Die Familie<em&gt; Poxviridae</em&gt; Besteht aus großen doppelsträngigen DNA-Viren enthalten, die ausschließlich im Zytoplasma der infizierten Zellen replizieren. Die Mitglieder des<em&gt; Orthopox</em&gt; Gattung gehören Variola, dem Erreger der menschlichen Pocken, Affenpocken und Vaccinia (VAC), der prototypische Mitglied der Virus-Familie. Innerhalb der relativ groß (~ 200 kb) Vaccinia-Genom gibt drei Klassen von Genen kodiert: frühe, mittlere und spät. Während alle drei Klassen von viral-kodierten RNA-Polymerasen transkribiert werden, dient jede Klasse eine andere Funktion im Lebenszyklus des Virus. Pockenviren nutzen mehrere Strategien zur Modulation der zellulären Umgebung Host während der Infektion. Um Regulierung der Wirt und Virus Genexpression verstehen, müssen wir genomweite Ansätze genutzt werden, um Transkript Fülle von beiden Viren und Wirtszellen zu analysieren. Hier zeigen wir, zeitlicher Verlauf Infektionen von HeLa-Zellen mit Vaccinia-Virus und Sampling-RNA zu verschiedenen Zeitpunkten nach der Infektion. Beide Gastgeber und virale RNA isoliert und verstärkt für die Hybridisierung an Microarrays zur Analyse der Genexpression.
