The mechanisms by which isoflurane injured the developing brain are not clear. Recent work has demonstrated that it is mediated in part by activation of p75 neurotrophin receptor. This receptor activates RhoA, a small guanosine triphosphatase that can depolymerize actin. It is therefore conceivable that inhibition of RhoA or prevention of cytoskeletal depolymerization might attenuate isoflurane neurotoxicity. This study was conducted to test these hypotheses using primary cultured neurons and hippocampal slice cultures from neonatal mouse pups.

Isoflurane neurotoxicity is mediated by p75NTR-RhoA activation and actin depolymerization.

The aged brain exhibits a loss in gray matter and a decrease in spines and synaptic densities that may represent a sequela for neurodegenerative diseases such as Alzheimer's. Membrane/lipid rafts (MLR), discrete regions of the plasmalemma enriched in cholesterol, glycosphingolipids, and sphingomyelin, are essential for the development and stabilization of synapses. Caveolin-1 (Cav-1), a cholesterol binding protein organizes synaptic signaling components within MLR. It is unknown whether loss of synapses is dependent on an age-related loss of Cav-1 expression and whether this has implications for neurodegenerative diseases such as Alzheimer's disease.

Loss of caveolin-1 accelerates neurodegeneration and aging.

BACKGROUND:: Propofol exposure to neurons during synaptogenesis results in apoptosis, leading to cognitive dysfunction in adulthood. Previous work from our laboratory showed that isoflurane neurotoxicity occurs through p75 neurotrophin receptor (p75) and subsequent cytoskeleton depolymerization. Given that isoflurane and propofol both suppress neuronal activity, we hypothesized that propofol also induces apoptosis in developing neurons through p75. METHODS:: Days in vitro 5-7 neurons were exposed to propofol (3 Î¼M) for 6 h and apoptosis was assessed by cleaved caspase-3 (Cl-Csp3) immunoblot and immunofluorescence microscopy. Primary neurons from p75 mice or wild-type neurons were treated with propofol, with or without pretreatment with TAT-Pep5 (10 Î¼M, 15 min), a specific p75 inhibitor. P75 neurons were transfected for 72 h with a lentiviral vector containing the synapsin-driven p75 gene (Syn-p75) or control vector (Syn-green fluorescent protein) before propofol. To confirm our in vitro findings, wild-type mice and p75 mice (PND5) were pretreated with either TAT-Pep5 or TAT-ctrl followed by propofol for 6 h. RESULTS:: Neurons exposed to propofol showed a significant increase in Cl-Csp3, an effect attenuated by TAT-Pep5 and hydroxyfasudil. Apoptosis was significantly attenuated in p75 neurons. In p75 neurons transfected with Syn-p75, propofol significantly increased Cl-Csp3 in comparison with Syn-green fluorescent protein-transfected p75 neurons. Wild-type mice exposed to propofol exhibited increased Cl-Csp3 in the hippocampus, an effect attenuated by TAT-Pep5. By contrast, propofol did not induce apoptosis in p75 mice. CONCLUSION:: These results demonstrate that propofol induces apoptosis in developing neurons in vivo and in vitro and implicate a role for p75 and the downstream effector RhoA kinase.

Propofol Neurotoxicity Is Mediated by p75 Neurotrophin Receptor Activation.

P75 neurones ont &#xE9;t&#xE9; transfect&#xE9;es pendant 72 h avec un vecteur lentiviral contenant le synapsine ax&#xE9;e p75 g&#xE8;ne (Syn-p75) ou la lutte antivectorielle (Syn-green fluorescent protein), avant le propofol. Pour confirmer notre r&#xE9;sultats obtenus in vitro, des souris de type sauvage et les souris (p75 PND5) ont &#xE9;t&#xE9; pr&#xE9;trait&#xE9;es avec soit TAT-Pep5 ou TAT-ctrl suivie par le propofol pendant 6 h. R&#xE9;sultats :: Les neurones expos&#xE9;s au propofol a montr&#xE9; une augmentation significative de la Cl-Csp3, un effet att&#xE9;nu&#xE9; par TAT-Pep5 et hydroxyfasudil. L&#x26;#39;apoptose a &#xE9;t&#xE9; significativement att&#xE9;nu&#xE9;e en p75 neurones. En p75 neurones transfect&#xE9;es avec Syn-p75, le propofol augment&#xE9; de fa&#xE7;on significative Cl-Csp3 en comparaison avec Syn-prot&#xE9;ine fluorescente verte-transfect&#xE9;es p75 neurones. Souris de type sauvage expos&#xE9;es au propofol expos&#xE9;es augment&#xE9; Cl-Csp3 dans l&#x26;#39;hippocampe, un effet att&#xE9;nu&#xE9; par TAT-Pep5. En revanche, le propofol n&#x26;#39;a pas induire l&#x26;#39;apoptose dans p75 souris. CONCLUSION :: Ces r&#xE9;sultats d&#xE9;montrent que le propofol induit l&#x26;#39;apoptose dans les neurones en d&#xE9;veloppement in vivo et in vitro et d&#x26;#39;impliquer un r&#xF4;le &#xE0; jouer p75 et la kinase en aval effecteur de RhoA.

