cellular toxicity of nanogenomedicine in mcf-7 cell line: mtt assay

<p class="jove_content">Cytotoxiciteit van de futuristische nanogenomedicine (bijvoorbeeld short interfering RNA en antisense) kunnen belemmeren haar klinische ontwikkeling. Van deze kunnen de gen-gebaseerde geneeskunde en / of de drager ervan te lokken cellulaire toxiciteit. Voor de beoordeling van deze cytotoxiciteit, een gemeenschappelijke methodologie is grotendeels afhankelijk van gebruik van de 3 - (4, 5-dimethyl-2-thiazolyl) -2, 5-difenyl-2H-tetrazolium bromide (MTT) test die veelvuldig gebruikt als een colorimetrische aanpak, gebaseerd op de activiteit van de mitochondriale dehydrogenase enzymen in de cellen. In dit huidige onderzoek werden MCF-7 cellen geïnoculeerd in 96-wells plaat en op 50% confluentie ze werden behandeld met verschillende nanopolyplexes en onderworpen aan test na 24 uur MTT. Water oplosbaar geel MTT wordt gemetaboliseerd door het metabool actieve cellen aan het water onoplosbare paarse formazan, die verder wordt opgelost in buffer dimethylsulfoxide en Sornson s pH 10,5. Het resulterende product kan worden gekwantificeerd door spectrofotometrie met behulp van een plaat reader bij 570 nm.</p>
