Some species of ciliates undergo massive DNA elimination and genome rearrangement to construct gene-sized "chromosomes" in their somatic nucleus. An example is the extensively scrambled DNA polymerase alpha gene that is broken into 48 pieces and distributed over two unlinked loci in Stylonychia. To understand the emergence of this complex phenomenon during evolution, we examined DNA polymerase alpha genes in several earlier diverging species, representing evolutionary intermediates. Mapping these data onto an evolutionary tree suggests that this gene became extensively fragmented and scrambled over evolutionary time through a series of steps, each leading to greater complexity. Our results also suggest a possible mechanism for intron loss by deletion of intron sequences as DNA during development of the somatic nucleus.

More effective therapies are urgently needed against hepatitis C virus (HCV), a major cause of viral hepatitis. We used in vitro protein expression and microfluidic affinity analysis to study RNA binding by the HCV transmembrane protein NS4B, which plays an essential role in HCV RNA replication. We show that HCV NS4B binds RNA and that this binding is specific for the 3' terminus of the negative strand of the viral genome with a dissociation constant (Kd) of approximately 3.4 nM. A high-throughput microfluidic screen of a compound library identified 18 compounds that substantially inhibited binding of RNA by NS4B. One of these compounds, clemizole hydrochloride, was found to inhibit HCV RNA replication in cell culture that was mediated by its suppression of NS4B's RNA binding, with little toxicity for the host cell. These results yield new insight into the HCV life cycle and provide a candidate compound for pharmaceutical development.

Terapias m&aacute;s efectivas se necesitan urgentemente contra el virus de la hepatitis C (VHC), una de las principales causas de la hepatitis viral. Utilizamos microflu&iacute;dicos afinidad an&aacute;lisis y expresi&oacute;n de la prote&iacute;na in vitro para estudiar la Uni&oacute;n de la RNA por la prote&iacute;na transmembrana de HCV NS4B, que desempe&ntilde;a un papel esencial en la replicaci&oacute;n del ARN de VHC. Mostramos que el VHC NS4B une RNA y que este enlace es espec&iacute;fico para la terminal 3&#39; de la hebra negativa del genoma viral con una constante de disociaci&oacute;n (Kd) de aproximadamente 3,4 nM. Hepatitis C Virus (HCV) is a leading cause of liver disease and represents a significant public health challenge. Treatments for this disease are inadequate and improved antiviral therapies are necessary. Several such antivirals are in development, most of which target the well-characterized NS3 protease or the NS5B polymerase. In contrast, the nonstructural 4B (NS4B) protein, though essential for HCV RNA replication, has been the subject of few pharmacological studies. One of the functions ascribed to this protein is the ability to form intracellular membrane-associated foci (MAF), which are believed to be related to the sites of viral replication. Here, we report the identification of a small molecule that inhibits HCV replication and disrupts the organization of these MAF. Genetic analysis links the compound's mode of action to the NS4B gene product, and transient transfections of NS4B-GFP demonstrate that treatment with this compound can lead to the formation of novel elongated assemblies of NS4B. Furthermore, an in vitro dynamic light scattering assay provides evidence that the second amphipathic helix of NS4B may be the target of the drug. Our results demonstrate that this molecule represents a new potential class of HCV inhibitors and also provides us with a useful tool for studying the HCV life cycle.

A small molecule inhibits HCV replication and alters NS4B's subcellular distribution.

Virus de l&#39;h&eacute;patite C (VHC) est des principales causes de maladie h&eacute;patique et repr&eacute;sente un d&eacute;fi important pour la sant&eacute; publique. Traitements pour cette maladie sont insuffisantes et l&#39;am&eacute;lioration des traitements antiviraux sont n&eacute;cessaires. Plusieurs de ces antiviraux est en d&eacute;veloppement, dont la plupart visent la prot&eacute;ase NS3 bien caract&eacute;ris&eacute;e ou la polym&eacute;rase NS5B. En revanche, les 4 b non structurales (NS4B) prot&eacute;ine, bien qu&#39;essentielle pour la r&eacute;plication de l&#39;ARN du VHC, a fait l&#39;objet de peu d&#39;&eacute;tudes pharmacologiques. Parmi les fonctions attribu&eacute;es &agrave; cette prot&eacute;ine est la capacit&eacute; de forme intracellulaire associ&eacute;e &agrave; la membrane foyers (MAF), qui sont soup&ccedil;onn&eacute;s d&#39;&ecirc;tre li&eacute;s aux sites de r&eacute;plication virale. Nous rapportons ici l&#39;identification d&#39;une petite mol&eacute;cule qui inhibe la r&eacute;plication du VHC et perturbe l&#39;organisation de ces CRG. Analyse g&eacute;n&eacute;tique des liens mode du compos&eacute; d&#39;action vers le produit du g&egrave;ne NS4B et transitoires transfections de NS4B-GFP d&eacute;montrent que le traitement avec ce compos&eacute; peut conduire &agrave; la formation de nouveaux assemblys allong&eacute;es de NS4B. En outre, un test in vitro diffusion dynamique de la lumi&egrave;re prouve que la deuxi&egrave;me h&eacute;lice amphipathique de NS4B peut &ecirc;tre la cible de la drogue. Nos r&eacute;sultats d&eacute;montrent que cette mol&eacute;cule repr&eacute;sente une nouvelle classe de potentielle d&#39;inhibiteurs du VHC et nous fournit aussi un outil utile pour &eacute;tudier le cycle de vie du VHC.

Trattamenti per questa malattia sono inadeguate e migliorate le terapie antivirali sono necessarie. Parecchi tali farmaci antivirali sono in via di sviluppo, la maggior parte dei quali target ben caratterizzata NS3 proteasi o la polimerasi NS5B. In contrasto, il 4B non strutturali della proteina (NS4B), anche se indispensabile per la replicazione di HCV-RNA, &egrave; stata oggetto di pochi studi farmacologici. Una delle funzioni attribuite a questa proteina &egrave; la capacit&agrave; di forma intracellulare membrana associato foci (MAF), che sono ritenute legate ai siti di replicazione virale. Riportiamo l&#39;identificazione di una piccola molecola che inibisce la replicazione di HCV e sconvolge l&#39;organizzazione di questi MAF. Exhaustion of CD8(+) T cells and upregulation of programmed death 1 (PD-1), a negative regulator of T cell activation, are characteristic features of individuals chronically infected with human immunodeficiency virus type 1. In a previous study, we showed in mice that a dendritic cell-directed lentiviral vector (DCLV) system encoding the human immunodeficiency virus (HIV)-1 Gag protein was an efficient vaccine modality to induce a durable Gag-specific T cell immune response. In this study, we demonstrate that blocking of the PD-1/PD-1 ligand (PD-L) inhibitory signal via an anti-PD-L1 antibody generated an enhanced HIV-1 Gag-specific CD8(+) immune response following both a single round of DCLV immunization and a homologous prime/boost regimen. The prime/boost regimen combined with PD-L1 blockade generated very high levels of Gag-specific CD8(+) T cells comprising several valuable features: improved ability to produce multiple cytokines, responding to a broader range of Gag-derived epitopes, and long-lasting memory. This enhanced cellular immune response generated by DCLV immunization combined with anti-PD-L1 blockade correlated with improved viral control following challenge with Gag-expressing vaccinia virus. Taken together, our studies offer evidence to support the use of PD-1/PD-L1 blockade as an adjuvant modality to enhance antigen-specific immune responses elicited by T cell-based immunizations such as DCLV.
