Drosophila is al lang een favoriet modelsysteem voor het bestuderen van de relatie tussen de structuur van chromatine en genregulatie als gevolg van de cytologische voordelen die door de reus speekselklieren polytheen chromosomen van de derde instar larven. In dit weefsel worden de chromosomen ondergaan vele ronden van replicatie in de afwezigheid van celdeling die aanleiding geven tot ongeveer 1000 exemplaren. Het DNA blijft uitgelijnd na elke replicatieve cyclus resulteert in sterk vergroot chromosomen die een unieke kans om chromatine morfologie correleren met de lokalisatie van specifieke eiwitten te verstrekken. Bijgevolg is er een hoge mate van interesse in het definiëren van de epigenetische modificaties aanwezig op verschillende genen en in verschillende stadia van het transcriptie proces. Een belangrijk instrument voor een dergelijk onderzoek is de etikettering van polytheen chromosomen met antistoffen tegen het enzym, transcriptie factor, of histon-eiwitten van belang.
