<p class="jove_content">Knaagdier operatie is vaak een belangrijk onderdeel bij de beoordeling van het nut van gemanipuleerde weefsels. Een grote verscheidenheid aan chirurgische ingrepen kunnen worden uitgevoerd in gemeenschappelijke laboratorium ratten of muizen en deze vrij vaak fungeren als een tussenstap tussen de bench-top experimenten en grote dieren testen of proeven op mensen. Gezien het feit dat knaagdieren een gevestigde, kosten-effectief, en fysiologisch relevante model systeem waarin tot nieuwe combinaties van materialen steigers en cellen te testen bieden, zijn ze bijzonder goed geschikt voor cardiovasculaire tissue engineering studies. Op dit moment, beschrijven we een open-hart chirurgische ingreep om engineered weefsel met myogene progenitor cellen in de atrioventriculaire (AV) groef van een rat hart implantaat. Deze implantaten zijn bedoeld om een ​​elektrische leiding tussen de rechterboezem en de rechterkamer te maken met het uiteindelijke doel van het verstrekken van een alternatieve behandeling voor conventionele pacemaker implantatie bij pediatrische patiënten met een complete hartblok [1]. De kunstmatige weefsel wordt geïmplanteerd in de AV-groef door middel van een thoracotomie. Voor ons doel, Lewis ratten zijn verdoofd en invasieve geventileerd om positive airway pressure tijdens de steriele chirurgische procedure te behouden. De aanpak van het hart wordt uitgevoerd door een rechter thoracotomie via een antero-laterale incisie op het 5<sup&gt; E</sup&gt; Intercostale ruimte. Het weefsel construct is vastgesteld in de AV-groef met een enkele 7-0 Prolene hechtdraad en gepositioneerd tussen de rechter ventrikel en atrium aan de ventrale deel van het hart. Het epicard is gedeeltelijk verwijderd om direct contact tussen de ontvanger myocardiale cellen en die welke zijn opgenomen in de gemanipuleerde weefsel mogelijk te maken. Na implantatie, de borstwand wordt gesloten in lagen, is elke pneumothorax geëvacueerd, en het dier is geëxtubeerd en behandeld met pijnstillende.</p>
