Rétro-orbitaire d&#39;injection chez le poisson zèbre adulte donne le meilleur rendement de la livraison du matériel injecté dans le sang avec la plus faible incidence de la mortalité. retro-orbital injection in adult zebrafish

Le traitement médicamenteux des animaux entiers est un outil essentiel dans tout système modèle pour pharmacologique et chimique des études génétiques. Intraveineuse (IV) est souvent la forme la plus efficace et non invasive de la livraison d&#39;un agent d&#39;intérêt. Dans le poisson zèbre (<em&gt; Danio rerio</em&gt;), L&#39;injection IV de médicaments a longtemps été un défi en raison du diamètre des petits vaisseaux. Cela a également été un obstacle important pour l&#39;injection de cellules lors de la transplantation de cellules souches hématopoïétiques. Historiquement, les injections dans le sang ont été effectuées directement par le cœur. Cependant, cette procédure intra-cardiaque a un taux de mortalité très élevé comme le cœur est souvent percé lors de l&#39;injection en laissant le poisson vulnérables à l&#39;infection, la perte de sang massive ou des dommages aux organes fatale. S&#39;appuyant sur notre expérience avec la souris, nous avons développé une procédure nouvelle injection dans le poisson zèbre dans lequel le site d&#39;injection est derrière l&#39;œil et dans le sinus veineux rétro-orbitaire. Cette technique d&#39;injection rétro-orbitale (RO) a été employée avec succès dans les deux l&#39;injection de drogues chez les poissons adultes ainsi que la transplantation de l&#39;ensemble des cellules de moelle des reins. Injection de RO a un taux de mortalité beaucoup plus bas que les traditionnels d&#39;injection intra-cardiaque. Les poissons qui sont injectés rétro-orbital tendance à saigner d&#39;injection moins après et sont à un risque beaucoup plus faible de blessure à un organe majeur comme le cœur. Cellules plus, lorsqu&#39;elle est effectuée correctement, injecté et / ou de la drogue d&#39;entrer rapidement dans la circulation sanguine permettant composés pour exercer leur effet sur le poisson entier et des cellules de rein de facilement la maison de leur niche. Ainsi, cette technique d&#39;injection de nouveaux minimise la mortalité, tout en permettant la prestation efficace du matériel dans le sang des poissons adultes. Ici, nous illustrent cette technique en rétro-orbitaires injection de Tg (<em&gt; Globine</em&gt;: GFP) dans les cellules adultes<em&gt; Casper</em&gt; Poisson ainsi que l&#39;injection d&#39;un colorant fluorescent rouge (dextran, Texas Red<em&gt;</em&gt;) Dans les adultes<em&gt; Casper</em&gt; Poisson. Nous avons ensuite visualiser les injections succès par microscopie à fluorescence animaux ensemble.
