Concanavalin A on polymyxin B (PmB) retained higher binding capacity for yeast mannan, compared with covalently immobilized lectin. Kinetics of mannan-concanavalin A interaction were significantly different in dependence on type of concanavalin A immobilization.

To gain new insights into the mechanism underlying cytoplasmic male sterility (CMS), we compared the nuclear gene expression profiles of flowers of a Brassica napus CMS line with that of the fertile B. napus maintainer line using Arabidopsis thaliana flower-specific cDNA microarrays. The CMS line used has a B. napus nuclear genome, but has a rearranged mitochondrial (mt) genome consisting of both B. napus and A. thaliana DNA. Gene expression profiling revealed that a large number of genes differed in expression between the two lines. For example, nuclear genes coding for proteins that are involved in protein import into organelles, genes expressed in stamens and pollen, as well as genes implicated in either cell-wall remodeling or architecture, were repressed in the CMS line compared with B. napus. These results show that the mt genome of the CMS line strongly influences nuclear gene expression, and thus reveal the importance of retrograde signalling between the mitochondria and the nucleus. Furthermore, flowers of the CMS line are characterized by a replacement of stamens with carpelloid organs, and thus partially resemble the APETALA3 (AP3) and PISTILLATA (PI) mutants. In accordance with this phenotype, AP3 expression was downregulated in the stamens, shortly before these organs developed carpelloid characteristics, even though it was initiated correctly. Repression of PI succeeded that of AP3 and might be a consequence of a loss of AP3 activity. These results suggest that AP3 expression in stamens depends on proper mt function and a correct nuclear-mt interaction, and that mt alterations cause the male sterility phenotype of the CMS line.

Pour acqu&eacute;rir de nouvelles connaissances sur le m&eacute;canisme qui sous-tend la st&eacute;rilit&eacute; m&acirc;le cytoplasmique (CMS), nous avons compar&eacute; les profils d&#39;expression de g&egrave;ne nucl&eacute;aire de fleurs d&#39;une ligne CMS de Brassica napus avec celui de la ligne de mainteneur fertile de b. napus en utilisant Arabidopsis thaliana fleur sp&eacute;cifique cDNA microarrays. La ligne CMS utilis&eacute;e poss&egrave;de un g&eacute;nome nucl&eacute;aire de b. napus, mais poss&egrave;de un g&eacute;nome mitochondrial r&eacute;arrang&eacute;s (mt) compos&eacute; de b. napus et a. thaliana ADN. Profil d&#39;expression g&eacute;nique a r&eacute;v&eacute;l&eacute; qu&#39;un grand nombre de g&egrave;nes diff&eacute;rente dans l&#39;expression entre les deux lignes. R&eacute;pression des PI succ&egrave;de &agrave; celle de AP3 et pourrait &ecirc;tre la cons&eacute;quence d&#39;une perte d&#39;activit&eacute; AP3. Per acquisire nuove conoscenze nel meccanismo sottostante maschiosterilit&agrave; citoplasmatica (CMS), abbiamo confrontato i profili di espressione genica nucleare di fiori di una linea di Brassica napus CMS con quello della linea manutentore fertile b. napus utilizzando Arabidopsis thaliana fiore specifico cDNA Microarray. La linea CMS utilizzata ha un genoma nucleare b. napus, ma ha un genoma mitocondriale riarrangiati (mt) consistente di b. napus e di a. thaliana del DNA. Profilo di espressione genica ha rivelato che un gran numero di geni differiva in espressione tra le due linee. Ad esempio, geni nucleari che codificano per le proteine che sono coinvolte nell&#39;importazione di proteine in organelli, geni espressi in stami e polline, cos&igrave; come i geni implicati nel rimodellamento della parete cellulare o architettura, furono repressi in linea CMS rispetto a b. napus. Questi risultati mostrano che il genoma di mt della linea CMS influenza fortemente l&#39;espressione di gene nucleare e quindi rivelare l&#39;importanza della segnalazione retrograda tra i mitocondri e il nucleo. Inoltre, fiori della linea CMS sono caratterizzati da una sostituzione di stami con gli organi carpelloid e quindi parzialmente assomigliano alla APETALA3 (AP3) e mutanti striata (PI). Para obtener nuevos conocimientos sobre el mecanismo subyacente de la androesterilidad citoplasm&aacute;tica (CMS), se compararon los perfiles de expresi&oacute;n de genes nucleares de flores de una l&iacute;nea de Brassica napus CMS con el de la l&iacute;nea mantenedora de f&eacute;rtil B. napus utilizando microarrays de cDNA de flor espec&iacute;fica Arabidopsis thaliana. La l&iacute;nea CMS utilizada tiene un genoma nuclear B. napus, pero tiene un genoma mitocondrial cambiados (mt) de a. thaliana ADN tanto B. napus. Perfil de expresi&oacute;n g&eacute;nica revel&oacute; que un gran n&uacute;mero de genes se diferenci&oacute; en expresi&oacute;n entre las dos l&iacute;neas. Por ejemplo, genes nucleares que codifican para prote&iacute;nas que intervienen en la importaci&oacute;n de prote&iacute;nas en org&aacute;nulos, genes expresan en estambres y polen, as&iacute; como de genes implicados en la remodelaci&oacute;n de la pared celular o arquitectura, fueron reprimidos en la l&iacute;nea CMS en comparaci&oacute;n con B. napus. Estos resultados demuestran que el genoma de mt de la l&iacute;nea CMS influye fuertemente en expresi&oacute;n de genes nucleares y revelar as&iacute; la importancia de la se&ntilde;alizaci&oacute;n retr&oacute;grada entre las mitocondrias y el n&uacute;cleo. Adem&aacute;s, flores de la l&iacute;nea CMS se caracterizan por un reemplazo de estambres con &oacute;rganos de carpelloid y as&iacute; se asemejan parcialmente el APETALA3 (AP3) y PISTILLATA (PI) mutantes. De acuerdo con este fenotipo, AP3 expresi&oacute;n era regulada en los estambres, poco antes de que estos &oacute;rganos desarrollados carpelloid caracter&iacute;sticas, aunque se inici&oacute; correctamente. Represi&oacute;n de PI de AP3 tuvo &eacute;xito y podr&iacute;a ser una consecuencia de una p&eacute;rdida de actividad AP3. Estos resultados sugieren que AP3 expresi&oacute;n en estambres depende mt adecuada funci&oacute;n y una interacci&oacute;n nuclear-mt correcto, y que alteraciones mt causan el fenotipo de la esterilidad masculina de la l&iacute;nea CMS.

