<p class="jove_content">RNA-interferentie (RNAi) is een systeem van gene silencing in levende cellen. In RNAi, genen homoloog in volgorde om korte interfererende RNA (siRNA) worden monddood gemaakt bij de post-transcriptionele staat. Short hairpin RNA&#39;s, voorlopers van siRNA, kan worden uitgedrukt met behulp van lentivirus, waardoor RNAi in een verscheidenheid van celtypen. Lentivirussen, zoals het Human Immunodeficiency Virus, in staat zijn om infecteren zowel delende als niet-delende cellen. We beschrijven een procedure die tot pakket lentivirussen. Verpakkingen verwijst naar de voorbereiding van de bevoegde virus uit DNA vectoren. Lentivirale vector productie systemen zijn gebaseerd op een &#39;split&#39; systeem, waar de natuurlijke virale genoom is opgesplitst in afzonderlijke helper plasmide constructen. Deze splitsing van de verschillende virale elementen in vier afzonderlijke vectoren vermindert het risico van een replicatie-capabele virus door toevallige recombinatie van de lentivirale genoom. Hier wordt een vector die de shRNA van rente en drie verpakkingen vectoren (p-VSVG, pRSV, pMDL) tijdelijk getransfecteerd in menselijke 293-cellen. Na ten minste een 48-uur incubatieperiode, is het virus bevat supernatant geoogst en geconcentreerd. Tot slot wordt virustiter bepaald door de verslaggever (TL), expressie met een flowcytometer.</p>
