Evaluering af migration og invasion af salidrosidbehandlede MCF-7-celler i et sårridseassay

Til at begynde med inkuberes to milliliter af de dyrkede MCF-7-celler i en seks-brøndplade indtil 90% sammenløb. I det dyrkede cellemonolag oprettes en lineær ridse afviklet i midten ved hjælp af en steril pipettespids. Tag derefter billederne med et optisk mikroskop med forskellige tidsintervaller.Til transwell-analysen suspenderes MCF-7-celler i et serumfrit medium. Se derefter cellerne i det øvre transwellkammer, enten forudkodet eller ikke-kodet med Matrigel. Der tilsættes DMEM-komplet substrat i bunden af transwellkammeret for at fungere som en kemisk inducer.Efter 24 timer skal du fjerne cellerne i det øverste kammer og fikse de resterende invasive og vandrende celler ved hjælp af methanol. Plet MCF-7-cellerne ved hjælp af krystalviolet opløsning. Billede derefter de farvede celler med et optisk mikroskop.Der blev observeret en dosisafhængig hæmmende virkning af salidrosid på celleproliferation med et fald på 50 % i cellevitalitet ved 40 mikromolar. Endvidere kunne salidrosid hæmme vitaliteten af MCF-7-celler over tid med et fald på 50% i MCF-7-cellevitalitet efter 24 timers co-inkubation. Sår-ridse-analysen viste en hæmmende virkning for salidrosidbehandling på MCF-7-celler.Endvidere reducerede salidrosidbehandling signifikant den uønskede migration og invasion af MCF-7-celler.