Phenotypic, genetic and molecular studies were made of Brassica napus lines with mitochondrial genomes consisting of DNA from both B. napus and Arabidopsis thaliana. The lines were isogenic regarding the nuclear and plastid genomes. Out of 21 lines, 10 were male-sterile, 3 semi-sterile and 8 male-fertile. Screening of the mitochondrial genomes with a dense set of A. thaliana specific markers showed that most lines contained large but variable portions of A. thaliana mitochondrial DNA. Several of the A. thaliana sequences in the mitochondrial genomes lead to the accumulation of novel transcripts. In addition, the restorer line showed different ability to restore male-fertility in the male-sterile lines. These results indicate that CMS is caused by several mitochondrial loci or combinations of loci. Beside petal and stamen morphology, growth rate and adenylate content varied among the lines. Furthermore, we found that the mitochondrial background had a distinct influence on nuclear gene expression. A clear example is the reduced expression of the two B-genes APETALA3 and PISTILATA in the male-sterile lines. From the studies made comparing the mitochondrial loci and the observed phenotypic alterations, our interpretation is that different loci in the mitochondrial genome influence nuclear gene expression via several retrograde signalling pathways.

Mitochondrial genotypes with variable parts of Arabidopsis thaliana DNA affect development in Brassica napus lines.

