5-bromo-2&#39;-deoxiuridin (BrdU) märkning av mitokondriellt DNA i HeLa-celler

För att börja mitokondrier märkning förbereda en 20 mikromolar lösning av BrdU i DMEM med 10% fetalt bovint serum eller FBS. Tillsätt sedan mitokondriernas spårningsfärg för att justera den slutliga koncentrationen till 1,1 mikromolar. Efter 15 timmar tillsätts 10 mikroliter av mitokondriernas spårningsfärglösning till HeLa-cellsuspensionen innehållande brunnar.Inkubera cellerna i en timme. Förhållandena som används för att märka nyligen syntetiserade DNA-molekyler med BrdU möjliggjorde detektion av BrdU-märkt DNA i mitokondrierna i HeLa-celler. Signalen erhållen med anti-BrdU-antikroppar var specifik och observerades endast i celler behandlade med BrdU.Oavsett BrdU-behandling detekterades ingen anti-BrdU- eller anti-DNA-antikroppssignal i mitokondrierna i radnollceller. Kvantifiering av den fluorescerande signalen från anti-BrdU-antikropparna indikerade att radnollceller behandlade med BrdU visade samma låga fluorescensnivå som BrdU-obehandlade celler, medan signalen för föräldralinjerna A549 och HeLa var 50 gånger högre.
