We bedanken Mark Terasaki voor een eerste advies over het gebruik van een twee-foton microscoop aan lokale weefselbeschadiging, Kathy Joubin voor advies over montage voor time-lapse imaging, en de Sagasti en portierster-Cailliau labs voor discussie te creëren. Wij hebben gebruik gemaakt van de methoden beschreven om precies te axotomize perifere sensorische axonen en om direct herstel waarnemen in de levende zebravis embryo. Lange-termijn time-lapse confocale beeldvorming in de zebravis kan worden gebruikt om een groot aantal ontwikkelingsprocessen in vivo te observeren. De twee-foton beschreven axotomy procedure kan worden aangepast voor verschillende experimentele doelen. Zebravissen zijn al lange tijd gebruikt om de cellulaire en moleculaire mechanismen van ontwikkeling studie van time-lapse beeldvorming van de levende transparante embryo. Hier beschrijven we een methode om de zebravis embryo&#39;s mount voor de lange termijn beeldvorming en demonstreren hoe de vangst van time-lapse beelden met behulp van een confocale microscoop te automatiseren. We beschrijven ook een methode om gecontroleerde, precieze schade aan de individuele vestigingen van perifere sensorische axonen in zebravis met behulp van de gerichte kracht van een femtoseconde laser gemonteerd op een twee-foton microscoop te creëren. De parameters voor succesvolle twee-foton axotomy moet worden geoptimaliseerd voor elke microscoop. We zullen aantonen twee-foton axotomy op zowel een op maat gemaakte twee-foton microscoop en een confocale Zeiss 510 / twee-foton om twee voorbeelden te geven. Zebravis trigeminale sensorische neuronen kunnen worden gevisualiseerd in een transgene lijn te drukken GFP aangedreven door een sensorisch neuron specifieke promoter 1. We hebben dit aangepast zebravis trigeminus model voor zintuiglijke regeneratie van axonen direct waarnemen in de levende zebravis embryo&#39;s. Embryo&#39;s worden verdoofd met tricaïne en gepositioneerd binnen een druppel agarose als het stolt. Geïmmobiliseerd embryo&#39;s worden afgesloten binnen een imaging kamer gevuld met phenylthiourea (PTU) Ringers. Wij hebben geconstateerd dat embryo&#39;s kan continu worden afgebeeld in deze kamers voor 12-48 uur. Een enkele confocale beeld wordt vervolgens opgeslagen op de gewenste plaats van axotomy te bepalen. De regio van belang is gelegen op de twee-foton microscoop door beeldvorming van de sensorische axonen onder lage, niet-beschadigende macht. Na het inzoomen op de gewenste plaats van axotomy, is de kracht verhoogd en een enkele scan van die bepaalde regio is voldoende om de axon te verbreken. Meerdere locatie time-lapse imaging wordt dan ingesteld op een confocale microscoop om direct waar te nemen axonale het herstel van een blessure.
