see Table 1) .</li>
		<li>Glue two 1 g magnets side-by-side onto the outer circumference of the wheel (Figure 2A). 
		NOTE: Here, it is helpful to use tape to hold the magnets in place until the glue firmly dries; otherwise, they may be attracted to the sensor magnet and become dislodged.</li>
		<li>Apply additional load in weeks 3, 4, and 6 by placing another 1 g magnet on top of either of the magnets already present. 
		NOTE: No glue is necessary as the magnets firmly adhere to one another. For example, with 6 g of load in week 6, the magnets will each be stacked three-high (Figure 2B).</li>
		<li>High loaded wheel running (HLWR): Follow steps 2.1.8-2.1.11. 
		NOTE: The HLWR protocol requires three sets of wheels. Assembling different sets of wheels allows the researcher to reuse wheel setups for other mice once the wheel is thoroughly cleaned and sanitized (numbers of each set should be determined by the researcher based on cohort/group size).</li>
		<li>The first set of wheels (required for week 2 only) will have a single 2.5 g magnet; glue (refer to the NOTE below Step 2.1.5) one 2.5 g magnet onto the outer circumference of the wheel (Figure 3A).</li>
		<li>The second set of wheels (required for week 3 only) will have two 2.5 g magnets; glue (refer to the NOTE below Step 2.1.5) two 2.5 g magnets side-by-side onto the outer circumference of the wheel (Figure 3B).</li>
		<li>The third set of wheels (required for week 4 and beyond) will have three 2.5 g magnets side-by-side; glue (refer to the NOTE below Step 2.1.5) three 2.5 g magnets side-by-side onto the outer circumference of the wheel (Figure 3C).</li>
		<li>Apply additional load for weeks 6 and 8 by placing another 2.5 g magnet on top of either of the magnets already present (Figures 3D, E).</li>
	</ol>
	</li>

3. Cage assembly
<ol>
	<li>Assemble running wheels using a cage equipped with a digital bike computer to monitor time exercising (h) and distance traveled (km). Average speed (km/h) is derived arithmetically.
	<ol>
		<li>Ensure that a fresh battery is inserted into the bike computer prior to assembly.</li>
		<li>Set wheel size during initial bike computer programming (see manufacturer&#39;s instructions); calculate per revolution distance by measuring the outer circumference of the running wheel (e.g., 3,580 mm for the wheel type used herein).</li>
	</ol>
	</li>
	<li>Place the bike computer sensor within a solid surface on the outside of the cage lid, directly above where the sensor magnet of the wheel is located. Ensure that all computer and sensor components are contained within a solid barrier outside the cage to prevent mice from chewing on components.
	<ol>
		<li>Utilize the lid of an empty pipette tip box with a small rectangle cut out for the magnetic bike sensor to reside, and the main part of the box (with the tip rack grid removed) to hold the bike computer and wire (Figure 4A).</li>
		<li>Drill two holes through the corners of the solid surface to secure the magnetic bike sensor and running wheel stand in place on the outside of the cage (Figure 4A).</li>
	</ol>
	</li>
	<li>Insert the running wheel base, upside down, through the gaps in the cage lid but on top of the solid surface described in step 3.2 (Figure 4B).
	<ol>
		<li>Secure the wheel base and computer sensor to the top of the cage with hardware (Figure 4C, D).</li>
	</ol>
	</li>
	<li>Ensure that the sensor magnet and computer sensor are spaced no more than 1 cm apart to allow for proper recording of wheel movement (most standard bike computer sensors are bi-directional and will record positive wheel movement in either direction of rotation).</li>
	<li>Attach the appropriate running wheel (as described above) to the wheel base from the inside of the cage lid, and securely place the lid onto the cage (Figure 4E, F).</li>
	<li>With the wheel hanging from the cage lid, ensure at least 2.5 cm of clearance from the cage floor. Place a minimal amount of bedding material in the cage to ensure that the wheel spins freely but does not become impeded by the buildup of bedding.</li>
	<li>During experimentation, record data from the bike computer at a consistent interval schedule to ensure accurate activity monitoring.
	<ol>
		<li>Recognize that mice are a nocturnal species; therefore, most of their natural cage activity (including wheel running) will be performed during the dark hours of the light cycle.</li>
	</ol>
	</li>

