Twee aspecten van de P. abies protocol werden geoptimaliseerd ten opzichte van de A. thaliana / B. napus protocol, RNase behandeling en proteinase K concentratie. De RNase verwijdert achtergrond signalen en verhoogt zo het signaal specificiteit 17. De proteinase K behandeling is nodig om de weefsels permeabilize, zodat de sonde kunt invoeren en hybridiseren met het RNA pool van belang. De DAL13 antisense probe gaf een hogere signaal zonder RNase-behandeling (Fig. 3C, E, G) in vergelijking met secties behandeld met RNase gedurende 30 minuten (figuur 2D, F en H). Sinds de RNase-behandeling niet verbeteren het signaal dat werd verwijderd uit het protocol. De proteinase K behandeling werden getest op drie verschillende concentraties, 1, 3 en 5 ug / ml. In het geval van P. abies een lage of geen signaal werd waargenomen met behulp van 1 ug / ml proteinase K (fig. 3C-D). Een sterk signaal werd verkregen met zowel 3 ug / ml (Afb. 3E-F) en 5 ng / ml (afb. 3G-H) proteïnase K. Aangezien er geen duidelijk verschil in signaalsterkte werd gevonden bij het gebruik van 5 ug / ml proteinase K in vergelijking met 3 ug / ml, hebben we ervoor gekozen om 3 ug / ml te gebruiken in ons protocol. Het is waarschijnlijk dat de noodzaak van een hogere proteinase K concentratie in de P. abies mannelijke kegels ten opzichte van de A. thaliana en B. napus bloemen knoppen is te wijten aan compacter weefsel met hogere niveaus van fenolen en polysachariden. Tijdens de fixatie en inbedding enkele verschillen tussen de A. thaliana / B. napus protocol en de P. abies protocol zijn evident. Het weefsel van belang is vast te RNA retentie te bieden en dit is een belangrijke stap, omdat slecht vaste materialen zou kunnen geven een lage of niet detecteerbare signaal ook al is het mRNA is zeer overvloedig. Het weefsel is uitgedroogd, gewist en ingebed in was in de geleidelijke veranderingen van weefsel schade te voorkomen, en in sommige protocollen 0,85% NaCl wordt toegevoegd aan de ethanol in de uitdroging te vermijden om verdere krimp en zwelling van de cellen 3. De uitdroging stap tijdens inbedden kan worden uitgevoerd op het ijs om RNase activiteit houden minimum te beperken. In de P. abies protocol van de 4% paraformaldehyde fix-oplossing bevat 0,25% glutaaraldehyde, die geacht wordt om een betere RNA retentie geven 17 ook al is dat sommige mensen beweren dat het waarschijnlijk geen verschil 3 te maken. Na het snijden van het materiaal is bevestigd op dia&#39;s en voorbehandeld (dat wil zeggen was ontdaan, gerehydrateerd, gepermeabiliseerde, getrakteerd op niet-specifieke binding van de probe en tenslotte gedroogd te verminderen) voor de hybridisatie. In het protocol hier gepresenteerde de hele detectie-procedure kan worden uitgevoerd in gewone laboratoria, het signaal ontwikkelt zich meestal binnen 1-3 dagen en de verhouding tussen signaal over achtergrond kan doorlopend worden gecontroleerd. Het DIG-gelabelde probes meestal geven een duidelijker signaal in vergelijking met radioactief gelabelde probes, zijn stabieler en kan worden hergebruikt in opeenvolgende experimenten. Aan de andere kant, is de gevoeligheid verminderd in vergelijking met radioactieve probes 17. In situ hybridisatie detecteert uitdrukking met een gemerkte probe die specifiek is voor een bepaalde mRNA. Radioactieve labeling is een robuuste en gevoelige etikettering methode, maar het vereist de behandeling van radioactiviteit en het duurt enkele weken voordat de resultaten kunnen worden gedetecteerd. Antigeen-gelabelde probes aan de andere kant, zijn stabieler, minder gevaarlijk en vereisen de blootstelling aan de detectie medium voor een paar dagen maar 17. Zo zijn antigeen-labelling methoden die momenteel domineert. De meest kritische fase, ongeacht het protocol is de sonde ontwerp. Zorg moeten worden genomen om ervoor te zorgen dat de sonde heeft een hoge specificiteit en is voorzien van een hoge kwaliteit UTPs. Soms zijn de hybridisatie temperatuur en / of de reactie lengte moeten worden aangepast voor specifieke probes. Onze gewijzigd protocol op basis van DIG-gelabelde probes zal vergemakkelijken lokalisatie van mRNA-expressie tegelijk in soorten, variërend van A. thaliana aan P. abies, en moet met kleine aanpassingen bruikbaar in andere plantensoorten. Het protocol heeft, naast de mannelijke kegels, is ook getest op verschillende stadia van vegetatieve scheuten en in zowel zygotische en somatische embryo&#39;s van P. abies met goede resultaten (niet gepubliceerde resultaten;. Karlgren et al.).

De high-throughput expressie analyse technieken die vandaag beschikbaar zijn geven wetenschappers een overloop van expressie profielen, maar hun resolutie op het gebied van weefsel-specifieke expressie is beperkt als gevolg van problemen in ontleden individuele weefsels. Uitdrukking gegevens moeten worden bevestigd en aangevuld met expressie patronen met behulp van bijvoorbeeld in situ hybridisatie, gebruikt een techniek om lokaliseren cel-specifieke mRNA-expressie. De in situ hybridisatie methode is omslachtig, tijdrovend en vaak vereist uitgebreide optimalisatie, afhankelijk van diersoort en weefsels. In situ experimenten zijn relatief moeilijker uit te voeren in een bosrijke soorten, zoals de conifeer Noorwegen spar (Picea abies). Hier hebben we een gewijzigd DIG in situ hybridisatie protocol, dat is snel en van toepassing op een breed scala aan plantensoorten, waaronder P. abies. Met slechts een paar aanpassingen, zoals gewijzigd RNase behandeling en proteinase K concentratie, kunnen we gebruiken het protocol bij weefselspecifieke expressie van homologe genen in mannelijke voortplantingsorganen van een naaktzadige en twee bloemplant soorten te bestuderen; P. abies, Arabidopsis thaliana en Brassica napus. Het protocol werkt even goed voor de soorten en genen bestudeerd. AtAP3 en BnAP3 werden waargenomen in de tweede en derde krans bloemen organen in A. thaliana en B. napus en DAL13 in microsporophylls van de mannelijke kegels van P. abies. Voor P. abies de proteinase K concentratie, gebruikt om de weefsels permeablize, moest worden verhoogd tot 3 g / ml in plaats van 1 g / ml, mogelijk als gevolg van meer compacte weefsels en hogere niveaus van fenolen en polysachariden. Voor alle soorten van de RNase behandeling werd als gevolg van verminderde signaalsterkte verwijderd zonder een corresponderende toename van de specificiteit. Door het vergelijken van weefsel-specifieke expressie patronen van homologe genen van beide bloeiende planten en een naaldboom we laten zien dat de DIG in situ-protocol hier voorgesteld, met slechts minuten aanpassingen, kan worden toegepast op een breed scala aan plantensoorten. Vandaar dat het protocol voorkomt zowel uitgebreide species specifieke optimalisatie en de moeizame gebruik van radioactief gelabelde probes in het voordeel van DIG gemerkte probes.
