<p class="jove_content">Trotz der bekannten engen Verbindung, hat einen direkten Beweis, der gestörten zur Atherogenese gefehlt. Vor kurzem haben wir eine modifizierte Version der Carotis partielle Ligation verwendeten Methoden [1,2] zeigen, dass es akut induziert niedrig und oszillatorische Strömungsverhältnisse, zwei wichtige Eigenschaften des gestörten, in der Maus Arteria carotis communis. Mit diesem Modell haben wir direkten Beweis dafür, dass gestörten Tat führt zu einer schnellen und robusten Atherosklerose Entwicklung in Apolipoprotein E-Knockout-Maus [3] zur Verfügung gestellt. Wir haben auch eine Methode entwickelt, von Endothel-RNA-Präparation mit hoher Reinheit aus dem Maus-Intima [3]. Mit dieser Maus-Modell und Methode fanden wir, dass partielle Ligation Ursachen endotheliale Dysfunktion in einer Woche, gefolgt von robusten und schnellen Atherombildung in zwei Wochen in einem hyperlipämischen Mausmodell mit Features von komplexen Läsionen Bildung wie intraplaque Neovaskularisation von vier Wochen. Diese schnelle<em&gt; In vivo</em&gt; Modell und die Endothel-RNA Präparationsmethode verwendet werden, um molekulare Mechanismen zugrunde liegenden Fluss-abhängige Regulation der vaskulären Biologie und Krankheiten zu bestimmen. Auch könnte es verwendet, um verschiedene therapeutische Interventionen gezielt endotheliale Dysfunktion und Atherosklerose in erheblich reduziert Studiendauer zu testen.</p>