4. Exercise training loading protocols
<ol>
	<li>Individually house sedentary (SED) mice for 9 weeks in a cage containing a locked running wheel to prevent any running. 
	NOTE: Table 1 provides the loading schedule for the LWR (PoWeR) and HLWR protocols used in the experimental design.</li>
	<li>Reduce load for the LWR and HLWR groups, if necessary, to ensure that mice continue to exercise for the entire 9 week protocol.</li>
</ol>

5. In situ muscle function testing, tissue harvesting, and tissue analysis
<ol>
	<li>Following the 9 week training intervention, anesthetize mice using inhaled isoflurane (4% induction; 2% maintenance) with supplemental oxygen, and ensure proper anesthetic plane monitoring throughout the procedure.</li>
	<li>Perform an in situ muscle function test on the gastrocnemius, plantaris, soleus (GPS) complex to test isometric muscular strength28. Establish a force-frequency curve by directly stimulating the sciatic nerve with 27 G electrode needles at 11 ascending frequencies between 1-300 Hz, with tetanic contractions occurring around 100-150 hz29.</li>
	<li>Immediately following the muscle function test, euthanize the mice via cervical dislocation and confirm euthanasia by removing the heart. Carefully excise the plantaris and soleus muscles and record the wet tissue mass.</li>
	<li>Coat each muscle sample in an embedding medium (OCT) and mount it on a cork. Freeze it in liquid nitrogen-cooled isopentane, and store it at -80 &#176;C until further immunohistochemical (IHC) analysis is performed on sections of muscle tissue (10 &#181;m thick).</li>
	<li>Analyze muscle fiber CSA using immunofluorescence for laminin. Measure fiber CSA using an automatic image quantification platform30.</li>
</ol>
6. Statistical analysis
<ol>
	<li>Express all data as mean &#177; SD.</li>
	<li>Perform statistical analyses using statistical analysis software with significance set at p &#8804; 0.05.</li>
	<li>Compare wheel running and training volume data with repeated measures two-way ANOVA.</li>
	<li>Compare body mass, tissue mass, CSA, and muscle function with a one-way ANOVA. If significant F-ratios are found, compare within-group differences using Fisher LSD post hoc analyses.</li>
	<li>Calculate effect sizes, then interpret them as 0.01, 0.06, and 0.14 for small, medium, and large effect sizes, respectively.</li>

