Een in vitro systeem dat recapituleert de ontwikkeling en differentiatie van voorlopers tot rijpe neuronen en glia in het centrale zenuwstelsel (CZS), zou een krachtig platform voor neurowetenschappers aan Axo-gliale interacties, eigenschappen en differentiatie van multipotente voorlopercellen, en de progressie van oligodendrogliale onderzoeken lijn cellen op cellulair en moleculair niveau. We beschrijven hier een CNS aggregaat cultuur systeem van embryonale rat forebrains, die kan worden gehandhaafd in een serum-vrij medium tot 3-4 weken en wordt in ons laboratorium als een model om neuron-glia interactie en CNS myelinisatie te bestuderen. Deze video clip zal tonen hoe te isoleren en deze CNS geaggregeerde culturen groeien van E16 rat hersenen. Bovendien, uit dezelfde hersenen dissectie, kan hoogverrijkt regelmatig los neuronale culturen gemakkelijk worden verkregen en gebruikt voor diverse studies over CNS neuronen of gebruikt worden voor co-culturen met andere cellen.
