<p style=";text-align:right;direction:rtl"> CD40 המופעל בתאי B (CD40-B תאים) זוהו כמקור חלופי חיסונית של גירוי התאים המציגים אנטיגנים (APC) עבור חיסוני סרטן <sup>1-3.</sup> לעומת תאים דנדריטים (DCS), APC מאופיין הטובה ביותר, תאים CD40-B יש מספר מאפיינים ביולוגיים טכני מובהק. בדומה DCs, תאים B להראות ביטוי מוגבר של MHC ושיתוף גירוי מולקולות (Fig.1b), מפגינים יכולת הנדידה חזק מצגת להציג ביעילות אנטיגן ותאי T, לאחר גירוי עם ליגנד interleukin-4 ו CD40 (CD40L). עם זאת, בניגוד DCs בוגר או בוגרת, CD40 בתאי B-לבטא את לימפה הצומת השלישיה ביות מלא המורכב CD62L, CCR7/CXCR4, ותפקוד ליקוציט אנטיגן-1 (LFA1, CD11a/CD18), הכרחי ביות הלימפה לאיברים משני (Fig.1a) <sup>3.</sup> תאים CD40-B יכול להיווצר ללא קשיים של כמויות קטנות מאוד של דם היקפיים אשר ניתן להרחבה נוספת <em>במבחנה</em> על כמויות גדולות מאוד של הפערים טהור CD40-B תאים סרטניים (&gt; 10 <sup>9</sup> תאים לכל מטופל) מתורמים בריאים, כמו גם חולים (Fig.1c, ד) <sup>1,4.</sup> </p><p style=";text-align:right;direction:rtl"> בפרוטוקול זה אנו מדגימים כיצד להשיג מופעל במלואו CD40-B תאים PBMC האדם. מולקולות מרכזיות בתרבות התא CD40 ליגנד, interleukin-4 (IL-4) ציקלוספורין A (CSA), אשר מתחדש בכל יום מחזור 3-4 התרבות. לענין מעבדה CD40 גירוי מסופק על ידי תאים NIH/3T3 להביע רקומביננטי אנושי CD40 ליגנד (tCD40L NIH/3T3) <sup>5.</sup> כדי למנוע זיהום עם הלא transfected תאים, ביטוי של ליגנד CD40 האדם על transfectants יש להיבדק באופן קבוע (Fig.2). </p><p style=";text-align:right;direction:rtl"> לאחר 14 ימים CD40-B בתרביות תאים מורכבים יותר טהור 95% תאים B ו הרחבה של CD40-B תאים מעל 65 ימים ניתן לעתים קרובות ללא כל אובדן של <sup>1 לתפקד, 4.</sup> תאים CD40-B ביעילות לקחת, תהליך להציג אנטיגנים לתאי T <sup>6.</sup> הם לא רק naϊve הממשלה, אלא גם להרחיב את הזיכרון בתאי T <sup>7,8.</sup> CD40 המופעל בתאי B יכול לשמש כדי ללמוד B-cell, הפעלת התמיינות ותפקוד. יתר על כן, הם מייצגים כלי מבטיח עבור חיסון או טיפול מונע נגד גידולים <sup>9.</sup> </p>
