To begin, culture the NF54 strain of Plasmodium falciparum in 2%hematocrit of O+human RBCs in supplemented complete RPMI medium. For sorbitol synchronization, pellet the asynchronous parasites in a 50 milliliter conical tube at 2, 300 G for three minutes at room temperature. After discarding the supernatant, incubate the parasitized red blood cells with nine volumes of 5%sorbitol for 10 minutes at 37 degrees Celsius.
Following incubation, pellet the red blood cells as demonstrated earlier, and discard the sorbitol. Wash the sorbitol treated parasites with incomplete RPMI and RPMI media without AlbuMAX and sodium bicarbonate. Subsequently incubate them in complete RPMI to obtain the ring stage parasites.
