optical mapping of langendorff-perfused rat hearts

Mappatura ottica della superficie cardiaca con coloranti fluorescenti voltaggio-sensibile è diventato un importante strumento per indagare l&#39;eccitazione elettrica in modelli sperimentali che vanno in scala da colture di cellule di interi organi<sup&gt; [1, 2]</sup&gt;. Utilizzando state-of-the-art sistemi di imaging ottico, generazione e propagazione dei potenziali d&#39;azione durante il normale ritmo cardiaco o per tutto l&#39;inizio e la manutenzione di aritmie possono essere visualizzati quasi istantaneamente<sup&gt; [1]</sup&gt;. L&#39;ultima disponibile in commercio di sistemi in grado di fornire informazioni spazio-temporali a risoluzioni estremamente elevate e si basano su custom-built attrezzature inizialmente sviluppato per superare gli ostacoli imposti dalla elettrofisiologici metodi più convenzionali<sup&gt; [1]</sup&gt;. I progressi in alta risoluzione e ad alta velocità Complementary Metal-Oxide-Semiconductor (CMOS), macchine fotografiche e intensamente brillante, diodi emettitori di luce (LED), così come tensione-sensibili coloranti, ottica, ed i filtri hanno cominciato a fare l&#39;aquisizione del segnale elettrico pratiche per i biologi cellulari cardiovascolari che sono più abituati a lavorare con microscopi. Anche se l&#39;ultima generazione di telecamere CMOS in grado di acquisire 10.000 frame al secondo su di un array 16.384 pixel, a seconda del tipo di preparazione del campione, ormai consolidata tecnologie di acquisizione di fluorescenza come array fotodiodo, sistemi di scansione laser, e raffreddato charged-coupled device (CCD telecamere) hanno ancora alcuni distinti vantaggi rispetto alla gamma dinamica e rapporto segnale-rumore e efficienza quantica<sup&gt; [1, 3]</sup&gt;. Nel presente studio, i cuori di ratto Lewis sono stati perfusi<em&gt; Ex vivo</em&gt; Con un perfusato cristalloidi (Krebs-Henseleit soluzione) a 37 ° C in un apparecchio Langendorff modificato. Dopo un periodo di stabilizzazione di 20 minuti, i cuori erano intermittente perfusi con 11 mmol / L 2,3-butanedione monoxime per eliminare le contrazioni associate movimento durante l&#39;acquisizione dell&#39;immagine.