Existing resistance exercise models in rodents have proven invaluable for skeletal muscle research; however, many of these models are invasive, involuntary, and/or time- and labor-intensive. Skeletal muscle mass comprises approximately 40% of body mass in adult humans; thus, maintaining skeletal muscle mass throughout life is critical. Skeletal muscle mass plays an integral role in energy metabolism, maintaining core body temperature, and glucose homeostasis1. The maintenance of skeletal muscle is a balance between protein synthesis and protein degradation, but many gaps still exist in the understanding of the intricate molecular mechanisms that drive these processes. To study the molecular mechanisms that regulate the maintenance and growth of muscle mass, human subjects&#39; research models often employ resistance exercise-based interventions, since mechanical stimuli play an integral role in the regulation of skeletal muscle mass. While human subjects research has been successful, the time necessary to exhibit adaptations and ethical concerns regarding invasive procedures (i.e., muscle biopsies) limit the quantity of data that can be obtained. While the adaptations to resistance exercise are fairly ubiquitous across mammalian species, animal models provide the benefit of being able to precisely control the diet and exercise regimen while also allowing for the collection of whole tissues throughout the body, such as the brain, liver, heart, and skeletal muscle.
Many resistance training models have been developed for use in rodents: synergistic ablation2, electrical stimulation3,4, weighted ladder climbing5, weighted sled pulling6, and canvassed squatting7. It is evident that all of these models, if done correctly, can be effective models to induce skeletal muscle adaptations, such as hypertrophy. However, the downfalls of these models are that they are mostly involuntary, not part of normal rodent behavior, time-/labor-intensive, and invasive.
Fortunately, many mouse and rat strains voluntarily run long distances when given access to a running wheel. Moreover, free-running wheel (FWR) exercise models do not rely on extensive conditioning, positive/negative reinforcement, or anesthesia to force movement or muscle activity8,9. Running activity depends greatly on mouse strain, sex, age, and an individual basis. Lightfoot et al. compared the running activity of 15 different mouse strains and found that daily running distance ranges from 2.93 km to 7.93 km, with C57BL/6 mice running the farthest, regardless of sex10. FWR is commonly accepted as an excellent model for inducing endurance adaptations in skeletal and cardiac muscles11,12,13,14,15,16; however, utilizing wheel running in resistance training models is less commonly investigated.
As one could suspect, the hypertrophic effect of wheel running might be augmented by adding resistance to the running wheel, termed loaded wheel running (LWR), thus requiring greater efforts to run on the wheel to more closely mimic resistance training. Using varied methods of load application, previous studies have demonstrated that the LWR model utilizing rats and mice routinely displayed increases in limb muscle mass of 5%-30% in a matter of 6-8 weeks17,18,19,20,21. Furthermore, D&#39;hulst et al. demonstrated that a single bout of LWR led to a 50% greater increase&#160;in activation of the protein synthesis signaling pathway compared to FWR22. Wheel resistance has been most commonly applied by a friction-based, constant loading method, whereby a magnetic brake or tension bolt is utilized to apply wheel resistance12,19,23,24. One caveat of the friction-based, constant load method is that when moderate to high resistance is applied, the animal cannot overcome the high resistance to initiate movement of the wheel, effectively ceasing training. Most importantly, many of the cage and wheel systems used for rodent running wheel models are quite costly and require specialized equipment.
Recently, Dungan et al. developed a progressive weighted-wheel-running (PoWeR) model, which applies a load to the wheel asymmetrically via external masses adhered to a single side of the wheel. The unbalanced wheel loading and variable resistance of the PoWeR model are thought to encourage continued running activity and promote shorter bursts of loaded wheel running in mice, more closely imitating the sets and repetitions performed with resistance training17. Despite the average running distance being 10-12 km per day, the PoWeR model yielded a 16% and 17% increase in plantaris muscle wet mass and fiber cross-sectional area (CSA), respectively. Despite many practical advantages, the PoWeR model of LWR does have some limitations. As recognized by the authors, the PoWeR model is a high-volume &#34;hybrid&#34; stimulus that is reflective of a blended endurance/resistance exercise model (i.e., concurrent training in humans), as opposed to a more strictly resistance exercise-based model, potentially introducing an interference effect and contributing to the less pronounced hypertrophy or different mechanisms by which hypertrophy is induced25. Ensuring that a concurrent training phenomenon does not occur in what is intended to be a resistance exercise training model is imperative. Therefore, the PoWeR model was modified to develop a LWR model that utilizes higher loads than previously used to more closely resemble a resistance training model. Herein, details are provided for a simple and inexpensive 9 week progressive resistance training LWR model in C57BL/6 mice.

Previously developed rodent resistance-based exercise models, including synergistic ablation, electrical stimulation, weighted-ladder climbing, and most recently, weighted-sled pulling, are highly effective at providing a hypertrophic stimulus to induce skeletal muscle adaptations. While these models have proven invaluable for skeletal muscle research, they are either invasive or involuntary and labor-intensive. Fortunately, many rodent strains voluntarily run long distances when given access to a running wheel. Loaded wheel running (LWR) models in rodents are capable of inducing adaptations commonly observed with resistance training in humans, such as increased muscle mass and fiber hypertrophy, as well as stimulation of muscle protein synthesis. However, the addition of moderate wheel load either fails to deter mice from running great distances, which is more reflective of an endurance/resistance training model, or the mice discontinue running nearly entirely due to the method of load application. Therefore, a novel high-load wheel running model (HLWR) has been developed for mice where external resistance is applied and progressively increased, enabling mice to continue running with much higher loads than previously utilized. Preliminary results from this novel HLWR model suggest it provides sufficient stimulus to induce hypertrophic adaptations over the 9 week training protocol. Herein, the specific procedures to execute this simple yet inexpensive progressive resistance-based exercise training model in mice are described.

