murine bioluminescent hepatic tumour model

<p class="jove_content">Deze video beschrijft de oprichting van levermetastasen in een muismodel die vervolgens kunnen worden geanalyseerd door bioluminescente beeldvorming. Tumor-cellen zijn specifiek toegediend aan de lever om een ​​gelokaliseerde tumor lever veroorzaken, via mobilisatie van de milt en splitsen in twee, waardoor intact de vasculaire pedikel voor elk de helft van de milt. Lewis longcarcinoom cellen die constitutief uitdrukken van de vuurvlieg luciferase gen (luc1) zijn geënt in een hemi-milt, die vervolgens wordt 10 minuten later gereseceerd. De andere hemi-milt is intact gelaten en keerde terug naar de buik. Lever groei van de tumor kan worden gecontroleerd door bioluminescentie beeldvorming met behulp van de IVIS hele lichaam imaging-systeem. Kwantitatieve beeldvorming van de groei van tumoren met behulp van IVIS zorgt voor een nauwkeurige kwantificering van levensvatbare tumorcellen. Tumor celdood en necrose als gevolg van medicamenteuze behandeling is vroeg aangegeven door een vermindering van de lichtgevende signaal. Deze muis model maakt voor het onderzoeken van de mechanismen die ten grondslag liggen aan uitgezaaide tumor-cel overleving en de groei en kan worden gebruikt voor de evaluatie van therapeutica van levermetastasen.</p>
