Reprogramming somatic cells into an embryonic stem (ES) cell-like state, or induced pluripotent stem (iPS) cells, has emerged as a promising new venue for customized cell therapies. In this study, we performed directed differentiation to assess the ability of murine iPS cells to differentiate into bone, cartilage and fat in vitro and to maintain an osteoblast phenotype on a scaffold in vitro and in vivo. Embryoid bodies derived from murine iPS cells were cultured in differentiation medium for eight to twelve weeks. Differentiation was assessed by lineage specific morphology, gene expression, histological stain and immunostaining to detect matrix deposition. After 12 weeks of expansion, iPS derived osteoblasts were seeded in a gelfoam matrix followed by subcutaneous implantation in syngenic ICR mice. Implants were harvested at 12 weeks, and histological analyses of cell, mineral and matrix content were performed. Differentiation of iPS cells into mesenchymal lineages of bone, cartilage and fat was confirmed by morphology, and expression of lineage specific genes. Isolated implants of iPS cell derived osteoblasts expressed matrices characteristic of bone, including osteocalcin and bone sialoprotein. Implants were also stained with alizarin red and von Kossa, demonstrating mineralization and persistence of an osteoblast phenotype. Recruitment of vasculature and microvascularization of the implant was also detected. Taken together, these data demonstrate functional osteoblast differentiation from iPS cells both in vitro and in vivo and reveal a source of cells which merit evaluation for their potential uses in orthopaedic medicine and understanding of molecular mechanisms of orthopaedic disease.

Rekrutierung von Gef&auml;&szlig;system und Microvascularization des Implantats wurde auch erkannt. In questo studio, abbiamo eseguito la differenziazione diretto per valutare la capacit&agrave; delle cellule iPS murino a differenziarsi in osso, cartilagine e grasso in vitro e di mantenere un fenotipo degli osteoblasti su un patibolo in vitro e in vivo. Corpi embryoid derivati da cellule iPS murino sono stati coltivati nel mezzo di differenziazione per otto-dodici settimane. La reprogramaci&#xF3;n de c&#xE9;lulas som&#xE1;ticas en una madre embrionarias (ES) similar a la c&#xE9;lula (IPS) del estado, o las c&#xE9;lulas madre pluripotentes inducidas, se ha convertido en un lugar nuevo y prometedor para las terapias celulares personalizadas. En este estudio, se realiz&#xF3; la diferenciaci&#xF3;n dirigida a evaluar la capacidad de las c&#xE9;lulas iPS murinas de diferenciarse en hueso, cart&#xED;lago y grasa in vitro y para mantener un fenotipo de osteoblastos en un andamio in vitro e in vivo. Cuerpos embrionarios derivados de las c&#xE9;lulas iPS murinas se cultivaron en medio de diferenciaci&#xF3;n de ocho a doce semanas. Diferenciaci&#xF3;n se evalu&#xF3; por la morfolog&#xED;a linaje espec&#xED;fico, la expresi&#xF3;n g&#xE9;nica, la deposici&#xF3;n histol&#xF3;gico matriz mancha y inmunotinci&#xF3;n de detectar. Reprogramming somatic cells into an ESC-like state, or induced pluripotent stem (iPS) cells, has emerged as a promising new venue for customized cell therapies. In this study, we performed directed differentiation to assess the ability of murine iPS cells to differentiate into bone, cartilage, and fat in vitro and to maintain an osteoblast phenotype on a scaffold in vitro and in vivo. Embryoid bodies derived from murine iPS cells were cultured in differentiation medium for 8â€“12 weeks. Differentiation was assessed by lineage-specific morphology, gene expression, histological stain, and immunostaining to detect matrix deposition. After 12 weeks of expansion, iPS-derived osteoblasts were seeded in a gelfoam matrix followed by subcutaneous implantation in syngenic imprinting control region (ICR) mice. Implants were harvested at 12 weeks, histological analyses of cell and mineral and matrix content were performed. Differentiation of iPS cells into mesenchymal lineages of bone, cartilage, and fat was confirmed by morphology and expression of lineage-specific genes. Isolated implants of iPS cell-derived osteoblasts expressed matrices characteristic of bone, including osteocalcin and bone sialoprotein. Implants were also stained with alizarin red and von Kossa, demonstrating mineralization and persistence of an osteoblast phenotype. Recruitment of vasculature and microvascularization of the implant was also detected. Taken together, these data demonstrate functional osteoblast differentiation from iPS cells both in vitro and in vivo and reveal a source of cells, which merit evaluation for their potential uses in orthopedic medicine and understanding of molecular mechanisms of orthopedic disease.

La reprogramaci&#xF3;n de c&#xE9;lulas som&#xE1;ticas en un estado similar al CES, o c&#xE9;lulas madre pluripotentes inducidas (iPS), se ha convertido en un lugar nuevo y prometedor para las terapias celulares personalizadas. En este estudio, se realiz&#xF3; la diferenciaci&#xF3;n dirigida a evaluar la capacidad de las c&#xE9;lulas iPS murinas de diferenciarse en hueso, cart&#xED;lago y grasa in vitro y para mantener un fenotipo de osteoblastos en un andamio in vitro e in vivo. Cuerpos embrionarios derivados de las c&#xE9;lulas iPS murinas se cultivaron en un medio de diferenciaci&#xF3;n para las 8-12 semanas. La diferenciaci&#xF3;n fue evaluada por el linaje espec&#xED;fico de la morfolog&#xED;a, la expresi&#xF3;n de genes, manchas histol&#xF3;gico, y la inmunotinci&#xF3;n para detectar dep&#xF3;sitos de matriz. Despu&#xE9;s de 12 semanas de expansi&#xF3;n, iPS derivadas de los osteoblastos fueron sembradas en una matriz de espuma de gel seguida de la implantaci&#xF3;n subcut&#xE1;nea en la regi&#xF3;n de control de impresi&#xF3;n sing&#xE9;nicas (ICR) en ratones. Implantes aislados de c&#xE9;lulas iPS derivadas de los osteoblastos expresan las matrices caracter&#xED;sticas de los huesos, incluyendo la osteocalcina y la sialoprote&#xED;na hueso. Los implantes tambi&#xE9;n fueron te&#xF1;idas con rojo de alizarina y Von Kossa, lo que demuestra la mineralizaci&#xF3;n y la persistencia de un fenotipo de los osteoblastos.