In this study, 24 C57BL/6 mice (6.3 &#177; 0.7 months at the start of this study) were randomly assigned to one of three treatment groups: sedentary (SED), loaded wheel running (LWR; same as PoWeR described by Dungan et al.17), or high LWR (HLWR), and then completed their respective 9 week protocol. Assessment of Murine Exercise Endurance Without the Use of a Shock Grid: An Alternative to Forced Exercise

Using laboratory mouse models, the molecular pathways responsible for the metabolic benefits of endurance exercise are beginning to be defined. The most common method for assessing exercise endurance in mice utilizes forced running on a motorized treadmill equipped with a shock grid. Animals who quit running are pushed by the moving treadmill belt onto a grid that delivers an electric foot shock; to escape the negative stimulus, the mice return to running on the belt. However, avoidance behavior and psychological stress due to use of a shock apparatus can interfere with quantitation of running endurance, as well as confound measurements of postexercise serum hormone and cytokine levels. Here, we demonstrate and validate a refined method to measure running endurance in na&#239;ve C57BL/6 laboratory mice on a motorized treadmill without utilizing a shock grid. When mice are preacclimated to the treadmill, they run voluntarily with gait speeds specific to each mouse. Use of the shock grid is replaced by gentle encouragement by a human operator using a tongue depressor, coupled with sensitivity to the voluntary willingness to run on the part of the mouse. Clear endpoints for quantifying running time-to-exhaustion for each mouse are defined and reflected in behavioral signs of exhaustion such as splayed posture and labored breathing. This method is a humane refinement which also decreases the confounding effects of stress on experimental parameters.

Using the Activity-based Anorexia Rodent Model to Study the Neurobiological Basis of Anorexia Nervosa

Anorexia nervosa (AN) is a psychiatric illness characterized by excessively restricted caloric intake and abnormally high levels of physical activity. A challenging illness to treat, due to the lack of understanding of the underlying neurobiology, AN has the highest mortality rate among psychiatric illnesses. To address this need, neuroscientists are using an animal model to study how neural circuits may contribute toward vulnerability to AN and may be affected by AN. Activity-based anorexia (ABA) is a bio-behavioral phenomenon described in rodents that models the key symptoms of anorexia nervosa. When rodents with free access to voluntary exercise on a running wheel experience food restriction, they become hyperactive &#8211; running more than animals with free access to food. Here, we describe the procedures by which ABA is induced in adolescent female C57BL/6 mice. On postnatal day 36 (P36), the animal is housed with access to voluntary exercise on a running wheel. After 4 days of acclimation to the running wheel, on P40, all food is removed from the cage. For the next 3 days, food is returned to the cage (allowing animals free food access) for 2 hr daily. After the fourth day of food restriction, free access to food is returned and the running wheel is removed from the cage to allow the animals to recover. Continuous multi-day analysis of running wheel activity shows that mice become hyperactive within 24 hr following the onset of food restriction. The mice run even during the limited time during which they have access to food. Additionally, the circadian pattern of wheel running becomes disrupted by the experience of food restriction. We have been able to correlate neurobiological changes with various aspects of the animals&#8217; wheel running behavior to implicate particular brain regions and neurochemical changes with resilience and vulnerability to food-restriction induced hyperactivity.

An Innovative Running Wheel-based Mechanism for Improved Rat Training Performance

This study presents an animal mobility system, equipped with a positioning running wheel (PRW), as a way to quantify the efficacy of an exercise activity for reducing the severity of the effects of the stroke&#160;in rats. This system provides more effective animal exercise training than commercially available systems such as treadmills and motorized running wheels (MRWs). In contrast to an MRW that can only achieve speeds below 20 m/min, rats are permitted to run at a stable speed of 30 m/min on a more spacious and high-density rubber running track supported by a 15 cm wide acrylic wheel with a diameter of 55 cm in this work. Using a predefined adaptive acceleration curve, the system not only reduces the operator error but also trains the rats to run persistently until a specified intensity is reached. As a way to evaluate the exercise effectiveness, real-time position of a rat is detected by four pairs of infrared sensors deployed on the running wheel. Once an adaptive acceleration curve is initiated using a microcontroller, the data obtained by the infrared sensors are automatically recorded and analyzed in a computer. For comparison purposes, 3 week training is conducted on rats using a treadmill, an MRW and a PRW. After surgically inducing middle cerebral artery occlusion (MCAo), modified neurological severity scores (mNSS) and an inclined plane test were conducted to assess the neurological damages to the rats. PRW is experimentally validated as the most effective among such animal mobility systems. Furthermore, an exercise effectiveness measure, based on rat position analysis, showed that there is a high negative correlation between the effective exercise and the infarct volume, and can be employed to quantify a rat training in any type of brain damage reduction&#160;experiments.