Les &eacute;tudes mol&eacute;culaires, g&eacute;n&eacute;tiques et ph&eacute;notypiques ont &eacute;t&eacute; faites de Brassica napus lignes avec des g&eacute;nomes mitochondriaux comprenant de l&#39;ADN de b. napus et l&#39;Arabidopsis thaliana. Les lignes &eacute;taient isog&eacute;niques concernant les g&eacute;nomes nucl&eacute;aires et des plastes. Sur les 21 lignes, 10 &eacute;taient des m&acirc;les-st&eacute;riles, 3 semi-st&eacute;rile et 8 m&acirc;le-fertile. Ces r&eacute;sultats indiquent que les CMS est caus&eacute;e par plusieurs locus mitochondriales ou combinaisons de loci. &Agrave; c&ocirc;t&eacute; de la morphologie des p&eacute;tales et des &eacute;tamines, croissance taux et l&#39;ad&eacute;nylate contenu vari&eacute; entre les lignes. En outre, nous avons constat&eacute; que l&#39;arri&egrave;re-plan mitochondrial avait un effet distinct sur l&#39;expression des g&egrave;nes nucl&eacute;aires. Un exemple clair est la r&eacute;duction de l&#39;expression des g&egrave;nes-B deux APETALA3 et PISTILATA dans les lign&eacute;es m&acirc;les-st&eacute;riles. Des &eacute;tudes faites en comparant les locus mitochondriales et les alt&eacute;rations ph&eacute;notypiques observ&eacute;es, notre interpr&eacute;tation est que les loci diff&eacute;rents dans le g&eacute;nome mitochondrial influence l&#39;expression des g&egrave;nes nucl&eacute;aires via plusieurs voies de signalisation r&eacute;trogrades.

Ph&auml;notypische, genetische und Molekulare Studien erfolgten Brassica Napus Linien mit mitochondrial Genome bestehend aus DNA von B. Napus und Arabidopsis Thaliana. Die Zeilen wurden hinsichtlich der nuklearen und Plastiden Genome isogene. Von 21 Zeilen, wurden 10 M&auml;nnchen steril, 3 semi-sterile und 8 M&auml;nnchen fruchtbar. Fr&uuml;herkennung der mitochondrial Genome mit einer dichten Reihe von A. Thaliana spezifische Marker zeigten, dass die meisten Reedereien gro&szlig;e aber Variable Teile von A. Thaliana mitochondriale DNA enthalten. Mehrere der A. Thaliana Sequenzen in die mitochondriale Genome dazu f&uuml;hren, dass die Anh&auml;ufung von Roman Abschriften. Dar&uuml;ber hinaus zeigte die Restaurator-Linie verschiedene M&ouml;glichkeit, m&auml;nnlich-Fruchtbarkeit in die m&auml;nnlichen-sterilen Linien wiederherzustellen. Diese Ergebnisse zeigen, dass CMS von mehreren mitochondrialen Loci oder Kombinationen von Loci verursacht wird. Neben Bl&uuml;tenblatt und staubbl&auml;tter Morphologie abwechslungsreiche Wachstum Rate und Adenylat-Inhalte zwischen den Zeilen. Dar&uuml;ber hinaus fanden wir, dass der mitochondriale Hintergrund einen deutlichen Einfluss auf die nukleare Genexpression hatte. Ein deutliches Beispiel ist der reduzierten Ausdruck der zwei B-Gene APETALA3 und PISTILATA in die m&auml;nnlichen-sterilen Linien. Aus den Studien gemacht, Vergleich der mitochondrialen Loci und die beobachteten ph&auml;notypischen Ver&auml;nderungen ist unsere Interpretation, dass verschiedene Loci im mitochondrialen Genom nukleare Genexpression &uuml;ber mehrere retrograde Signalwege beeinflussen.

Mitochondriale Genotypen mit Variablen Teilen von Arabidopsis Thaliana DNA beeinflussen Entwicklung in Brassica Napus Linien.

