Insgesamt internen Reflexion Fluoreszenz (TIRF)-Mikroskopie ist eine Technik, dass die Studie die Geschehnisse an der Zellmembran ermöglicht, durch selektive Darstellung von fluoreszierenden Molekülen, die zu einem hohen Brechungsindex Substanz wie Glas am nächsten sind,<sup&gt; 1</sup&gt;. In diesem Artikel wenden wir diese Technik, um Bild Exozytose synaptischer Vesikel in der Netzhaut bipolaren Zellen aus dem Goldfisch Netzhaut isoliert. Diese Neuronen sind sehr geeignet für diese Art der Studie aufgrund ihrer großen Axonterminalen. Durch die gleichzeitige Patch-Clamp der bipolaren Zellen, ist es möglich, die Beziehung zwischen präsynaptischen Spannung und synaptische Freisetzung untersuchen<sup&gt; 2,3</sup&gt;. Synaptischer Vesikel innerhalb der bipolaren Zellen Terminals sind mit einem Fluoreszenzfarbstoff (FM 1-43 ®) von geladenen Co-schnaufend dem Farbstoff und einem Ringer-Lösung mit einem hohen K<sup&gt; +</sup&gt; Konzentration auf die synaptischen Terminals. Dies depolarisiert die Zellen und regt die Endozytose und damit Farbstoffaufnahme in die glutamaterge Vesikel. Nach dem Waschen der überschüssige Farbstoff entfernt für etwa 30 Minuten werden die Zellen bereit für sein Patch gespannt und bebildert gleichzeitig mit einem 488 nm-Laser. Die Patch-Pipette Lösung enthält ein Rhodamin-basierte Peptid, das selektiv an den synaptischen Bandes Protein RIBEYE<sup&gt; 4</sup&gt;, Damit die Kennzeichnung Bänder speziell, wenn die Klemmen mit einem 561 nm Laser abgebildet werden. Dies ermöglicht die genaue Lokalisierung der aktiven Zonen und die Trennung von synaptischen aus extra-synaptischen Ereignisse.
