synthesis and calibration of phosphorescent nanoprobes for oxygen imaging in biological systems

Zuurstof meting door fosforescentie afschrikken [1, 2] bestaat uit de volgende stappen: 1) de sonde wordt geleverd in het medium van belang (bijv. bloed of interstitiële vloeistof), 2) het object wordt verlicht met het licht van de juiste golflengte te prikkelen de sonde in zijn triplet toestand, 3) de uitgezonden fosforescentie wordt verzameld, en het tijdsverloop is geanalyseerd om de fosforescentie levensduur, die wordt omgezet in de zuurstofconcentratie (of de partiële druk, PO opbrengst<sub&gt; 2</sub&gt;). De sonde moet geen interactie met de biologische omgeving en in sommige gevallen te worden 4) uitgescheiden uit het medium op het einde van de meting. Elk van deze stappen stelt eisen aan de moleculaire ontwerp van de fosforescerende sondes, die de enige invasieve component van het meetprotocol vormen. Hier bespreken we het ontwerp van dendritische fosforescerende nanosensoren voor zuurstof metingen in biologische systemen. De sondes bestaan ​​uit Pt of Pd porfyrine-based polyarylglycine (AG) dendrimeren, perifeer gemodificeerd met polyethyleenglycol (PEG&#39;s) residuen. Voor een effectieve twee-foton excitatie, kan termini van de dendrimeren worden aangepast met twee-foton antenne chromoforen, die de excitatie-energie op te vangen en het kanaal naar de triplet kernen van de sondes via intramoleculaire FRET (Förster Resonance Energy Transfer). We beschrijven de belangrijkste fotofysische eigenschappen van de sondes en de huidige gedetailleerde kalibratie protocollen.