Studi fenotipici, genetici e molecolari erano fatte di Brassica napus linee con genoma mitocondriale che consiste di DNA da entrambi b. napus e Arabidopsis thaliana. Le linee erano isogeni per quanto riguarda il genoma nucleare e plastidio. Fuori 21 linee, 10 erano maschio-sterile, 3 8 e SEMISTERILE maschio fertile. Lo screening del genoma mitocondriale con un insieme denso di marcatori specifici di a. thaliana ha mostrato che la maggior parte delle linee contenevano variabile ma grandi porzioni di DNA mitocondriale di a. thaliana. Molte delle sequenze a. thaliana nel genoma mitocondriale portano all&#39;accumulo di trascrizioni romanzo. Inoltre, la linea ha mostrato diverse capacit&agrave; di ripristinare maschio fertilit&agrave; nelle linee maschio-sterile. Questi risultati indicano che CMS &egrave; causato da diversi loci mitocondriale o combinazioni di loci. Accanto a morfologia petalo e stame, crescita tasso e adenilato contenuto variava tra le linee. Inoltre, abbiamo trovato che lo sfondo mitocondriale ha avuto un&#39;influenza distinta sull&#39;espressione del gene nucleare. Un chiaro esempio &egrave; la ridotta espressione dei due geni-B APETALA3 e PISTILATA nelle linee maschio-sterile. Dagli studi fatti confrontando i loci mitocondriali e le alterazioni fenotipiche osservate, la nostra interpretazione &egrave; che diversi loci del genoma mitocondriale influenzano l&#39;espressione di gene nucleare tramite diverse vie di segnalazione retrograde.

Se realizaron estudios fenot&iacute;picos, gen&eacute;ticos y moleculares de Brassica napus l&iacute;neas con genomas mitocondriales consistente en DNA de B. napus tanto Arabidopsis thaliana. Las l&iacute;neas fueron isog&eacute;nicas sobre los genomas nucleares y Plasto. De 21 l&iacute;neas, 10 eran androest&eacute;riles, 3 semi-sterile y 8 hombre f&eacute;rtil. Proyecci&oacute;n de los genomas mitocondriales con un denso conjunto de marcadores espec&iacute;ficos de a. thaliana demostr&oacute; que la mayor&iacute;a de las l&iacute;neas contuvo variables pero grandes porciones del ADN mitocondrial de a. thaliana. Varias de las secuencias de a. thaliana en los genomas mitocondriales conducen a la acumulaci&oacute;n de las transcripciones de la novela. Adem&aacute;s, la l&iacute;nea restauradora demostr&oacute; capacidad diferente para restaurar la fertilidad del hombre en las l&iacute;neas androest&eacute;riles. Estos resultados indican que CMS es causada por varios loci mitocondriales o combinaciones de loci. Al lado de la morfolog&iacute;a del p&eacute;talo y estambre, crecimiento tasa y adenilato contenido vari&oacute; entre las l&iacute;neas. Adem&aacute;s, encontramos que el fondo mitocondrial ten&iacute;a una influencia clara en la expresi&oacute;n de genes nucleares. Un ejemplo claro es la expresi&oacute;n reducida de los dos B-genes APETALA3 y PISTILATA en las l&iacute;neas androest&eacute;riles. De los estudios realizados comparando los loci mitocondriales y las alteraciones fenot&iacute;picas observadas, nuestra interpretaci&oacute;n es que diversos lugares geom&eacute;tricos en el genoma mitocondrial influyan en la expresi&oacute;n de genes nucleares mediante varias v&iacute;as de se&ntilde;alizaci&oacute;n retr&oacute;gradas.

Flower development in plants depends not only on a set of nuclear genes but also on the coordinate action of the mitochondrion. Certain mitochondrial genomes in combination with certain nuclear genomes lead to the expression of cytoplasmic male-sterility (CMS). Both mitochondrial genes that determine male-sterility and nuclear Restorer-of-fertility genes that suppress the male-sterile phenotype have been cloned. Lately, the interactions between mitochondrial and nuclear genes through retrograde signalling in CMS-systems have been dissected. Of special interest are the altered expression patterns of floral homeotic genes in certain CMS-systems. Here, we review the mitochondrial influence on flower development and give examples from CMS-systems developed in Brassica, Daucus carota, Nicotiana tabacum and Triticum aestivum.