This study presents an innovative running wheel-based animal mobility system to quantify an effective exercise activity in rats. A rat-friendly testbed is built, using a predefined adaptive acceleration curve, and a high correlation between the effective exercise rate and the infarct volume suggests the protocol&#39;s potential for stroke prevention experiments.

A Simple and Low-cost Assay for Measuring Ambulation in Mouse Models of Muscular Dystrophy

Measuring functional outcomes in the treatment of muscular dystrophy is an essential aspect of preclinical testing. The assessment of voluntary ambulation in mouse models is a non-invasive and reproducible activity assay that is directly analogous to measures of patient ambulation such as the 6-minute walk test and related mobility scores. Many common methods for testing mouse ambulation speed and distance are based on the open field test, where an animal&#39;s free movement within an arena is measured over time. One major downside to this approach is that commercial software and equipment for high-resolution motion tracking is expensive and may require transferring mice to specialized facilities for testing. Here, we describe a low-cost, video-based system for measuring mouse ambulation that utilizes free and open-source software. Using this protocol, we demonstrate that voluntary ambulation in the dystrophin-null mdx mouse model for Duchenne muscular dystrophy (DMD) is decreased relative to wild-type mouse activity. In mdx mice expressing the utrophin transgene, these activity deficits are not observed and the total distance traveled is indistinguishable from wild-type mice. This method is effective for measuring changes in voluntary ambulation associated with dystrophic pathology, and provides a versatile platform that can be readily adapted to diverse research settings.

This protocol describes a flexible, low-cost system for measuring mouse ambulation in an open field activity assay. We show that a 6-minute ambulation assay based on this system detects a decrease in voluntary movement in mdx mice, and accurately distinguishes improvement in a muscle-specific rescue of these animals.

A Method to Study the Impact of Chemically-induced Ovarian Failure on Exercise Capacity and Cardiac Adaptation in Mice

The risk of cardiovascular disease (CVD) increases in post-menopausal women, yet, the role of exercise, as a preventative measure for CVD risk in post-menopausal women has not been adequately studied. Accordingly, we investigated the impact of voluntary cage-wheel exercise and forced treadmill exercise on cardiac adaptation in menopausal mice. The most commonly used inducible model for mimicking menopause in women is the ovariectomized (OVX) rodent. However, the OVX model has a few dissimilarities from menopause in humans. In this study, we administered 4-vinylcyclohexene diepoxide (VCD) to female mice, which accelerates ovarian failure as an alternative menopause model to study the impact of exercise in menopausal mice. VCD selectively accelerates the loss of primary and primordial follicles resulting in an endocrine state that closely mimics the natural progression from pre- to peri- to post-menopause in humans. To determine the impact of exercise on exercise capacity and cardiac adaptation in VCD-treated female mice, two methods were used. First, we exposed a group of VCD-treated and untreated mice to a voluntary cage wheel. Second, we used forced treadmill exercise to determine exercise capacity in a separate group VCD-treated and untreated mice measured as a tolerance to exercise intensity and endurance.

Tracking Individual Running Metrics in Mice Using a Voluntary Wheel Running Protocol that Minimizes Social Isolation

Voluntary wheel running (VWR) in rodents is used to evaluate performance and endurance and to simulate exercise. In contrast to other exercise paradigms that require aversive stimulus to induce active movement, wheel running is voluntary. VWR generally consists of placing a running wheel inside the mouse home cage. However, analyzing exercise parameters can typically only be conducted when mice are individually housed, while overall cage effects can be reported when mice are group-housed. Therefore, the current models force experimenters to choose between measuring individual exercise parameters in single-housed mice or measuring group effects in group-housed mice; depending on the research question and model, this could be limiting. 
Therefore, the goal of this method is to demonstrate a protocol that assesses VWR in a mouse model by quantifying running parameters in individual mice, with minimal isolation. We report on device design, preoperative preparations, and experiment setup. Results of a 6-week training protocol using this VWR model suggest it provides sufficient stimulus to increase spatial object recognition memory, comparable to the clinical effect of exercise. This simple and inexpensive model facilitates investigation into the effect of VWR on multiple measurements of wheel activity, minimizing factors that could limit or affect experimental outcomes.

