Drosophila ist seit langem ein beliebtes Modellsystem für die Erforschung der Beziehung zwischen Chromatinstruktur und Genregulation durch die zytologische Vorteile durch die riesigen Speicheldrüse Polytänchromosomen des dritten Larvenstadium zur Verfügung gestellt worden. In diesem Gewebe die Chromosomen durchlaufen viele Runden der Replikation in Abwesenheit der Zellteilung, die zu etwa 1000 Exemplaren. Die DNA ausgerichtet bleibt nach jedem replikativen Zyklus resultierende stark vergrößert Chromosomen, die eine einzigartige Gelegenheit, Chromatin Morphologie mit der Lokalisierung von spezifischen Proteinen korreliert sind. Folglich hat es ein hohes Maß an Interesse an der Festlegung der epigenetischen Modifikationen derzeit an verschiedenen Genen und in den verschiedenen Phasen der Transkription Prozess. Ein wichtiges Instrument für solche Untersuchungen ist die Kennzeichnung von Polytänchromosomen mit Antikörpern gegen das Enzym, Transkriptionsfaktor, oder Histon-Modifikation von Interesse. Dieses Video-Protokoll zeigt die Squash-Technik in der Johansen Labor verwendet werden, um Drosophila Polytänchromosomen für Antikörpermarkierung vorzubereiten.
