assembly, tuning and use of an apertureless near field infrared microscope for protein imaging

Este trabalho tem como objetivo instruir o leitor na montagem e operação de um microscópio de campo próximo infravermelho para imagens além do limite de difração. O microscópio de campo próximo apertureless é um instrumento de espalhamento de luz do tipo que fornece espectros de infravermelho em cerca de 20 nm resolução. Uma lista completa de componentes e um protocolo passo a passo para uso é fornecida. Erros comuns na montagem e afinação do instrumento são discutidas. Um conjunto de dados representativos que mostra a estrutura secundária de uma fibrilas amilóides é apresentado.
