Ratten met een gewicht van ongeveer 250-300g worden gekocht van Charles River (Sandhofer Weg 7, D-97633 Sulzfeld). orthotopic aortic transplantation: a rat model to study the development of chronic vasculopathy

Onderzoek modellen van chronische afstoting zijn essentieel voor pathobiological en pathofysiologische processen te onderzoeken tijdens de ontwikkeling van de transplantatie vasculopathie (TVP). De meest gebruikte diermodel voor hart-en chronische afstoting studies is het heterotope harttransplantatie model uitgevoerd in het laboratorium knaagdieren. Dit model wordt veel gebruikt in experimenten, omdat Ono en Lindsey (3) publiceerden hun techniek. Voor het analyseren van de bevindingen in de bloedvaten, het hart moet worden coupes en alle schepen moeten worden gemeten. Een andere methode om chronische afstoting te onderzoeken in cardiovasculaire ondervragingen is de aorta transplantatie model (1, 2). In de orthotope aorta transplantatie model, kan de aorta gemakkelijk histologisch geëvalueerd worden (2). De PVG-to-ACI model is vooral nuttig voor CAV studies, omdat acute vasculaire afstoting is geen belangrijke storende factor en ciclosporine A (CsA) behandeling belet niet dat de ontwikkeling van CAV, vergelijkbaar met wat we vinden in de klinische setting (4 ). A7-dagen periode van CsA is vereist in dit model van acute afstoting te voorkomen en om overleving op lange termijn te bereiken met de ontwikkeling van TVP. Dit model kan ook gebruikt worden om acute cellulaire afstoting en media necrose in xenogene modellen (5) te onderzoeken.
