vaccinia virus infection &amp; temporal analysis of virus gene expression: part 1

Familjen<em&gt; Poxviridae</em&gt; Består av stora dubbelsträngat DNA som innehåller virus som replikerar endast i cytoplasman av infekterade celler. Medlemmar i<em&gt; Orthopox</em&gt; Släktet inkluderar variola, vilka smittämnen av mänskliga smittkoppor, monkeypox och vaccinia (VAC), den prototypic medlem av viruset familjen. Inom det relativt stora (~ 200 kb) vaccinia genomet, finns tre klasser av gener kodade: tidig, mellan-och sena. Även om alla tre klasser Kopierat av viralt-kodade RNA-polymeraser, serverar varje klass en annan funktion i livscykeln av viruset. Poxviruses utnyttja flera strategier för modulering i den mottagande cellulära miljön under infektion. För att förstå regleringen av både värd och virus genuttryck, har vi utnyttjat genomet-strategier för att analysera avskrift överflöd från både virus och celler värd. Här visar vi infektioner tid under HeLa celler med Vaccinia virus och RNA provtagning vid flera olika tidpunkter efter infektion. Både värd och virus total RNA isoleras och förstärks för hybridisering med microarrays för analys av genuttryck.
