<p class="jove_content">Menselijke pluripotente stamcellen (hPSCs) dat de menselijke embryonale stamcellen (hESC &#39;s) en de mens geïnduceerde pluripotente stamcellen (hiPSCs) omvatten zijn spannende cel bronnen door hun grenzeloze zelfvernieuwing mogelijkheden en hun potentieel om te differentiëren in verschillende celtypes. De pluripotente toestand van hPSCs wordt meestal bepaald door technieken zoals qPCR, immunocytochemie, en door andere<em&gt; In vitro</em&gt; En<em&gt; In vivo</em&gt; Het hanteren van strategieën in meerdere celtypen. Onder deze, zijn immunocytochemische technieken ontwikkeld voor routine karakterisering van de ongedifferentieerde toestand van hPSCs gebaseerd op een analyse van de kandidaat-intracellulaire en cel-oppervlak van biomarkers. Flowcytometrie analyses dienen om dit probleem aan te pakken, maar moet het aantal cellen en van de reagentia die normaal niet bevorderlijk voor routine kwaliteitscontrole beoordeling van hPSC culturen gebruiken. Zo is de ontwikkeling van praktische en reproduceerbare middel om monolaag cel monsters met bewaard integriteit voor marker evaluatie heeft vele voordelen in het stamcelonderzoek. Dit heeft veel baat immunocytochemische analyse, omdat de individuele cellen van de monolaag kunnen gemakkelijk worden waargenomen en gekwantificeerd voor de expressie van specifieke markers. Dit doel te bereiken, was een self-made Cytospin apparaat gebouwd en geoptimaliseerd voor gebruik met immunocytochemische kleuring. Twee celoppervlak markers (SSEA3/SSEA4) expressie werden geanalyseerd in een variant BG01 stamcellijn voor het doel van dit protocol.</p>
