De familie<em&gt; Poxviridae</em&gt; Bestaat uit grote double-stranded DNA die virussen bevatten die uitsluitend repliceren in het cytoplasma van de geïnfecteerde cellen. De leden van de<em&gt; Orthopoxvirus</em&gt; Genus behoren variola, de verwekker van de menselijke pokken, apenpokken, en vaccinia (VAC), het prototype lid van het virus familie. Binnen de relatief grote (~ 200 Kb) vaccinia genoom, zijn drie klassen van genen gecodeerd: vroege, intermediaire en late. Terwijl alle drie de klassen zijn getranscribeerd door viraal gecodeerde RNA-polymerasen, elke klas heeft een ander functie in de levenscyclus van het virus. Poxviruses gebruik van meerdere strategieën voor modulatie van de host-cellulaire omgeving tijdens de infectie. Met het oog op regulering van zowel de gastheer en virus genexpressie te begrijpen, hebben we gebruik gemaakt genoom-brede aanpak van transcript overvloed analyseren van zowel virus en gastheercellen. Hier laten we zien tijdsverloop infecties van de HeLa cellen met Vaccinia virus en bemonstering RNA op verschillende tijdstippen na infectie. Zowel de host-en virale totaal RNA wordt geïsoleerd en versterkt voor hybridisatie met microarrays voor de analyse van genexpressie.
