<p class="jove_content">Nager Operation ist oft ein wichtiger Bestandteil bei der Beurteilung der Nützlichkeit von technischen Geweben. Eine Vielzahl von chirurgischen Verfahren können in gemeinsamen Labor Ratten oder Mäusen durchgeführt werden und diese sehr häufig als ein Zwischenschritt zwischen Tisch-Experimente und große Tierversuche oder Versuche am Menschen dienen. Da die Nagetiere eine etablierte, kostengünstige und physiologisch-relevanten Modellsystem, in dem neuartige Kombinationen von Gerüsten Materialien und Zellen zu testen bieten, eignen sie sich besonders für Herz-Kreislauf-Tissue-Engineering-Studien gut geeignet. Derzeit stellen wir eine am offenen Herzen chirurgische Verfahren entwickelt, Gewebe mit myogenen Vorläuferzellen in der atrioventrikulären (AV) Nut einer Ratte Herz zu implantieren. Diese Implantate sollen eine elektrische Leitung zwischen dem rechten Vorhof und die rechte Herzkammer mit dem ultimativen Ziel, eine alternative Behandlung zu konventionellen Schrittmacher-Implantation bei pädiatrischen Patienten mit komplettem AV-Block [1] zu schaffen. Die gezüchteten Gewebe ist in der AV-Nut mittels einer Thorakotomie implantiert. Für unsere Zwecke sind Lewis-Ratten betäubt und invasiv beatmet, um positive Atemwegsdruck während des sterilen OP-Verfahren aufrecht zu erhalten. Der Ansatz des Herzens wird durch ein Recht Thorakotomie durch eine anterolaterale Inzision am 5 durchgeführt<sup&gt; Th</sup&gt; Interkostalraum. Das Gewebe Konstrukt ist in der AV-Nut mit einem einzigen 7-0 Prolene Naht fixiert und positioniert zwischen dem rechten Ventrikel und Atrium am ventralen Teil des Herzens. Das Epikard ist teilweise entfernt, um den direkten Kontakt zwischen dem Empfänger Herzmuskelzellen und die in der gezüchteten Gewebe enthalten kann. Nach der Implantation, der Brustwand wird schichtweise verschlossen ist jede Pneumothorax evakuiert, und das Tier extubiert und mit Analgetikum.</p>
