In dit rapport presenteren we twee alternatieve methodes voor het opruimen van de muis uiervet pad ter voorbereiding van mamma-epitheel transplantaties. De standaard procedure is in gebruik sinds zijn ontwikkeling in 1959 door DeOme et al.. 1 Terwijl meer invasieve, de standaard methode is de beste methode voor de opleiding van beginnende omdat de structuur en de oriëntatie van de borstklier gemakkelijk bekeken worden. Dit artikel beschrijft en vergelijkt de dikke pad procedure voor de goedkeuring ontwikkeld door DeOme KB et al.. 1 en het spaarzaam procedure ontwikkeld door Brill B et al.. Twee, gevolgd door de borstklieren epitheliale transplantatie procedure. De borstklieren transplantatie procedure wordt veel gebruikt door mamma-biologen, omdat het profiteert van het feit dat de belangrijke ontwikkeling van de borstklier epitheel niet plaatsvinden tot na de puberteit. Op leeftijd van 3 weken, is de groei van de borstklier epitheliale boom beperkt tot de omgeving van de tepel en het vet pad is grotendeels verstoken van mamma-epitheel, maar door 7 weken oud zijn de epitheliale ductaal boom strekt zich de hele dikke pad. Daarom, als dit kleine gedeelte van het vet pad met epitheel, het gebied tussen de tepel en de lymfeklieren, verwijderd wordt op 3 weken oud zijn, zal de endogene epitheel nooit bevolken het uiervet pad en het vet pad wordt beschreven als &quot;gewist &quot;. Op dit moment kan mamma-epitheel uit een andere bron worden getransplanteerd in de gewist vet pad waar het de potentie heeft om mammae ductaal bomen uit te breiden door uit het vet pad. Deze procedure is gebruikt in veel experimentele modellen, inclusief het onderzoek tumor fenotype in transgene borstklieren epitheelweefsel zonder de verstorende effecten van het genotype op de gehele dier drie, in de identificatie van de mamma-stamcellen door het transplanteren van cellen in beperkte verwatering 4,5, het bepalen van Als hyperplastische knobbeltjes overgaan tot mammatumoren 6, en het effect van voorafgaande hormoon blootstelling op het gedrag van de borstklier epitheel 7,8 beoordelen. Drie week oude host muizen zijn verdoofd, schoongemaakt en vastgebonden op een chirurgische podium. Een mid-sagittale incisie wordt gemaakt door de huid, maar niet het buikvlies, dat zich uitstrekt van het schaambeen tot het borstbeen. Schuine sneden worden gemaakt door de huid van het midden van de sagittale incisie in het bekken in de richting van elk been. De huid is verwijderd van het buikvlies getrokken naar de 4e lies borstklier bloot te leggen. Het vet pad wordt gewist door het verwijderen van het vet pad weefsel juist voor de lymfeklier. Epitheel fragmenten of epitheelcellen worden getransplanteerd in de resterende gewist vet pad en de muis is gesloten.
