Important: There has been an erratum issued for this article. Read more …Konjuge aşılar aşılarda vaksinolojide dikkate değer gelişmeler vardır. Polisakkarit konjuge aşıların hazırlanması için, polisakkaritler rahatça işlevsel hale getirilebilir ve kullanımı kolay bir siyantüre reaktif olan 1-siyano-4-dimetilamopyridine tetrafluoroborate (CDAP) kullanılarak aşı taşıyıcı proteinlere bağlanabilir. CDAP, pH 7-9'da karbonhidrat hidroksil grupları ile reaksiyona geçerek polisakkaritleri aktive eder. CDAP'nin stabilitesi ve reaktivitesi yüksek oranda pH'a bağlıdır. Reaksiyonun pH'ı, CDAP'nin hidrolizi nedeniyle aktivasyon sırasında da azalır, bu da iyi pH kontrolünü tekrarlanabilir aktivasyonun anahtarı haline getirir. Orijinal CDAP aktivasyon protokolü, oda sıcaklığında unbuffered pH 9 çözeltilerinde gerçekleştirildi.
Bu durumdaki hızlı reaksiyon (<3 dk) ve hızlı CDAP hidrolizinden gelen hızlı pH düşüşü nedeniyle, hedef reaksiyon pH'ını kısa zaman diliminde hızlı bir şekilde ayarlamak ve korumak zordu. Geliştirilmiş protokol, etkinleştirme işlemini daha az çılgın, daha tekrarlanabilir ve ölçeklendirmeye daha uygun hale getirir.
