Vorbereitung <ol><li> Erhalten entsprechenden Drosophila-Linien mit einer hemocyte-spezifische Gal4-Treiber (zB SRP-Gal4 2) und einem genetisch kodierten fluoreszierenden Reporter unter UAS Kontrolle (z. B. UAS-GFP). Fliegen homozygot für SRP-Gal4, UAS-GMA 3 oder CRQ-Gal4, UAS-GFP 4, 5 sind besonders nützlich für die Bildgebung Zwecke (nb GMA ist GFP fusioniert mit dem Aktin-bindende Domäne von Moesin), siehe unten für eine Diskussion ?...

Die wichtigsten Elemente dieses Verfahrens sind die Auswahl gesunder Embryonen mit deutlich gekennzeichneten Blutzellen und sie vorsichtig montieren, ohne sie zu beschädigen. Sobald die Embryonen in die Halocarbonöl sind, sind sie resistent gegen Austrocknung und einmal montiert werden kann für mehrere Stunden abgebildet werden. In unseren Händen können wir Bild Blutzellen für drei Stunden, mit vernachlässigbarer Austrocknung des Embryos oder offensichtliche Foto-Schäden, wobei ein z-Stapel von Bildern alle drei...

Viele Studien befassen sich mit der Zellwanderung<em&gt; In-vitro-</em&gt; Methoden, während die physiologisch relevanten Umwelt ist, dass der Organismus selbst. Hier präsentieren wir ein Protokoll für die Montage von<em&gt; Drosophila melanogaster</em&gt; Embryonen und anschließender Live-Darstellung von fluoreszenzmarkierten Blutzellen, die embryonale Makrophagen dieses Organismus. Mit dem Gal4-UAS-System<sup&gt; 1</sup&gt; Fahren wir die Expression einer Vielzahl von genetisch kodierten, fluoreszenzmarkierten Marker in Blutzellen, ihre Entwicklungs-Verbreitung in den Embryo zu folgen. Nach dem Sammeln von Embryonen in der gewünschten Ausbaustufe ist die äußere Chorion entfernt und die Embryonen werden dann in Halocarbonöl zwischen einem hydrophoben, Gas-Membran und einem Deckglas für Live-Bildgebung montiert. Zusätzlich zu grob wandernde Parameter wie Geschwindigkeit und Ausrichtung, höhere Auflösung Bildgebung mit der Verwendung von fluoreszierenden Reporter von F-Aktin und Mikrotubuli Nähere Informationen über die Dynamik dieser Zytoskelett-Komponenten bieten gekoppelt.
