The error rate of RNA-dependent RNA polymerases (RdRp) affects the mutation frequency in a population of viral RNAs. Using chikungunya virus (CHIKV), we describe a unique arbovirus fidelity variant with a single C483Y amino acid change in the nsP4 RdRp that increases replication fidelity and generates populations with reduced genetic diversity. In mosquitoes, high fidelity CHIKV presents lower infection and dissemination titers than wild type. In newborn mice, high fidelity CHIKV produces truncated viremias and lower organ titers. These results indicate that increased replication fidelity and reduced genetic diversity negatively impact arbovirus fitness in invertebrate and vertebrate hosts.

Le taux d&#x26;#39;erreur des ARN polym&#xE9;rases ARN-d&#xE9;pendantes (RdRp) affecte la fr&#xE9;quence de mutation dans une population d&#x26;#39;ARN viraux. En utilisant le virus chikungunya (CHIKV), nous d&#xE9;crivons un arbovirus uniques variante fid&#xE9;lit&#xE9; avec un seul changement d&#x26;#39;acide amin&#xE9; dans la C483Y RdRp nsP4 qui augmente la fid&#xE9;lit&#xE9; de r&#xE9;plication et g&#xE9;n&#xE8;re des populations &#xE0; la diversit&#xE9; g&#xE9;n&#xE9;tique r&#xE9;duite. Il tasso di errore delle polimerasi RNA-dipendente RNA (RdRp) colpisce la frequenza di mutazione in una popolazione di RNAs virale. Utilizzando virus chikungunya (CHIKV), descriviamo una variante di fedelt&agrave; da arbovirus unico con una singola modifica dell&#39;amminoacido C483Y nella RdRp nsP4 che aumenta la fedelt&agrave; di replicazione e genera delle popolazioni con ridotta diversit&agrave; genetica.
