Många studier adress cell migration med<em&gt; In vitro</em&gt; Metoder, medan den fysiologiskt relevanta miljön är att av organismen själv. Här presenterar vi ett protokoll för montering av<em&gt; Drosophila melanogaster</em&gt; Embryon och efterföljande leva avbildning av fluorescerande hemocyter, den embryonala makrofager av denna organism. Använda Gal4-UAS-system<sup&gt; 1</sup&gt; Kör vi ett uttryck för en variation av genetiskt kodade, fluorescerande taggade markörer i hemocyter att följa deras utveckling spridning i hela embryot. Efter insamlingen av embryon på önskad utvecklingsstadium, är det yttre chorion bort och embryona sedan monteras i Halocarbon olja mellan en hydrofoba, gas-membran och ett glas täckglas för live avbildning. Förutom grov vandrande parametrar som hastighet och riktning, högre upplösning bildbehandling i kombination med användning av fluorescerande reportrar för F-aktin och mikrotubuli kan ge mer detaljerad information om dynamiken i dessa cytoskelettala komponenter.
