Organotypischen Kultur von funktionellen neuronalen Gewebe sind wichtige Werkzeuge in der neurobiologischen Forschung. Idealerweise sollte ein solches System kompatibel sein mit bildgebenden Verfahren, genetische Manipulation und elektrophysiologische Ableitung. Hier präsentieren wir Ihnen eine einfache Interphase Gewebekultursystem für erwachsene Kaninchen Netzhaut, die keine spezielle Ausrüstung und nur sehr wenig Wartung benötigt. Wir zeigen die Präparation und Inkubation von Kaninchen-Netzhaut und Partikel-vermittelte Gentransfer von Plasmiden GFP oder eine Vielzahl von subzellulären Marker in retinalen Ganglienzellen. Kaninchen Netzhaut kultivierten diese Weise am Leben erhalten für bis zu 6 Tage mit sehr wenig Veränderungen der allgemeinen anatomischen Struktur oder die Morphologie der einzelnen Ganglien-und Amakrinzellen werden.
