ここでは、単一細胞における絶対ミトコンドリア(mt)DNAコピー数とmtDNA欠失ヘテロプラスミーレベルを測定するためのプロトコルを紹介します。
哺乳類のミトコンドリア(mt)DNAは、電子伝達鎖の13サブユニットをコードする、小さな環状の二本鎖ミトコンドリア内DNA分子です。二倍体核ゲノムとは異なり、ほとんどの細胞にはmtDNAのコピーが多く含まれており、細胞の種類に応じて100コピー未満から200,000コピーを超える範囲です。MtDNAコピー数は、多くの加齢に伴う変性状態や疾患のバイオマーカーとしてますます使用されているため、mtDNAコピー数の正確な測定は、研究と診断の両方の設定において重要なツールになりつつあります。mtDNAの変異は、一塩基多型(SNP)または欠失として発生することが多く、細胞内のmtDNAのすべてのコピーに存在するか(ホモプラスミーと呼ばれる)、変異したmtDNAコピーとWTmtDNAコピーの混合物として存在する可能性があります(ヘテロプラスミーと呼ばれます)。ヘテロプラズマmtDNA変異は、希少疾患またはパーキンソン病などの一般的な遅発性疾患の増加のいずれかにおいて、臨床ミトコンドリア病理の主な原因です。細胞内に存在するヘテロプラスミーのレベルを決定することは、まれなミトコンドリア病の診断およびミトコンドリアが役割を果たす可能性のある一般的な遅発性疾患を理解することを目的とした研究において重要なステップです。MtDNAコピー数とヘテロプラスミーは、従来、定量的(q)PCRベースのアッセイまたはディープシーケンシングによって測定されてきました。しかし、ddPCR技術の最近の導入は、両方のパラメータを測定するための代替方法を提供しました。絶対mtDNAコピー数を測定する能力や、低いコピー数でも単一細胞から正確な測定を行うのに十分な感度など、既存の方法に比べていくつかの利点があります。ここでは、ddPCRを使用した単一細胞のmtDNAコピー数の測定を説明する詳細なプロトコル(以下、液滴生成PCRと呼びます)と、mtDNA欠失のある細胞のヘテロプラスミーを同時に測定するオプションを紹介します。mtDNA SNPを有する細胞のヘテロプラスミーを測定するためにこの方法を拡張する可能性についても議論されています。
哺乳類のミトコンドリア(mt)DNAは、ミトコンドリアマトリックスに存在する小さな(約16.5 Kb)環状DNAゲノムであり、2つのrRNA、22のtRNA、および13のタンパク質コード遺伝子で構成される37の遺伝子をコードしています1。細胞ごとに各遺伝子のコピーが1つ(一倍体)または2つ(二倍体)含まれる核ゲノムとは異なり、mtDNAは各細胞のミトコンドリアに複数のコピーとして存在し、数十コピー(成熟精母細胞など)から数十万コピー(卵母細胞など)の範囲で存在します2,3。このマルチコピーの性質の結果、一塩基多型(SNP)、欠失、または重複として存在する可能性のあるmtDNAゲノムの変異が、任意の細胞にさまざまなレベルで存在し、細胞の総mtDNA集団の0%から100%を占める可能性があります。同じ細胞内に野生型および変異型mtDNAゲノムが存在することはヘテロプラスミーと呼ばれ、病原性ヘテロプラズマmtDNA変異はミトコンドリア病の主な原因であり、いくつかの一般的な神経学的症候群が根底にあるヘテロプラスミックmtDNA変異に関連しています4。
