Cell-Based Drug Screening for Inhibitors of Autophagy Related 4B Cysteine Peptidase

Autofagie Protease Tag-Compound Additie en Cellulaire Supernatant Oogst voor Drug Screening

Begin met het ontdooien van de Selleckchem Small Molecule Compound Library-platen bij kamertemperatuur. Open de software en klik op Nieuw protocol. Klik op het tabblad Protocol op Sample Plate Format, gevolgd door 384PP.Selecteer vervolgens op Sample Plate Type 384PP_DMSO2. Kies vervolgens Cell Carrier 384 Ultra PN als het type bestemmingsplaat. Klik op Keuzelijst en selecteer de importopties om de spreadsheet met het doseerprogramma te importeren.Klik op de optie Protocol uitvoeren om de juistheid van de weergegeven informatie te controleren en klik vervolgens op Uitvoeren. Druk vervolgens op Start in het nieuwe venster met de titel Status uitvoeren en volg de stappen die worden weergegeven in de promptvensters. Gebruik een akoestische vloeistofdispenser op nanoschaal en doseer 50 nanoliter per putje van de Small Molecule Compound Library in een plaat met 384 putjes.Om te beginnen met het oogsten van cellulair supernatant, begint u met het configureren van het dek van het laboratoriumautomatiseringswerkstation. Plaats een wegwerprek met 96 tips op de P1-positie en plaats de testplaten op posities P2 en P4. Plaats ten slotte de lege, effen zwarte platen met 384 putjes op posities P3 en P5. Zuig met behulp van de tips van P1 10 microliter van het supernatans uit de plaat op P2 en breng het vervolgens over in de lege vaste zwarte plaat op P3. Gooi de tips op positie P6 weg. Herhaal de overdracht van supernatans van P4 naar P5.
