in situ quantification of pancreatic beta-cell mass in mice

Tracing Veränderungen spezifischer Zellpopulationen in Gesundheit und Krankheit ist ein wichtiges Ziel der biomedizinischen Forschung. Der Prozess der Überwachung pankreatischen Beta-Zell-Proliferation und Inselchen Wachstum ist eine besondere Herausforderung. Wir haben eine Methode, um die Verteilung von Beta-Zellen im intakten Pankreas transgener Mäuse mit Fluoreszenz-markierten beta-Zellen mit einem Makro für ImageJ (rsb.info.nih.gov / ij /) geschrieben Bildaufzeichnung entwickelt. Nach Pankreas-Dissektion und Gewebe Lichtung, wird die gesamte Bauchspeicheldrüse als virtuelle Scheibe gefangen genommen, nach denen die GFP-markierten beta-Zellen untersucht werden. Die Analyse beinhaltet die Quantifizierung von insgesamt Beta-Zell-Bereich, Inselchen Anzahl und Größenverteilung in Bezug auf bestimmte Parameter und Standorte für jede Insel und für kleine Gruppen von beta-Zellen. Die gesamte Verteilung der Inseln kann in drei Dimensionen dargestellt werden, und die Informationen aus der Verteilung auf die Größe und Form der einzelnen Insel ermöglicht eine quantitative und qualitative Vergleich der Veränderungen in insgesamt Beta-Zell-Bereich auf einen Blick.
