Het vermogen van embryonale kiemcellen (EG) om te differentiëren in primordiale kiemcellen (PGC) en later in de gameten tijdens de vroege ontwikkelingsstadia is een perfect model om onze hypothese te pakken over kanker en onvruchtbaarheid. Dit protocol laat zien hoe primordiale kiemcellen van de ontwikkeling van gonaden isoleren in 10.5-11.5 dagen na coitum (DPC) muizenembryo&#39;s. Het ontwikkelen van gonadale richels van muis embryo&#39;s (C57BL6J) zijn gedissocieerd door een mechanische storing met collagenase, en afgewerkt in een muis embryo fibroblast feeder layer (MEF-CF1) die eerder mitotisch was met mitomycine C geïnactiveerd in de aanwezigheid van knock-out media en aangevuld met leukemie Inhibitor Factor (LIF), basic fibroblast groeifactor (bFGF), en Stem Cell Factor (SCF). Met behulp van deze geoptimaliseerde methoden voor PCG identificatie, isolatie, en de vestiging van de kweekomstandigheden vergunningen op lange termijn culturen van EG-cellen voor meer dan 40 dagen. De embryonale germinale cellijnen toonde embryonale fenotype en expressie van de gemeenschappelijke gebruikte markers van de pluripotente staat. Isolatie en afleiding van germinale cellen in de cultuur een middel om hun ontwikkeling te begrijpen in vitro en bieden de mogelijkheid om cumulatieve schade monitor genetische en epigenetische niveaus na blootstelling aan oxidatieve stress.