Fr&uuml;here Arbeiten von unserem Labor zeigte, dass Isofluran Neurotoxizit&auml;t bis p75 Neurotrophin Rezeptor (p75) und anschlie&szlig;ende Zytoskeletts Depolymerisation stattfindet. Angesichts der Tatsache, dass sowohl Isofluran als auch Propofol neuronalen Aktivit&auml;t unterdr&uuml;cken, wir die Hypothese, die Propofol auch induziert Apoptose in der Entwicklung von Neuronen durch p75. P75 Neuronen wurden f&uuml;r 72 h mit einem lentiviralen Vektor mit der Synapsiden-gesteuerte p75-gen (Syn-p75) oder dem Steuerelement Vektor (Syn-Green fluorescent Protein) vor Propofol transfected. Um unsere in-vitro-Ergebnisse zu best&auml;tigen, wurden Wildtyp M&auml;usen und p75-M&auml;use (PND5) vorbehandelt mit TAT-Pep5 oder TAT-Ctrl gefolgt von Propofol f&uuml;r 6 h. Ergebnisse:: Neuronen ausgesetzt Propofol zeigte eine deutliche Steigerung in Cl-Csp3, einen Effekt abgeschw&auml;cht durch TAT-Pep5 und Hydroxyfasudil. Apoptose wurde in p75 Neuronen deutlich abgeschw&auml;cht. In p75 Neuronen, die mit Syn-p75 transfected erh&ouml;ht Propofol deutlich Cl-Csp3 im Vergleich zu Syn-gr&uuml;n fluoreszierendes Protein-transfected p75 Neuronen. Wildtyp M&auml;usen Propofol ausgestellt ausgesetzt Anstieg der Hippocampus, eine Wirkung abgeschw&auml;cht durch TAT-Pep5 Cl-Csp3. Antecedentes :: La exposici&#xF3;n propofol a las neuronas durante la sinaptog&#xE9;nesis resultados en la apoptosis, que conducen a la disfunci&#xF3;n cognitiva en la edad adulta. Trabajos anteriores de nuestro laboratorio mostraron que la neurotoxicidad del isoflurano produce a trav&#xE9;s del receptor de neurotrofinas p75 (p75) y despolimerizaci&#xF3;n del citoesqueleto posterior. Teniendo en cuenta que el isoflurano y el propofol, ambos suprime la actividad neuronal, la hip&#xF3;tesis de que el propofol tambi&#xE9;n induce la apoptosis en las neuronas en desarrollo a trav&#xE9;s de p75. Mouse models of DS, especially the Ts65Dn model, have been pivotal in furthering our understanding of the genetic, molecular and neurobiological mechanisms that underlie learning and memory impairments in DS. Cognitive and pharmacological insights from the Ts65Dn mouse model have led to remarkable translational progress in the development of therapeutic targets and in the emergence of DS clinical trials. Unravelling the pathogenic role of trisomic genes on human chromosome 21 and the genotype-phenotype relationship still remains a pertinent goal for tackling cognitive deficits in DS.

The cognitive impairment in people with DS is virtually universal but varies considerably with respect to expressivity and severity. Significant advances in medical treatment and social inclusion have increased longevity in people with DS resulting in an increased aging population, thus highlighting the significance of early onset of dementia and the importance of identifying pharmacotherapies to treat DS-associated health complications in adults. Given its prevalence and established mouse models, this review will focus on ID in the DS population; specifically, the superimposed effect of aging on the complications already manifest in DS adults and the cognitive insights gained from studies on mouse models of DS.