Dosage-Adjusted Resistance Training in Mice with a Reduced Risk of Muscle Damage

Progressive resistance training (PRT), which involves performing muscle contractions against progressively greater external loads, can increase muscle mass and strength in healthy individuals and in patient populations. There is a need for precision rehabilitation tools to test the safety and effectiveness of PRT to maintain and/or restore muscle mass and strength in preclinical studies on small and large animal models. The PRT methodology and device described in this article can be used to perform dosage-adjusted resistance training (DART). The DART device can be used as a standalone dynamometer to objectively assess the concentric contractile torque generated by the ankle dorsiflexors in mice or can be added to a pre-existing isokinetic dynamometry system. The DART device can be fabricated with a standard 3D printer based on the instructions and open-source 3D print files provided in this work. The article also describes the workflow for a study to compare contraction-induced muscle damage caused by a single bout of DART to muscle damage caused by a comparable bout of isometric contractions (ISOM) in a mouse model of limb-girdle muscular dystrophy type 2B/R2 (BLAJ mice). The data from eight BLAJ mice (four animals for each condition) suggest that less than 10% of the tibialis anterior (TA) muscle was damaged from a single bout of DART or ISOM, with DART being less damaging than ISOM.

Behavior is shaped by actions, and actions necessitate motor skills such as strength, coordination, and learning. None of the behaviors essential for sustaining life would be possible without the ability to transition from one position to another. Unfortunately, motor skills can be compromised in a wide array of diseases. Therefore, investigating the mechanisms of motor functions at the cellular, molecular, and circuit levels, as well as understanding the symptoms, causes, and progression of motor disorders, is crucial for developing effective treatments. Mouse models are frequently employed for this purpose.
This article describes a protocol that allows the monitoring of various aspects of motor performance and learning in mice using an automated tool called the Erasmus Ladder. The assay involves two phases: an initial phase where mice are trained to navigate a horizontal ladder built of irregular rungs (&#34;fine motor learning&#34;), and a second phase where an obstacle is presented in the path of the moving animal. The perturbation can be unexpected (&#34;challenged motor learning&#34;) or preceded by an auditory tone (&#34;associative motor learning&#34;). The task is easy to conduct and is fully supported by automated software.
This report shows how different readouts from the test, when analyzed with sensitive statistical methods, allow fine monitoring of mouse motor skills using a small cohort of mice. We propose that the method will be highly sensitive to evaluate motor adaptations driven by environmental modifications as well as early-stage subtle motor deficits in mutant mice with compromised motor functions.

Activating Autophagy by Aerobic Exercise in Mice

Autophagy is a lysosomal degradation pathway essential for cell homeostasis, function and differentiation. Under stress conditions, autophagy is induced and targets various cargos, such as bulk cytosol, damaged organelles and misfolded proteins, for degradation in lysosomes. Resulting nutrient molecules are recycled back to the cytosol for new protein synthesis and ATP production. Upregulation of autophagy has beneficial effects against the pathogenesis of many diseases, and pharmacological and physiological strategies to activate autophagy have been reported. Aerobic exercise is recently identified as an efficient autophagy inducer in multiple organs in mice, including muscle, liver, heart and brain. Here we show procedures to induce autophagy in vivo by either forced treadmill exercise or voluntary wheel running. We also demonstrate microscopic and biochemical methods to quantitatively analyze autophagy levels in mouse tissues, using the marker proteins LC3 and p62 that are transported to and degraded in lysosomes along with autophagosomes.

Exploring T2DM Exercise Interventions: A Static Training Device for Mice

Static Strength Training Method for Type 2 Diabetic Mice

The treatment of type 2 diabetes mellitus (T2DM) is a major difficulty in improving patient health. Exercise is one of the main interventions for T2DM. Static strength training is one of the key forms of traditional sports in China. Research shows that static strength training is an effective clinical method for T2DM intervention, but there is no experimental device suitable for static training in mice. One of the difficulties in moving from clinical to basic research is to design appropriate experimental devices. In order to further study the mechanism of static training intervention in T2DM, a simple method for making a static training device for mice is introduced in this paper. This device has the advantages of simple operation, cheap material, and high feasibility. Previous studies conducted under this protocol have shown that static training can effectively reduce blood glucose levels and improve the mitochondrial function of skeletal muscle cells in T2DM mice. The purpose of introducing this device is to promote research on the mechanism of traditional exercise in the intervention of T2DM and to lay a foundation for the quantitative intervention of exercise.