Certains g&eacute;nomes mitochondriaux en combinaison avec certains g&eacute;nomes nucl&eacute;aires conduisent &agrave; l&#39;expression de la st&eacute;rilit&eacute; m&acirc;le cytoplasmique (CMS). Les deux g&egrave;nes mitochondriaux qui d&eacute;terminent la st&eacute;rilit&eacute; m&acirc;le et g&egrave;nes nucl&eacute;aires de restaurateur de la fertilit&eacute; qui suppriment le ph&eacute;notype m&acirc;le-st&eacute;rile ont &eacute;t&eacute; clon&eacute;s. Ces derniers temps, les interactions entre les g&egrave;nes mitochondries et nucl&eacute;aires par le biais de la signalisation r&eacute;trograde dans les syst&egrave;mes CMS ont &eacute;t&eacute; diss&eacute;qu&eacute;es. Lo sviluppo del fiore nelle piante dipende non solo su un set di geni nucleari, ma anche sull&#39;azione coordinata del mitocondrio. Alcuni genomi mitocondriali in combinazione con alcuni genomi nucleari portano all&#39;espressione del maschio-sterilit&agrave; citoplasmatica (CMS). Geni mitocondriali che determinano la sterilit&agrave; maschile sia geni nucleari restauratore della fertilit&agrave; che sopprimono il fenotipo maschio-sterile sono stati clonati. Ultimamente, sono state sezionate le interazioni tra geni mitocondriali e nucleari attraverso segnalazione retrograda in sistemi di CMS. Di particolare interesse sono i modelli alterata espressione dei geni omeotici floreale in taluni sistemi di CMS. Desarrollo de la flor en las plantas depende no s&oacute;lo de un conjunto de genes nucleares, sino tambi&eacute;n en la acci&oacute;n coordinada de la mitocondria. Algunos genomas mitocondriales en combinaci&oacute;n con ciertos genomas nucleares conducen a la expresi&oacute;n del hombre-esterilidad citopl&aacute;smica (CMS). Genes mitocondriales que determinan hombre-esterilidad tanto de genes nucleares de restauradora de la fertilidad que suprimen el fenotipo androest&eacute;riles han sido clonados. &Uacute;ltimamente, las interacciones entre genes mitocondriales y nucleares a trav&eacute;s de se&ntilde;alizaci&oacute;n retr&oacute;grada en CMS-sistemas han sido disectadas. De especial inter&eacute;s son los patrones de expresi&oacute;n alterada de genes home&oacute;ticos floral en ciertos sistemas CMS. Aqu&iacute; revisamos mitocondrial influencia en el desarrollo de la flor y dar ejemplos de sistemas CMS desarrollaron en Brassica, Daucus carota, Nicotiana tabacum y Triticum aestivum.

Global glycosylation changes of serum proteins in type 1 diabetic patients have in this paper been investigated based on the interaction of the saccharide moiety of serum proteins with different lectins. Lectins are proteins, which bind carbohydrates specifically and reversibly. Panels with lectins of various carbohydrate specificities were immobilized on gold surfaces. Sera from healthy individuals, newly diagnosed type 1 diabetes patients and type 1 diabetes patients having had the disease for 4-6 years, respectively, were applied to the lectin panel. The biorecognition was evaluated with null ellipsometry. Data obtained were related to an internal standard of lactoferrin. Multivariate data analysis (MVDA) techniques were used to analyze data. Principal component analysis showed that the lectin panel enabled discrimination between sera from the three different above-mentioned groups. Using an artificial neuronal net (ANN), it was possible to correctly categorize unknown serum samples into one of the three groups.

