<p> Große Fortschritte in unserem Verständnis der Entwicklung des ZNS und Stammzellbiologie wurden möglich durch unsere Fähigkeit zu ernten, zu isolieren und Kultur embryonalen neuralen Stammzellen. Dieses Video zeigt die Zerlegung von E12.5 Maus Großhirnrinde und die anschließende Zerlegung und Kultivierung von embryonalen neuralen Stammzellen. Viele andere ähnliche Methoden wurden erfolgreich von anderen Forschern eingesetzt. </p>

<p class="jove_content">Primäre neuronale Stammzellen Kulturen sind nützlich für die Untersuchung der Mechanismen des zentralen Nervensystems Entwicklung. Die Stammzellforschung wird unser Verständnis des Nervensystems und kann es uns ermöglichen, Therapien für bisher unheilbare Krankheiten und Verletzungen des Gehirns zu entwickeln. Darüber hinaus sollten Stammzellen für die Stammzellenforschung an die detaillierte Untersuchung von Mechanismen der neuronalen Differenzierung und Transdifferenzierung und die genetischen und umweltbedingten Signale unterstützt werden sollen, dass die direkte Spezialisierung der Zellen in bestimmten Zelltypen. Dieses Video zeigt eine Technik verwendet, um Zellen aus dem Embryonaltag 12,5 Maus dorsalen Vorderhirn aufzuschlüsseln. Die Dissektion Verfahren schließt die Gewinnung E12.5 Maus Embryonen aus der Gebärmutter, die Beseitigung der &quot;Haut&quot; mit feinen Pinzette und schließlich isoliert Stücke Großhirnrinde. Nach der Präparation wird das Gewebe verdaut und mechanisch dissoziiert. Die resuspendierten dissoziierten Zellen werden dann in &quot;Stammzelle&quot; Medien, die das Wachstum von neuralen Stammzellen bevorzugt kultiviert.</p>
