Dorsal root (DR) axons regenerate in the PNS but turn around or stop at the dorsal root entry zone (DREZ), the entrance into the CNS. Earlier studies that relied on conventional tracing techniques or postmortem analyses attributed the regeneration failure to growth inhibitors and lack of intrinsic growth potential. Here, we report the first in vivo imaging study of DR regeneration. Fluorescently labeled, large-diameter DR axons in thy1-YFPH mice elongated through a DR crush site, but not a transection site, and grew along the root at >1.5 mm/d with little variability. Surprisingly, they rarely turned around at the DREZ upon encountering astrocytes, but penetrated deeper into the CNS territory, where they rapidly stalled and then remained completely immobile or stable, even after conditioning lesions that enhanced growth along the root. Stalled axon tips and adjacent shafts were intensely immunolabeled with synapse markers. Ultrastructural analysis targeted to the DREZ enriched with recently arrived axons additionally revealed abundant axonal profiles exhibiting presynaptic features such as synaptic vesicles aggregated at active zones, but not postsynaptic features. These data suggest that axons are neither repelled nor continuously inhibited at the DREZ by growth-inhibitory molecules but are rapidly stabilized as they invade the CNS territory of the DREZ, forming presynaptic terminal endings on non-neuronal cells. Our work introduces a new experimental paradigm to the investigation of DR regeneration and may help to induce significant regeneration after spinal root injuries.

In vivo imaging of dorsal root regeneration: rapid immobilization and presynaptic differentiation at the CNS/PNS border.

Diese Daten legen nahe, dass Axone sind weder abgesto&szlig;en noch kontinuierlich an die DREZ durch Wachstum-hemmenden Molek&uuml;le gehemmt, sondern werden rasch stabilisiert, wie sie CNS Gebiet DREZ dringen, pr&auml;synaptische terminal Endungen auf nicht-neuronale Zellen bilden. La ra&#xED;z dorsal (RD) regenerar los axones en el SNP, sino dar la vuelta o detenerse en la zona de la ra&#xED;z dorsal de entrada (DREZ), la entrada en el SNC. Estudios anteriores que se basaban en las t&#xE9;cnicas convencionales de rastreo o an&#xE1;lisis post-mortem atribuy&#xF3; el fracaso de la regeneraci&#xF3;n a los inhibidores de crecimiento y la falta de potencial de crecimiento intr&#xED;nseco. Aqu&#xED;, se presenta la primera en el estudio de im&#xE1;genes in vivo de la regeneraci&#xF3;n DR. Marcado con fluorescencia, de gran di&#xE1;metro en los axones DR-thy1 YFPH ratones alargada a trav&#xE9;s de un sitio de aplastamiento DR, pero no un sitio de corte transversal, y creci&#xF3; a lo largo de la ra&#xED;z a&#x26;gt; 1,5 mm / d&#xED;a, con poca variabilidad. Sorprendentemente, pocas veces se dio la vuelta en el DREZ al encontrarse con los astrocitos, pero penetrado m&#xE1;s profundamente en el territorio del SNC, donde r&#xE1;pidamente se estanc&#xF3; y luego se mantuvo completamente inm&#xF3;vil o se mantiene estable, incluso despu&#xE9;s de las lesiones de aire acondicionado que un mayor crecimiento a lo largo de la ra&#xED;z. Estancados consejos ax&#xF3;n y ejes adyacentes fueron immunolabeled intensamente con los marcadores de sinapsis. El an&#xE1;lisis ultraestructural dirigido al enriquecimiento DREZ con axones reci&#xE9;n llegados, adem&#xE1;s, revel&#xF3; abundantes perfiles axonal presin&#xE1;ptica exhiban caracter&#xED;sticas tales como las ves&#xED;culas sin&#xE1;pticas que se ordenan a las zonas activas, pero las funciones no postsin&#xE1;pticos. Estos datos sugieren que los axones no son ni rechazado, ni inhibe de forma continua en el DREZ por mol&#xE9;culas inhibidoras del crecimiento, pero son r&#xE1;pidamente estabilizado, ya que invaden el territorio del SNC de la DREZ, formando terminaciones presin&#xE1;pticas de terminales en las c&#xE9;lulas no neuronales. Nuestro trabajo presenta un nuevo paradigma experimental para la investigaci&#xF3;n de la regeneraci&#xF3;n y la Rep&#xFA;blica Dominicana puede ayudar a inducir la regeneraci&#xF3;n significativa despu&#xE9;s de las lesiones de ra&#xED;ces espinales.