Lectines sont des prot&eacute;ines qui lient les hydrates de carbone sp&eacute;cifiquement et de fa&ccedil;on r&eacute;versible. Cambios globales de la glicosilaci&oacute;n de prote&iacute;nas de suero en pacientes con diabetes tipo 1 en el presente trabajo se investigaron basado en la interacci&oacute;n de la mol&eacute;cula de sacarosa de prote&iacute;nas s&eacute;ricas con diferentes lectinas. Lectinas son prote&iacute;nas que unen carbohidratos espec&iacute;ficamente y reversible. Paneles con lectinas de diversas especificidades de carbohidratos fueron inmovilizados sobre superficies de oro. Sueros de individuos sanos, reci&eacute;n diagnosticado pacientes con diabetes 1 tipo y tipo 1 pacientes de diabetes, haber tenido la enfermedad durante 4 a 6 a&ntilde;os, respectivamente, se aplicaron al panel de lectinas. El biorreconocimiento fue evaluada con Elipsometr&iacute;a nulo. Datos obtenidos se relacionaron con una norma interna de lactoferrina. Se utilizaron t&eacute;cnicas de an&aacute;lisis (MVDA) de datos multivariantes para analizar los datos. We have developed a sensitive and specific method based on surface plasmon resonance (SPR) for detection of insulin autoantibodies (IAA) in serum samples from individuals at high risk of developing type 1 diabetes (T1D). When measuring trace molecules in undiluted sera with label-free techniques like SPR, non-specific adsorption of matrix proteins to the sensor surface is often a problem, since it causes a signal that masks the analyte response. The developed method is an indirect competitive immunoassay designed to overcome these problems. Today, IAA is mainly measured in radio immunoassays (RIAs), which are time consuming and require radioactively labeled antigen. With our SPR-based immunoassay the overall assay time is reduced by a factor of >100 (4 days to 50min), while sensitivity is maintained at a level comparable to that offered by RIA.

BACKGROUND: The Swedish pharmacy market is presently being reregulated. The state-owned pharmacy monopoly company was divided and sold during 2009, and certain nonprescription medicines are now allowed in nonpharmacy settings. The changes will likely affect the pharmacist's role and the image of the community pharmacist in society. This change may affect how pharmacists are seen by society at large, and therefore, a baseline showing how pharmacists are depicted before the reregulation is of great value. OBJECTIVES: The aim of this study was to describe how the pharmacist profession is depicted in print media in Sweden, with a focus on community pharmacy. METHODS: A deductive qualitative content analysis with material from print media was conducted, using professional criteria as a framework. Swedish print media from October 2005 to October 2008 were searched and all relevant articles included. RESULTS: A total of 139 articles were included. Most articles came from professional journals, that is, journals directed toward pharmacist or related professions. The results show that the pharmacist profession is not highly visible and that this lack of visibility is disappointing to pharmacists. CONCLUSIONS: Society, as reflected in print media, does not display an awareness of the pharmacist role in Sweden. Although this is disappointing for the profession, it allows pharmacists to influence the depiction and hence their position in society.

FONDO: El mercado de farmacia Sueca actualmente est&aacute; siendo reregulated. La compa&ntilde;&iacute;a de monopolio estatal de la farmacia fue dividida y vendida durante el 2009, y ciertos medicamentos sin receta ahora est&aacute;n permitidos en la configuraci&oacute;n de nonpharmacy. Los cambios afectar&aacute;n probablemente papel del farmac&eacute;utico y la imagen del farmac&eacute;utico comunitario en la sociedad. Este cambio puede afectar c&oacute;mo farmac&eacute;uticos son vistos por la sociedad en general, y por lo tanto, es una l&iacute;nea de base que muestra c&oacute;mo se representan los farmac&eacute;uticos antes la re-regulaci&oacute;n de gran valor. OBJETIVOS: El objetivo de este estudio fue describir c&oacute;mo se representa la profesi&oacute;n de farmac&eacute;utico en medios impresos en Suecia, con un enfoque en la farmacia comunitaria. M&Eacute;TODOS: Se realiz&oacute; un an&aacute;lisis de contenido cualitativo deductivo con material de prensa, utilizando criterios profesionales como marco. Sueco medios impresos desde octubre de 2005 a octubre de 2008 fueron buscados y todos los art&iacute;culos incluidos. Resultados: Se incluyeron un total de 139 art&iacute;culos. La mayor&iacute;a de art&iacute;culos proceden de revistas profesionales, es decir, revistas destinaba a su farmac&eacute;utico o relacionadas con las profesiones. Los resultados muestran que la profesi&oacute;n de farmac&eacute;utico no es muy visible y que esta falta de visibilidad es decepcionante para farmac&eacute;uticos. CONCLUSIONES: La sociedad, como se refleja en los medios impresos, no muestra una conciencia sobre el papel del farmac&eacute;utico en Suecia. Aunque esto es decepcionante para la profesi&oacute;n, permite farmac&eacute;uticos influir en la representaci&oacute;n y por lo tanto su posici&oacute;n en la sociedad.
