De zebravis is een ideaal model voor beeldvorming cel gedrag tijdens de ontwikkeling in vivo. Zebravis embryo&#39;s worden extern bevrucht en dus gemakkelijk te bereiken in alle stadia van ontwikkeling. Bovendien, hun optische helderheid maakt hoge resolutie imaging van cel-en moleculaire dynamica in de natuurlijke omgeving van de intacte embryo. We maken gebruik van een live-imaging benadering naar cel gedrag te analyseren tijdens de neurale lijst cel migratie en de uitgroei en begeleiding van neuronale axonen. Live-imaging is vooral nuttig voor het begrijpen van mechanismen die celmotiliteit processen te reguleren. Te visualiseren details van celbeweeglijkheid, zoals stuwend activiteit en moleculaire dynamica, is het voordelig om individuele cellen label. In de zebravis, plasmide DNA injectie levert een voorbijgaande mozaïek expressiepatroon en biedt duidelijke voordelen ten opzichte van andere cel etikettering methoden. Bijvoorbeeld, transgene lijnen vaak label hele cel populaties en kan dus visualisatie van de boete uitsteeksels (of veranderingen in de moleculaire distributie) in een enkele cel onduidelijk. Daarnaast, injectie van DNA in de een-cellig stadium is minder invasief en preciezer dan kleurstof injecties in latere stadia. Hier beschrijven we een methode voor het labelen van de individuele ontwikkeling van neuronen of neurale cellen en imaging hun gedrag in vivo. We injecteren plasmide DNA in een-cellig stadium embryo&#39;s, wat resulteert in mozaïek transgenexpressie. De vectoren bevatten cel-specifieke promotors die rijden expressie van een gen van interesse in een subset van de sensorische neuronen of neurale lijst cellen. Wij bieden voorbeelden van cellen gelabeld met membraan gerichte GFP of met een biosensor sonde die visualisatie van F-actine laat in levende cellen 1.
